Adult skin stem cells are recognized as potential therapeutics to rejuvenate aged skin. We previously demonstrated that human dermal stem/progenitor cells (hDSPCs) with multipotent capacity could be enriched from human dermal fibroblasts using collagen type IV. However, the effects of hDSPCs on cellular senescence remain to be elucidated. In the present study, we investigated whether conditioned medium (CM) collected from hDSPC cultures (hDSPC-CM) exhibits beneficial effects on senescent fibroblasts. We found that hDSPC-CM promoted proliferation and decreased the expression level of senescence-associated β-galactosidase in senescent fibroblasts. In addition, p53 phosphorylation and p21 expression were significantly reduced in senescent fibroblasts treated with hDSPC-CM. hDSPC-CM restored the expression levels of collagen type I, collagen type III, and tissue inhibitor of metalloproteinase, and antagonized the increase of matrix metalloproteinase 1 expression. Finally, we demonstrated that hDSPC-CM
First, light entering the eye must pass through its clear outer surface, or cornea, which is kept smooth, clean, and lubricated by a thin coating of tears. These tears are produced by specialized cells in the eyelids, but damage from environmental factors like smoke, pollution, and ultraviolent light can, over time, cause the eyelids to become inflamed, potentially impairing tear production. This condition, commonly known as blepharitis, creates the sensation that a cloudy film is covering the eye, partly obscuring vision. As evaporation of the tears and drying out of the corneal surface gets worse, the condition can evolve into chronic dry eye, which may require the use of surgical tear plugs to fully relieve. In the early stages, however, blepharitis can be treated simply with warm compresses, artificial tears, oral antibiotics and even topical immunosuppressive agents that decrease the inflammatory response.. Once the light enters the eye it is focused by a clear, flexible lens, which can ...
Since the adult myocardium lacks the inherent ability to repair itself following ischemic injury, exogenous cell sources for the repair of infarcted myocardium have been investigated, and it has been demonstrated that transplanted cells may impart functional benefits via the secretion of growth factors and extracellular matrix (ECM) molecules.  Such mitigation of tissue repair by cell-derived molecules represents a significant paradigm shift in the therapeutic application of stem and progenitor cells and provides a novel approach to myocardial repair.  My research aims are to (1) examine the effects of three-dimensional cell culture systems on adult progenitor cell differentiation, growth factor synthesis, and ECM production, and (2) to determine the effects of progenitor cell-derived factors on ischemic cells and tissues in vitro and in vivo.  These studies should yield new insights into the cues presented to injured myocardium by exogenously administered ...

Group B streptococci (GBS) are pathogens of both neonates and adults, with serotype III strains in particular being associated with invasive disease and meningitis. In this study, a novel GBS surface antigen, ε, was found to be co-expressed with the previously reported δ antigen on an identical subset of serotype III GBS. Expression of δ/ε on the surface of serotype III GBS was shown to distinguish the restriction digest pattern (RDP) III-3 and multilocus sequence typing (ST)-17 lineage. ε-Specific antibodies were reactive with a unique, high-molecular-mass, serine-rich repeat protein (Srr-2) found exclusively in RDP III-3 strains. The gene encoding Srr-2 was located within a putative accessory secretory locus that included secY2 and secA2 homologues and had a genetic organization similar to that of the secY2/A2 locus of staphylococci. In contrast, serotype III δ/ε-negative strains and strains representative of serotypes Ia, Ib, Ic and II shared a common Srr-encoding gene, srr-1, and an

