Nine nurses from the post-anaesthesia care unit, with no prior experience of LMA or PLMA insertion, were observed inserting the LMA and PLMA in 60 ASA 1 to 2 anaesthetized, paralysed adults following manikin-only training. Our data indicate that laryngeal tubes such as the LTS-II/D are a valuable tool in "in-hospital" emergency airway management. Placement is safe and can be easily performed by anesthetists and nurse specialists, even outside the OR, within a short time.. BACKGROUND: To prevent endothelium and nerve lesions, tracheal tube and laryngeal mask cuff pressure is to be maintained at a low level and yet be high enough to secure air sealing. METHOD: In a prospective quality-control study, 201 patients undergoing surgery during anaesthesia (without the use of nitrous oxide) were included for determination of the cuff pressure of the tracheal tubes and laryngeal masks. RESULTS: In the 119 patients provided with an endotracheal tube, the median cuff pressure was 30 (range 8 - 100) cm H2O and the pressure exceeded 30 cm H2O (upper recommended level) for 54 patients. During the development of a perioperative information system, we found it useful to devise a grading system similar to that used for grading the view during laryngoscopy. Initially, we chose grades 0-4, defined in table 1. There was also a means by which practitioners could type in a text description of mask ventilation. In genetic manipulation, transformation refers specifically to the successful insertion of foreign DNA (or more accurately exogenous DNA) into the bacterium. In the early 1970s, E coli was the best characterised bacterium, and the only bacterium known to possess plasmids. Plasmids are little circular rings of DNA, independent of the bacteriums single chromosome. They can even replicate themselves on their own, which, for biologists, is a very useful thing indeed. In genetics, pBluescript (pBS) or pBluescript II is a commercially available phagemid containing several useful sequences for use in cloning with bacteriophage. The sequences include a polylinker sequence (MCS), antibiotic resistance sequence to ampicillin and an E. coli and f1 helper phage origin of replication. The polylinker sequence is located within a LacZ controlled gene designed to provide a blue coloration when expressed in bacteria. This is usually achieved via IPTG found in agarose growth media used to culture bacteria with pBS. If the gene is disrupted by successful insertion of a DNA sequence, the bacteria exhibit a white coloration in Blue white screening, distinguishing successful recombination from those phagemids which were not altered.
