The detection of lymph node metastasis by RT-PCR (37-40) and by QRT-PCR (12, 19-25) has been studied previously. CK19 mRNA has been described as having the highest sensitivity at nearly 90%. However, there are drawbacks using CK19 mRNA due to the concomitant amplification of pseudogenes in genomic DNA that lead to false positive results. For this reason, a combination of two or three markers has been used.. We evaluated 45 potential mRNAs and finally selected CK19 mRNA as the best marker for the OSNA assay. To use CK19 mRNA as a marker, we designed RT-LAMP primers that do not amplify the known CK19 pseudogenes (see Materials and Methods). Squamous cell carcinoma antigen: a role in the early identification of nodal metastases in men with squamous cell carcinoma of the penis. To evaluate whether serum squamous cell carcinoma antigen (SCCAg) measurements may be of use in identifying nodal metastases in patients with SCC of the penis after treating the primary tumour. The levels of SCCAg were analysed in 11 men with penile SCC between 1994 and 2001. An elevated SCCAg level had a sensitivity of 57% (95% confidence interval, CI, 18-90%) and a specificity of 100% (CI 40-100%) for nodal metastases. Levels of SCCAg increased exponentially in patients who developed nodal metastases after treatment of the primary tumour, and were elevated before clinical or radiological evidence of nodal disease. Despite extensive research, the role of the commonly employed tumour markers in the diagnosis of lung carcinoma is yet to be clarified. The utility of a new marker, CYFRA 21-1, in the preoperative evaluation of patients with bronchogenic carcinoma was investigated. CYFRA 21-1 was determined with a radiometric assay in serum of 280 patients with lung cancer and 208 patients with various nonmalignant lung diseases. The levels of the marker were significantly higher in lung cancer patients. Among benign lung diseases, elevated CYFRA 21-1 levels were found in pulmonary fibrosis. Using a cut-off of 3.2 ng.ml-1 (95th percentile of levels obtained in benign lung disease), the total sensitivity of the marker was 48%.
