Lactoperoxidase is a peroxidase enzyme secreted from mammary, salivary, and other mucosal glands that functions as a natural antibacterial agent. Lactoperoxidase is a member of the heme peroxidase family of enzymes. In humans, lactoperoxidase is encoded by the LPO gene. Lactoperoxidase catalyzes the oxidation of a number of inorganic and organic substrates by hydrogen peroxide. These substrates include bromide and iodide and therefore lactoperoxidase can be categorised as a haloperoxidase. Another important substrate is thiocyanate. The oxidized products produced through the action of this enzyme have potent bactericidal activities. Lactoperoxidase together with its inorganic ion substrates, hydrogen peroxide, and oxidized products is known as the lactoperoxidase system. The lactoperoxidase system plays an important role in the innate immune system by killing bacteria in milk and mucosal (linings of mostly endodermal origin, covered in epithelium, which are involved in absorption and secretion) ...
Peroxidases catalyse reactions in which hydrogen peroxide is reduced and a suitable electron donor is subsequently oxidised. Most assays are based on the principle that the electron donor in oxidised form absorbs light and can be determined by a spectrophotometric assay. Currently 2, 2-azino-di-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) is accepted as the electron donor of choice for peroxidase assay.

The enzyme lactoperoxidase was used to catalyse the radioiodination of membrane proteins in intact human erythrocytes and in erythrocyte ghosts. Two major proteins of the erythrocyte membrane were isolated after iodination of these two preparations, and the peptide maps of each protein so labelled were compared. Peptides from both proteins are labelled in the intact cell. In addition, further mobile peptides derived from one of the proteins are labelled only in the ghost preparation. Various sealed ghost preparations were also iodinated, lactoperoxidase being present only at either the cytoplasmic or extra-cellular surface of the membrane. The peptide maps of protein E (the major membrane protein) labelled in each case were compared. Two discrete sets of labelled peptides were consistently found. One group is obtained when lactoperoxidase is present at the extra-cellular surface and the other group is found when the enzyme is accessible only to the cytoplasmic surface of the membrane. ...

When deprived of oxygen, Bacille Calmette-Guérin (BCG)-activated macrophages no longer lysed P388 lymphoma cells. Both H2O2 release and cytotoxicity by BCG-activated macrophages and by granulocytes triggered with phorbol myristate acetate (PMA) were markedly inhibited when the glucose concentration in the medium was reduced to 0.03 mM or less, or if glucose were replaced with galactose. Catalase abolished PMA-triggered cytotoxicity by both types of effector cells, whereas superoxide dismutase had no effect. Ferricytochrome C reduced the cytotoxicity of BCG-activated macrophages, an effect which was largely reversed by superoxide dismutase. 10 drugs, thought to quench singlet oxygen and/or scavenge hydroxyl radical, did not affect cytotoxicity in this system. Neither azide nor cyanide reduced cytolysis, but there was marked inhibition by lactoperoxidase and iodide. This suggested that cytotoxicity was not dependent upon myeloperoxidase, and that lactoperoxidase may have diverted H2O2 from the ...

AU - Salathe, Matthias. PY - 2001/10/16. Y1 - 2001/10/16. N2 - Enzymes secreted onto epithelial surfaces play a vital role in innate mucosal defense, but are believed to be steadily removed from the surface by mechanical actions. Thus, the amount and availability of enzymes on the surface are thought to be maintained by secretion. In contrast to this paradigm, we show here that enzymes are retained at the apical surface of the airway epithelium by binding to surface-associated hyaluronan, providing an apical enzyme pool ready for use and protected from ciliary clearance. We have studied lactoperoxidase, which prevents bacterial colonization of the airway, and kallikrein, which mediates allergic bronchoconstriction that limits the inhalation of noxious substances. Binding to hyaluronan inhibits kallikrein, ...
Zymox Otic Enzymatic Solution with Hydrocortisone uses a patented LP3 Enzyme System to create a biological system that is effective against a variety of bacterial, fungal and yeast infections in the ear. The three enzymes (Lactoperoxidase, Lysozyme and Lactoferrin) react with infectious pus and debris, forming their antimicrobial properties to counteract infections. Using a cleaner prior to use will disrupt this biological system, so the solution acts as an ear cleaner and medication in one daily dose. With no memory, its effective against even antibiotic-resistant microorganisms for fast otic treatment. The antibiotic-free formula is safe for use in animals of any age.
Hypothiocyanite is the anion [OSCN]− and the conjugate base of hypothiocyanous acid (HOSCN). It is an organic compound part of the thiocyanates as it contains the functional group SCN. It is formed when an oxygen is singly bonded to the thiocyanate group. Hypothiocyanous acid is a fairly weak acid; its dissociation constant is 5.3. Hypothiocyanite is formed by peroxidase catalysis of hydrogen peroxide and thiocyanate: H2O2 + SCN− → OSCN− + H2O Hypothiocyanite occurs naturally in the antimicrobial immune system of the human respiratory tract in a redox reaction catalyzed by the enzyme lactoperoxidase. It has been researched extensively for its capabilities as an alternative antibiotic as it is harmless to human body cells while being cytotoxic to bacteria. The exact processes for making hypothiocyanite have been patented as such an effective antimicrobial has many commercial applications. Lactoperoxidase-catalysed reactions yield short lived intermediary oxidation products of SCN−, ...