At least six mutations in the KRT4 gene have been found to cause white sponge nevus, a condition that results in the formation of white patches of tissue called nevi (singular: nevus) that appear as thickened, velvety, sponge-like tissue. These nevi most often occur on the mouth (oral) mucosa (plural: mucosae). Rarely, white sponge nevus occurs on the mucosae of the nose, esophagus, genitals, or anus.. The KRT4 gene mutations that cause white sponge nevus disrupt the structure of keratin 4. As a result, keratin 4 does not fit together properly with keratin 13, leading to the formation of irregular intermediate filaments that are easily damaged with little friction or trauma. Fragile intermediate filaments in the oral mucosa might be damaged when eating or brushing ones teeth. Damage to intermediate filaments leads to inflammation and promotes the abnormal growth and division (proliferation) of epithelial cells, causing the mucosae to thicken and resulting in white sponge nevus. ...
Short-term efficacy of calcium fructoborate on subjects with knee discomfort: a comparative, double-blind, placebo-controlled clinical study Zbigniew Pietrzkowski,1 Michael J Phelan,2 Robert Keller,3 Cynthia Shu,1 Ruby Argumedo,1 Tania Reyes-Izquierdo11FutureCeuticals, Inc., Applied BioClinical Laboratory; 2Department of Statistics, School of Information and Computer Science, University of California at Irvine; 3NutraClinical Inc., Irvine, CA, USAAbstract: Calcium fructoborate (CFB) at a dose of 110 mg twice per day was previously reported to improve knee discomfort during the first 14 days of treatment. In this study, 60 participants with self-reported knee discomfort were randomized into two groups receiving CFB or placebo. Initial levels of knee discomfort were evaluated by Western Ontario and McMaster Universities Arthritis Index (WOMAC) and McGill Pain Questionnaire (MPQ) scores at the beginning of the study and also at 7 and 14 days after treatment. Results showed that

The antinuclear antibodies (ANA) test has been a cornerstone of the evaluation of connective tissue disease. The aim of this study was to investigate the diagnostic value of the ANA test in pleural or pericardial effusions of unknown causes. Over a 3-yr period, a total of 126 pleural fluid and 30 pericardial fluid samples were analysed. ANA tests were performed using a commercially available kit. The ANA kit used an indirect immunofluorescent antibody method with a human epithelial (HEP-2) cell line as substrate. Patients with high fluid ANA titre (,1:160) received a second aspiration 2 weeks after the initial aspiration if diagnosis was not confirmed. ANA results were positive in 39 pleural and 10 pericardial fluid samples. All but one of the effusions with positive ANA testing were exudative. Eleven pleural or pericardial effusions due to active systematic lupus erythematosus were identified and all had high ANA titres (1:160) with various staining patterns. Thirty-eight of 145 patients (26%) ...
Human epithelial (HEp-2) cell specimens is obtained from indirect immunofluorescence (IIF) imaging for diagnosis and management of autoimmune diseases. Analysis of HEp2 cells is important and in this work we consider automatic cell segmentation and classification using spatial and texture pattern features and random forest classifiers. In this paper, we summarize our efforts in classification and segmentation tasks proposed in ICPR 2016 contest. For the cell level staining pattern classification (Task 1), we utilized texture features such as rotational invariant co-occurrence (RIC) versions of the well-known local binary pattern (LBP), median binary pattern (MBP), joint adaptive median binary pattern (JAMBP), and motif cooccurrence matrix (MCM) along with other optimized features. We report the classification results utilizing different classifiers such as the k-nearest neighbors (kNN), support vector machine (SVM), and random forest (RF). We obtained the best mean class accuracy of 94.29% for ...
Abstract Purpose: To compare short-term efficacy of cough technique (CT) versus forced expiration technique (FET) on sputum removal in individual patients with cystic fibrosis (CF), and to investigate patient´s self-reported preferences of the two techniques. Background: Physiotherapy for Airway Clearance is recommended in CF but no strong evidence exist that one technique is better than another. Sputum removal is essential, with FET commonly used, though CT might represent an alternative. Method: Randomized controlled individual trials (N of 1 RCTs) with six patients were conducted. Each trial included 8 weeks of treatment periods with two interventions each week. Primary outcome measure was sputum wet weight (g). Secondary outcomes were self-reported preference and perceived utility value. In addition, spirometry, oxygen saturation and heart rate were measured and health-related quality of life before and after the study. Results: The majority removed more sputum with CT than FET. In three ...
Preliminary data from the HIVNAT PK laboratory indicate that out of 5/60 patients treated with nevirapine (200 mg bid) and rifampicin had sub-therapeutic nevirapine levels (,3.0 mg mg/L). In a control group of 38 patients using nevirapine without rifampicin there were no sub-therapeutic levels. A dose increase of nevirapine while patients who are receiving that rifampicin may be required. Both nevirapine and rifampicin are tepatotoxic agents as are other agents used in treatment of HIV or tuberculosis. Using a higher nevirapine may prevent the occurrence of sub-therapeutic nevirapine levels, but may also induce more liver toxicity. To address these issues, we designed a randomized prospective study to evaluate the safety, efficacy and pharmacokinetics of nevirapine 400 mg/day versus 600 mg/day with a two weeks lead-in 200 mg/day and 400 mg/day respectively, in TB-HIV co-infected patients who taking rifampicin and short-term efficacy and toxicity ...

