Tear proteins are supplied by the regulated fusion of secretory vesicles with the apical surface of lacrimal gland acinar cells, utilizing trafficking mechanisms largely yet uncharacterized. We investigated the role of Rab27b in regulating the exocytotic pathway of these secretory vesicles. Evaluation of morphological changes by transmission electron microscopy of lacrimal glands from Rab27b-/- and Rab27ash/ash/Rab27b-/- mice, but not Ashen mice deficient in Rab27a, showed significant changes in organelle morphology which included an approximate 50% decrease in abundance of secretory vesicles and decreased vesicle localization in the subapical region. Along with an apparent secretory pathway effect, knockout of Rab27b also resulted in a two-fold increase in the number of lysosomes, four-fold increase number of damaged mitochondria, two-fold increase in the formation of autophagosome-like organelles, and observed increased ER swelling and vesiculation. Confocal fluorescence microscopy analysis, ...
Glycosaminoglycans (GAGs) play a central role in embryonic development by regulating the movement and signaling of morphogens. We have previously demonstrated that GAGs are the co-receptors for Fgf10 signaling in the lacrimal gland epithelium, but their function in the Fgf10-producing periocular mesenchyme is still poorly understood. In this study, we have generated a mesenchymal ablation of UDP-glucose dehydrogenase (Ugdh), an essential biosynthetic enzyme for GAGs. Although Fgf10 RNA is expressed normally in the periocular mesenchyme, Ugdh mutation leads to excessive dispersion of Fgf10 protein, which fails to elicit an FGF signaling response or budding morphogenesis in the presumptive lacrimal gland epithelium. This is supported by genetic rescue experiments in which the Ugdh lacrimal gland defect is ameliorated by constitutive Ras activation in the epithelium but not in the mesenchyme. We further show that lacrimal gland development requires the mesenchymal expression of the heparan sulfate ...
Lacrimal gland acinar cells (LGACs) are the major contributor to ocular surface proteins extruded into the tear film. It has been suggested that a reduction in LGAC output is attributed to onset of dry eye syndromes ranging from mild dye eye to the more severe Sjögrens Syndrome. We investigated the role of the actin-based myosin motor, Myosin 5c (Myo5c) in vesicle transport in exocrine secretion. Confocal fluorescence and immunogold electron microscopy revealed that Myo5c was associated with secretory vesicles (SVs) in primary rabbit LGAC. Upon stimulation with carbachol (CCh), Myo5c was also detected in association with actin-coated fusion intermediates. Adenovirus-mediated expression of GFP fused to the tail domain of Myo5c (Ad GFP-Myo5c-tail) showed that this protein was localized to SVs. Furthermore, its expression induced a significant (p less than or equal to 0.05) decrease in CCh-stimulated release of two SV content markers, secretory component (SC) and syncollin-GFP (Syn-GFP) of 58.14% ...
Histological examination of 99 human lacrimal glands showed a relationship between atrophy of the secretory acini and secretory duct obstruction, ascending periductal fibrosis, and obliteration of the adjacent blood vessels caused by lymphocytic and polymorphonuclear inflammation. Investigation of the subgroups of the B lymphocytic series by immunohistochemistry did not show any statistical change with age, sex, fibrosis, or lymphocytic inflammation. The concept of senile atrophy occurring as a result of senescent involution of the lacrimal gland is challenged on the basis of the histological findings. ...