In this chapter, the discussion of spoilage of milk and dairy products is based on the types of microorganisms associated with various defects. These include gram-negative psychrotrophic microorganisms, gram-positive bacteria including lactic acid bacteria and spore-forming bacteria, yeasts, and molds. The chapter describes the interactions of these microorganisms with dairy foods that lead to commonly encountered product defects. Citrate in milk can be utilized by many microorganisms but is not present in sufficient amount to support significant growth. The major microbial inhibitors in raw milk are lactoferrin and the lactoperoxidase system. As much as milk and dairy products are important components of a healthy diet, the presence of pathogenic microorganisms poses a potential health hazard to the consumers. Proteases of psychrotrophic bacteria cause product defects either at the time they are produced in the product or as a result of the enzymes surviving a heat process. Spoilage of milk and dairy
TY - JOUR. T1 - Radioiodination and binding characteristics of a highly active opioid peptide. AU - Kream, R. M.. AU - Zukin, R. S.. PY - 1979/1/1. Y1 - 1979/1/1. N2 - D-ala2, N-Me-phe4, met (O)5-ol-enkephalin (Sandoz, FK 33-824) was radiolabeled to a specific activity of 1000 Ci/mmol using soluble lactoperoxidase and carrier-free Na125l. The [125l]-labeled enkephalin analog was purified by a combination of gel filtration and ion-exchange chromatography; it cochromatographed with its [127l] labeled analog on thin layers. [125l] D-ala2, N-Me-phe4, met (O)5-ol-enkephalin binds stereospecifically to whole brain membrane preparations from rat with saturation at 5 nM at 25°C. Scatchard analysis reveals two classes of binding sites with apparent Kds of 0.42 x 10-9 M and 3.70 x 10-9 M. The ratio of high affinity to low affinity binding sites is approximately 1:3. At 25°C, 40 mM sodium ion elicits half-maximal inhibition of stereospecifically bound counts, while 100 mM potassium inhibits binding by ...

The interest for the development of new active packaging materials has rapidly increased in the last few years. Antimicrobial active packaging is a potential alternative to protect perishable products during their preparation, storage and distribution to increase their shelf-life by reducing bacterial and fungal growth. This review underlines the most recent trends in the use of new edible coatings enriched with antimicrobial agents to reduce the growth of different microorganisms, such as Gram-negative and Gram-positive bacteria, molds and yeasts. The application of edible biopolymers directly extracted from biomass (proteins, lipids and polysaccharides) or their combinations, by themselves or enriched with natural extracts, essential oils, bacteriocins, metals or enzyme systems, such as lactoperoxidase, have shown interesting properties to reduce the contamination and decomposition of perishable food products, mainly fish, meat, fruits and vegetables. These formulations can be also applied to ...

Oxidative iodination of human lactoferrin (Lf) as commonly performed by using the chloramine-T, the Iodogen or the lactoperoxidase method produces an unreliable tracer protein because of excessive and heterogeneous polymer formation. Before iodination a minor tetramer fraction may be demonstrable in iron-saturated Lf only. Iodination-induced polymerization of iron-poor as well as iron-saturated Lf occurs independently of the presence or absence of 10 mM-EDTA and the 125I-/Lf molar ratio used for iodination. 125I-Lf polymers are mainly covalently linked, as suggested by the lack of substantial dissociation in SDS/polyacrylamide-gel electrophoresis. Damage to the 125I-Lf monomer may be another consequence of oxidative iodination. This is demonstrated in SDS/polyacrylamide-gel electrophoresis where 50% of the radioactivity of apparently normal monomer (Mr 75,000) is displaced to a lower-Mr region (30,000-67,000) after reduction with dithiothreitol. Non-oxidative iodination by the Bolton-Hunter ...
Among the human heme-peroxidase family, myeloperoxidase (MPO) has a unique disulfide-linked oligomeric structure resulting from multi-step processing of the pro-protein monomer (proMPO) after it exits the endoplasmic reticulum (ER). Related family members undergo some, but not all, of the processing steps involved with formation of mature MPO. Lactoperoxidase has its pro-domain proteolytically removed and is a monomer in its mature form. Eosinophil peroxidase undergoes proteolytic removal of its pro-domain followed by proteolytic separation into heavy and light chains and is a heterodimer. However, only MPO undergoes both these proteolytic modifications and then is further oligomerized into a heterotetramer by a single inter-molecular disulfide bond. The details of how and where the post-ER processing steps of MPO occur are incompletely understood. We report here that T47D breast cancer cells stably transfected with an MPO expression plasmid are able to efficiently replicate all of the processing steps
The adhesive properties of Chinese hamster V79 cells were analyzed and characterized by various cell dissociation treatments. The comparisons of aggregability among cells dissociated with EDTA, trypsin + Ca2+, and trypsin + EDTA, revealed that these cells have two adhesion mechanisms, a Ca2+-independent and a Ca2+-dependent one. The former did not depend on temperature, whereas the latter occurred only at physiological temperatures. Both mechanisms were trypsin sensitive, but the Ca2+-dependent one was protected by Ca2+ against trypsinization. In morphological studies, the Ca2+-independent adhesion appeared to be a simple agglutination or flocculation of cells, whereas the Ca2+-dependent adhesion seemed to be more physiological, being accompanied by cell deformation resulting in the increase of contact area between adjacent cells. Lactoperoxidase-catalyzed iodination of cell surface proteins revealed that several proteins are more intensely labeled in cells with Ca2+-independent adhesiveness ...

