In taxonomy, the Korarchaeota are a phylum of the Archaea. The name is derived from the Greek noun koros or kore, meaning young man or young woman, and the Greek adjective archaios which means ancient. They are also known as Xenarchaeota. Korarchaeota is regarded as a phylum, which itself is part of the archaeal TACK superphylum which encompasses Thaumarchaeota, Aigarchaeota, Crenarchaeota and Korarchaeota. Analysis of their 16S rRNA gene sequences suggests that they are a deeply branching lineage that does not belong to the main archaeal groups, Crenarchaeota and Euryarchaeota. Analysis of the genome of one korarchaeote that was enriched from a mixed culture revealed a number of both Crenarchaeota- and Euryarchaeota-like features and supports the hypothesis of a deep-branching ancestry. In recent years several laboratories have developed effective plating techniques, identifying genetic markers that do not target cell wall synthesis, fusing archaeal promoters with recombinant genes, and isolating native vectors and promiscuous nonnative vectors. This chapter focuses on tractable systems that are currently available for the Archaea. Due to fundamental differences between gene transfer systems for each archaeal branch, the chapter is divided into three inclusive sections covering the halophilic and methanogenic Euryarchaeota and the hyperthermophilic Crenarchaeota. Despite varying degrees of difficulty growing Archaea, all three systems are routinely used by laboratories conducting research on archaeal genetics and can be mastered by anyone with a fundamental knowledge of microbial genetic techniques. A tripartite comparison of Archaea phylogeny and taxonomy at and above the rank order is reported: (1) the whole-genome-based and alignment-free CVTree using 179 genomes; (2) the 16S rRNA analysis exemplified by the All-Species Living Tree with 366 archaeal sequences; and (3) the Second Edition of Bergeys Manual of Systematic Bacteriology complemented by some current literature. A high degree of agreement is reached at these ranks. From the newly proposed archaeal phyla, Korarchaeota, Thaumarchaeota, Nanoarchaeota and Aigarchaeota, to the recent suggestion to divide the class Halobacteria into three orders, all gain substantial support from CVTree. In addition, the CVTree helped to determine the taxonomic position of some newly sequenced genomes without proper lineage information. A few discrepancies between the CVTree and the 16S rRNA approaches call for further investigation.
A gene for membrane-integral inorganic pyrophosphatase (miPPase) was found in the composite genome of the extremophile archaeon Candidatus Korarchaeum cryptofilum (CKc). This korarchaeal genome shows unusual partial similarity to both major archaeal phyla Crenarchaeota and Euryarchaeota. Thus this Korarchaeote might have retained features that represent an ancestral archaeal form, existing before the occurrence of the evolutionary bifurcation into Crenarchaeota and Euryarchaeota. In addition, CKc lacks five genes that are common to early genomes at the LUCA border. These two properties independently suggest a pre-LUCA evolutionary position of this extremophile. Our finding of the miPPase gene in the CKc genome points to a role for the enzyme in the energy conversion of this very early archaeon. My work is focused on plant community restoration in the Great Basin through the development of native seed supplies. From utility corridors to mining and grazing to wildfire, disturbances in the Great Basin over the last decade impact on average about 1,000,000 acres annually necessitating reseeding on hundreds of thousands of acres. Federal policy changes implemented over a decade ago require, with exceptions, the use of native plant materials for restoration projects on the 95 million acres of federal lands in the Great Basin. The result is a huge demand for native plant materials. Private production of native grass cultivars and wild land collected native shrubs largely fills the volume requirement to meet the restoration need but lacks the diversity of sources restoration practitioners desire to return appropriately adapted and genetically diverse population to the landscape. Natural and anthropogenic boundaries have been shown to affect population dynamics and population structure for many species with movement patterns at the landscape level. The Great Basin desert climate begins with the Sierra Nevada in eastern California. Rising 14,000 feet (4,300 m) above sea level, this mountain range casts a large rain shadow over the desert. Quantifying genomic variation provides information that can be used to understand the evolutionary history of populations. Here, I examined populations of Euphilotes pallescens, a species of butterfly that lives within the Great Basin of western North America. I genotyped 376 butterflies at over 90,000 loci to address questions surrounding gene exchange among lineages of E. pallescens and other geographically proximate Euphilotes species. I also investigated what relative contributions of historical and contemporary admixture to the patterns I saw. I stratified loci into common and rare loci based on minor allele frequencies to investigate historical and contemporary genetic structure, respectively. I used a Bayesian hierarchical model to visualize and quantify genetic variation in two analyses: one included only E. pallescens populations, while the second analysis was performed at the genus-level. In order to obtain a suitable red phosphor for mercury-free lamps and plasma display panels (PDPs), samples of trivalent europium-activated polyphosphate LiGd(PO3)4:Eu3+ (LGP:Eu3+) were prepared by a solid-state reaction technique at high temperature. The vacuum ultraviolet (VUV)-visible spectroscopic properties were investigated. Because the phosphor LiGd1-xEux(PO3)4 for x = 0.50 shows broad and strong absorption in VUV region and exhibits intensive emission under 147/172 nm excitation in comparison with the PDP commercial red phosphor (Y, Gd)BO3:Eu3+, it is considered to be a promising red phosphor for mercury-free lamps and plasma display panels application.. ©2009 Optical Society of America. This odd jay, looking more like a small blue-gray crow, lives mainly in the Great Basin region of the west. Appropriately named, it feeds heavily on the seeds of pinyon pines, and its distribution is tied closely to the range of these trees. Pinyon Jays are sociable at all seasons, traveling in flocks, nesting in colonies. When on the move they fly close together, giving harsh nasal calls.
