The present study documents a number of important features relevant to signal transduction systems operating in the IMCD. First, increased extracellular tonicity reduces expression of the ET gene, which typically displays robust expression in IMCD cells under isotonic conditions. Second, endogenous ET stimulates eNOS and inhibits NPR-A gene expression in these cells. These latter effects appear to be maximally activated under basal conditions in which administration of exogenous ET had little additional activity. Third, the osmotic stimulation of NPR-A and suppression of eNOS gene expression noted previously does not appear to be mechanistically linked solely to the osmotic inhibition of ET gene expression.. Vasopressin is the primary hormone regulating urine-concentrating ability. Vasopressin phosphorylates the UT-A1 urea transporter in rat inner medullary collecting ducts (IMCDs). To assess the effect of UT-A1 phosphorylation at S486, we developed a phospho-specific antibody to S486-UT-A1 using an 11 amino acid peptide antigen starting from amino acid 482 that bracketed S486 in roughly the center of the sequence. Our results also demonstrate that 18-OH-DOC can stimulate Na+ transport by these cells and that the majority of its action appears to be mediated via the mineralocorticoid receptor rather than the glucocorticoid receptor.. Mathematical modeling techniques have been useful in providing insights into biological systems, including the kidney. This article considers some of the mathematical models that concern urea transport in the kidney. Modeling simulations have been conducted to investigate, in the context of urea cycling and urine concentration, the effects of hypothetical active urea secretion into pars recta. Simulation results suggest that active urea secretion induces a "urea-selective" improvement in urine concentrating ability. Two urea transporters, UT-A1 and UT-A3, are expressed in the kidney terminal inner medullary collecting duct (IMCD) and are important for the production of concentrated urine. UT-A1, as the largest isoform of all UT-A urea transporters, has gained much attention and been extensively studied; however, the role and the regulation of UT-A3 are less explored. In this study, we investigated UT-A3 regulation by glycosylation modification. A site-directed mutagenesis verified a single glycosylation site in UT-A3 at Asn279. Loss of the glycosylation reduced forskolin-stimulated UT-A3 cell membrane expression and urea transport activity. UT-A3 has two glycosylation forms, 45 and 65 kDa. Lithium is the most common cause of nephrogenic diabetes insipidus (Li-NDI). Hydrochlorothiazide (HCTZ) combined with amiloride is the mainstay treatment in Li-NDI. The paradox antidiuretic action of HCTZ in Li-NDI is generally attributed to increased sodium and water uptake in proximal tubules as a compensation for increased volume loss due to HCTZ inhibition of the NaCl-co-transporter (NCC), but alternative actions for HCTZ have been suggested. In the kidney, the antidiuretic hormone vasopressin (AVP) is a critical regulator of water homeostasis by controlling the water movement from lumen to the interstitium for water reabsorption and adjusting the urinary water excretion. In normal physiology, AVP is secreted into the circulation by the posterior pituitary gland, in response to an increase in serum osmolality or a decrease in effective circulating volume. When reaching the kidney, AVP binds to V2 receptors on the basolateral surface of the collecting duct epithelium, triggering a G-protein-linked signaling cascade, which leads to water channel aquaporin-2 (AQP2) vesicle insertion into the apical plasma membrane. Abstract: Aldosterone effects are mediated by the mineralocorticoid receptor (MR), a transcription factor highly expressed in the distal nephron. Given that MR expression level constitutes a key element controlling hormone responsiveness, there is much interest in elucidating the molecular mechanisms governing MR expression. To investigate whether hyper- or hypotonicity could affect MR abundance, we established by targeted oncogenesis a novel immortalized cortical collecting duct (CCD) cell line and examined the impact of osmotic stress on MR expression. Our study contributes to the literature that show that hyperosmotic stress induces an ER stress‐like response. Water restriction or dDAVP infusion, which both increase medullary osmolality, increased Grp78 and ATF4 expression in the inner medulla (van Balkom et al. 2004; Cai et al. 2006, 2010). Conversely, GRP78 protein abundance decreased in IMCD of animals subjected to vasopressin escape (Hoorn et al. 2005). Expression levels of ER stress‐responsive genes CHOP and ATF3 were increased by either NaCl or urea challenge in cultured IMCD cells (Kultz et al. 1998; Zhang et al. 1999; Tian and Cohen 2002) and several ER stress‐responsive genes were upregulated by NaCl challenge in renal fibroblast cell lines (Dihazi et al. 2011). The renal collecting duct is the nephron segment where the final urine content of acid equivalents and inorganic ions are determined. The role of two different cell types present in this nephron segment has been determined many years ago: principal cells that express the epithelial sodium channel ENaC and aquaporin 2, regulate electrolyte reabsorption, while intercalated cells, which express acid-base transporters and vacuolar H+-ATPase, maintain an apropriate acid-base balance. Recent evidence challenges this historical view. Rather than having independent and non-overlapping functions, the two cell types in the collecting duct appear to functionally cooperate to regulate acid-base and volume homeostasis via complex paracrine and endocrine interplay. This review summarizes these recent findings.