Purpose: Sox11, a transcription factor expressed in trigeminal and dorsal root ganglion neurons early in development, is induced in adult sensory neurons following nerve injury. As a promotor of nerve regeneration, we hypothesized that Sox11 may also be expressed during dry eye in order to increase trophic support to the cornea provided by corneal afferent neurons. The present study examined Sox11 expression and examined corneal epithelial integrity and mechanical sensitivity in Sox11 conditional knockout animals following lacrimal gland excision.. Methods: Male C57BL/6 mice were obtained from Jackson Labs and Sox11 conditional knockout (Sox11CKO) mice were generated using Cre-LoxP-mediated recombination under the control of a NaV1.8 promoter. Under isoflurane, unilateral lacrimal gland excision was performed. Either the left exorbital lacrimal gland, or both the exorbital and intraorbital lacrimal glands were excised. For sham surgeries, incisions were made and glands were partially exposed on ...
The globe is formed from neuroectoderm of the forebrain, surface ectoderm from the head, mesoderm lying between these layers, and neuronal crest cells.19 The surface ectoderm forms the corneal and conjunctival epithelium, lens, and lacrimal glands.19,20 In human lacrimal gland morphogenesis, the formation of epithelial bud at the level of the superior conjunctival fornix constitutes the onset of glandular development in embryos between 22 and 25 mm.21 In the 5-mm embryo, the choroidal fissure exists in the inferior edge of the optic cup19-20; later, the choroidal fissure closes at the 15-mm stage. Thereafter, the iris and ciliary body complete their development within the globe after the closure of the choroidal fissures.22 Therefore, it is not well understood how lacrimal tissue reaches in the intraocular structures, because the choroidal fissure closes at the 15-mm stage, and the lacrimal gland does not appear until the 25-mm stage.. Some theories have been proposed concerning the origin of ...
The lacrimal gland (LG) is an exocrine gland important for secretion of the tear film. The kinase p38 has important signal transduction functions, e.g. in gene transcription, but has previously not been known to modulate exocrine secretion. The aim of the current study was to investigate the role of p38 in carbachol (Cch)-induced LG secretion in LG acinar cells in vitro. Western blotting was used to determine the phosphorylation status of p38 and p42/44 and determine expression of p38 isoforms. To determine the effect of p38 inhibition on LG secretion, PD 169316, a general p38 inhibitor, and SB 239063, an inhibitor of p38α and β, were added to the cells prior to secretion measurements. The results revealed activation of p38 mediated by Cch stimulation and inhibition of Cch-induced secretion as a result of p38 inhibition. The inhibition was observed with PD 169316 isoforms, but not with SB 239063. The p38δ isoform was shown to have robust expression both by Western blotting of acinar cells and ...
Numerous eye complications are associated with that relating to diabetes mellitus, of which the dry eyes Disease is one of the most typical as a result of improvement in the tear proteins from that relating to the healthy people .Diabetes could damage certain nerves in your body. In the eyes, such damage can block the device that controls tear secretion. When this occurs, the lacrimal glands fail to produce sufficient tears, resulting in dry eyes. Insulin deficiency is another symptom associated with diabetes. Aside from controlling blood sugar, insulin comes with a major effect, on several glands in your body. In the eyes, lacrimal gland metabolism is affected by insulin. If you find low insulin in your body, the biomechanical balance of the eyes is disrupted resulting in ocular dryness. Another reaction of diabetes is lacrimal gland inflammation thats on account of abnormal lacrimal secretion. When this gland is inflamed, tear secretion is affected, which leads to dry eyes. Remedial ...
Purpose: Our group has been studying the feasibility of viral vector gene transfer to the lacrimal gland. The aims of this work are: a) to evaluated changes in the tear secretion and cells of ocular surface, b) to evaluate the changes in the lacrimal gland of rats with an adenovirus encoding the human erythropoietin (Epo) gene (AdLTR2EF1a-hEPO) or GFP (AdGFP).. Methods: Male Wistar rats 6-8 weeks were divided into 3 groups (n = 5/group), where the control group received an injection of saline and the treated group received an injection of erythropoietin adenovirus (Ad-Epo) (20ul, 1012/ml ) and a group that received an injection of GFP adenovirus (Ad-GFP) (20 ul, 1012/ml) in the lacrimal gland. Data was collected 6 days after injection. The main parameters evaluated were tear secretion (phenol red thread), lacrimal gland (LG) and ocular globe histology, ocular surface impression citology (IC), expression of erythropoietin in the western blotting.. Results: Tear secretion was not affected by viral ...

Glycosaminoglycans (GAGs) are the major constituents of the extracellular matrix that control the transport and signaling of numerous growth factors (Hacker et al., 2005). Consisting of 50-400 repeats of disaccharide units, GAGs can be divided by their composition, sulfation and epimerization into chondroitin sulfate (CS), dermatan sulfate (DS), heparan sulfate (HS)/heparin, and keratan sulfate (KS). A common precursor of all GAGs is UDP-glucuronic acid, which is synthesized from UDP-glucose by a single mammalian enzyme, UDP-glucose dehydrogenase (Ugdh). Together with an amino sugar such as N-acetylglucosamine for HS and N-acetylgalactosamine for CS, these monosaccharides are incorporated by polymerization enzymes into the backbone of the polysaccharide and are further modified by a series of sulfotransferase enzymes (Esko and Selleck, 2002). For example, the polymerization of HS is exclusively catalyzed by Ext enzymes, which are followed by N-deacetylase/N-sulfotransferase (Ndst) enzymes to ...