There are many ways adults can increase their participation in PA [1]. The goal of this pilot study was to examine feasibility and short-term efficacy of a PA program that targeted changing a traditionally sedentary leisure-time behavior (TV viewing) into an active one. The results suggest that a small behavior change, like stepping in place during TV commercials, was both effective and acceptable to adults. At baseline, no participants were meeting the guidelines for aerobic activity (≥150 min/wk). TV Commercial Stepping and Walking both resulted in about 70% of the previously inactive participants meeting the volume requirements of the 2008 PA guidelines, but only the walking group accumulated their PA in continuous bouts of 10 minutes or more. Despite the different approaches to increasing PA, the volume of exercise for both groups was approximately equal. Stepping in place during commercials for least 90 min/d of TV viewing resulted in an increase in daily steps (~3000) roughly equivalent ...
OBJECTIVES: To study the pharmacokinetics and short-term efficacy of low and standard dose lopinavir/ritonavir and saquinavir combinations in Thai, human immunodeficiency virus (HIV)-infected, treatment-naive patients. METHODS: In this open-label, 24-week, prospective study, 48 treatment-naive patients were randomized to lopinavir/ritonavir 400/100 mg+saquinavir 1000 mg twice daily (arm A), lopinavir/ritonavir 400/100 mg+saquinavir 600 mg twice daily (arm B), lopinavir/ritonavir 266/66 mg+saquinavir 1000 mg twice daily (arm C), or lopinavir/ritonavir 266/66 mg+saquinavir 600 mg twice daily (arm D). A 12 h. pharmacokinetic profile in all patients was performed. Plasma concentrations of saquinavir and lopinavir were determined using an HPLC technique. HIV-1 RNA was measured over 24 weeks. RESULTS: Forty-three subjects were included in the pharmacokinetic analysis. The total exposure differed significantly for the different arms. Median values for lopinavir area under the curve at 0-12 h were ...

Vasioukhin V., Bowers E., Bauer C., Degenstein L., Fuchs E.. We have generated an epidermis-specific desmoplakin (DP) mouse knockout, and show that epidermal integrity requires DP; mechanical stresses to DP-null skin cause intercellular separations. The number of epidermal desmosomes in DP-null skin is similar to wild type (WT), but they lack keratin filaments, which compromise their function. DP-null keratinocytes have few desmosomes in vitro, and are unable to undergo actin reorganization and membrane sealing during epithelial sheet formation. Adherens junctions are also reduced. In vitro, DP transgene expression rescues these defects. DP is therefore required for assembly of functional desmosomes, maintaining cytoskeletal architecture and reinforcing membrane attachments essential for stable intercellular adhesion.. Nat. Nonsteroidal anti-inflammatory drugs, especially Indomethacin and Ibuprofen, have been widely used for both prevention and treatment of PDA. Short-term efficacy of Indomethacin or Ibuprofen is equivalent, while Ibuprofen results show a higher safety profile. Ibuprofen is associated with fewer clinical gastrointestinal and renal side effects with respect to Indomethacin even if subclinical potential effects are reported. When administered as prophylaxis, Ibuprofen has no effects on prevention of intraventricular haemorrhage unlike Indomethacin. Considering the potential adverse effects of both these drugs, a careful monitoring during and after the treatment ...

Ive had success cleaning the marks off our white toilet seat using a magic sponge (NOT the type that they use on the football pitch!!). Its a cleaning product thats great for loads of different stains. It looks just like a fine white sponge which you cut a piece off, dip it in water and gently rub at the stain. Its basically a very fine abrasive that is good at removing surface marks without damaging the finish underneath. It even cleaned the crayon off our matt-painted walls created by my art-loving daughter when she was a toddler - saved me from repainting the whole room ...