Saliva provides critical defense against dental decay. When food is chewed salivary flow increases which provides a large liquid volume for plaque acids to diffuse into and an increased concentration of bicarbonate buffer which neutralizes acids. Saliva provides a constant remineralization mechanism because it is supersaturated with calcium and phosphate ions. The environment of the tooth surface therefore favors mineral deposition. Additionally, salivary proteins and glycoproteins such as lysozyme, lactoperoxidase, lactoferrin, and high-molecular weight agglutinins, exhibit antibacterial activity although S. mutans is minimally affected by lysozyme. [1] Exposure to fermentable carbohydrates such as sucrose causes a rapid pH drop in dental plaque. A study took pH readings of subjects with varying degrees of decay and then had subjects rinse with a 10% glucose solution. The pH was read at periodic intervals until it returned to its original level. The rapid drop in pH shows that glucose was ...

The products obtained using freeze-drying also had a bactericidal effect on Escherichia coli, but the activity of these samples was 30%. The other whey products had no apparent effect on the bacterial cells.. The antimicrobial activity of whey products can be attributed to the iron-binding property of α-La, β-Lg, LF, lactoperoxidase (LP), BSA and lysozyme. These proteins decrease the iron available to the microorganisms and also act by direct binding to microbial membranes. However, their effects are not limited to bacteria with iron requirements (e.g., coliforms), particularly in the case of LF. LF can damage the outer membranes of Gram-negative bacteria via binding to Lipid A lipopolysaccharides (LPS) [55] and enhance bacterial susceptibility to hydrophobic antimicrobials such as lysozyme. However, other studies have showed that addition of cations, such as Ca2+ inhibits LF binding to LPS, as does the addition of polymyxin B [56]. Further studies have shown that LF is bactericidal only when ...

Respiratory syncytial virus (RSV) is the most frequent cause of bronchiolitis in infants and children worldwide. Many animal models are used to study RSV, but most studies investigate disease in adult animals which does not address the unique physiology and immunology that makes infants more susceptible. The perinatal (preterm and term) lamb is a useful model of infant RSV disease as lambs have similar pulmonary structure including airway branching, Clara and type II cells, submucosal glands and Duox/lactoperoxidase (LPO) oxidative system, and prenatal alveologenesis. Lambs can be born preterm (90% gestation) and survive for experimentation although both preterm and term lambs are susceptible to ovine, bovine and human strains of RSV and develop clinical symptoms including fever, tachypnea, and malaise as well as mild to moderate gross and histologic lesions including bronchiolitis with epithelial injury, neutrophil infiltration and syncytial cell formation. RSV disease in preterm lambs is more severe

Mouse lymphocyte H-2 and Ia glycoproteins have been analyzed with a two-dimensional (2-D) acrylamide gel electrophoresis technique, in which proteins are separated first according to their charge in isoelectrofocusing gels and then according to their size in sodium dodecyl sulfate gels. Individual polypeptide chains from radiolabeled cells are resolved as discrete spots on autoradiograms of the gels, forming patterns which are characteristic of the proteins in the sample. 2-D gels of H-2K, H-2D, and Ia glycoproteins immunoprecipitated from 35S-methionine-labeled cells reveal that these proteins exist in the cells as complex arrays of molecules heterogeneous in both size and charge. Lactoperoxidase-catalyzed radioiodination of lymphocyte surfaces labels only subsets of the total H-2 and Ia molecules with 125I, indicating that some of the molecules may represent cytoplasmic precursors of the cell surface proteins. This theory is supported by the kinetics of labeling of various spots in ...