Tumors of the lacrimal gland constitute less than 5% of all biopsied orbital lesions [3, 4]. The most common epithelial malignancy is primary adenoid cystic carcinoma; adenocarcinoma is rare, representing only 5 to 7% of epithelial tumors of the lacrimal gland [2-6]. Because there is no specific histopathologic classification for lacrimal gland tumors, they are often classified according to the histological classification of salivary gland tumors with which they share many similarities [1, 3, 7].. This subtype of lacrimal gland, adenocarcinoma, was described for the first time in 1996 by Katz et al. [2]. Since then, most reports in the literature are case reports describing an aggressive clinical pattern and poor prognosis [1, 2, 8].. The tumor usually begins in the upper eyelid as a growth and is accompanied by clinical symptoms such as lid pseudoptosis, exophthalmos, dystopia, pain, and reduced visual acuity [4, 6].. The patients short life expectancy after surgical treatment for ...
A ductal cyst of the major lacrimal gland, or dacryops, is a rare clinical entity, encountered mainly in its palpebral lobe (1, 3, 4). Ductal cysts of the accessory lacrimal glands of Wolfring and Krause are even more uncommon than those of the major lacrimal gland and are rarely reported in the ophthalmologic literature (1, 2, 5-7). Clinically, cysts of the accessory gland develop insidiously and present as painless cystic masses of the lids. They are of variable size and are located in the previously described anatomic locations of the accessory glands. Most frequently, these cysts occur in patients with previous traumatic or inflammatory conditions of the conjunctiva, particularly in patients with trachoma (2, 5). Less commonly, a congenital anomaly of an excretory duct or alteration in the composition of secretions may be the cause of cyst formation (1). Histopathologically, these lesions are simple benign cysts lined by a layer of nonciliated cuboidal epithelial cells and occasional goblet ...
A collaborative research team led by GSDM Professor of Molecular & Cell Biology and Associate Dean for Research Dr. Maria Kukuruzinska has identified a molecular explanation for at least a subset of cases of the disabling autoimmune disease Sjogrens Syndrome. The teams findings are published in the journal Laboratory Investigation.. Sjogrens Syndrome (SS) is a complex autoimmune disease manifested by defective salivary and lacrimal gland function. The most common symptoms of SS are dry mouth and dry eyes, which can lead to dental cavities, vision problems, and thrush. SS also carries an increased risk for developing B cell lymphoma.. While the cause of SS has remained controversial, the prevailing dogma has been that immune system pathology drives the disease. The manuscript now provides evidence that defects in cell polarity, or cell structure, are the likely culprits in at least some cases of SS.. The collective expertise of the international multidisciplinary team of investigators-which ...

Punctal plugs are available in two styles. One plug feature completely stops tear flow and it is placed at the opening of the tear duct. The other plug type is designed to control the flow of tears outward and is usually imbedded into the tear duct channel.
The Chloropidae are a family of flies commonly known as frit flies or grass flies. About 2000 described species are in over 160 genera distributed worldwide. These are usually very small flies, yellow or black and appearing shiny due to the virtual absence of any hairs. The majority of the larvae are phytophagous, mainly on grasses, and can be major pests of cereals. However, parasitic and predatory species are known. A few species are kleptoparasites. Some species in the genera Hippelates and Siphunculina (S. funicola being quite well known in Asia) are called eye gnats or eye flies for their habit of being attracted to eyes. They feed on lachrymal secretions and other body fluids of various animals, including humans and are of medical significance. There are scant records of chloropids from amber deposits, mostly from the Eocene and Oligocene periods although some material may suggest the family dates back to the Cretaceous or earlier. For terms see Morphology of Diptera. Chloropidae are ...

Dry Eye (Keratoconjunctivitis Sicca) Symptoms and Treatments in Dogs - Keratoconjunctivitis sicca is a disorder of the tear glands that results in insufficient aqueous (water like) tear production and leads to a dry cornea. The tear film contains less of the aqueous layer and more of the mucus layer.
Exocrine glands are glands that produce and secrete substances onto an epithelial surface by way of a duct. Examples of exocrine glands include sweat, salivary, mammary, ceruminous, lacrimal, sebaceous, and mucous. Exocrine glands are one of two types of glands in the human body, the other being endocrine glands, which secrete their products directly into the bloodstream. The liver and pancreas are both exocrine and endocrine glands; they are exocrine glands because they secrete products-bile and pancreatic juice-into the gastrointestinal tract through a series of ducts, and endocrine because they secrete other substances directly into the bloodstream. Exocrine glands contain a glandular portion and a duct portion, the structures of which can be used to classify the gland. The duct portion may be branched (called compound) or unbranched (called simple). The glandular portion may be tubular or acinar, or may be a mix of the two (called tubuloacinar). If the glandular portion branches, then the ...

John D. Sheppard, MD, Chief Medical Editor. The understanding of keratoconjunctivitis sicca (KCS), also known as Chronic Dry Eye Disease or Dry Eye Syndrome, has changed dramatically in recent years. Until the late 1990s, KCS was thought to have been due to aqueous insufficiency. Today, KCS is understood to be a multifactorial disease that also involves inflammation of the ocular surface and lacrimal gland, neurotrophic deficiency, and meibomian dysfunction. Delay in treatment may cause disease of the ocular surface. The American Academy of Ophthalmology, in November 2003, released guidelines for treatment that recognizes these contributing factors to the epidemiology of KCS and provides evidence-based guidelines for treatment. This collection of review articles places the etiology of KCS, the AAO recommendations, and the most recent advances in treatment in perspective for practitioners, MCOs, and physicians and pharmacists who serve on pharmacy and therapeutic committees..
