To investigate cell surface-associated keratan sulfate on the corneal endothelium. METHODS. Immunolabeling techniques were used at the light, scanning, and transmission electron microscopic level to localize keratan sulfate on the corneal endothelium. The investigation included human, bovine, and rabbit corneal endothelia. A quantitative study of the relationship between cell size and keratan sulfate levels was conducted on normal bovine corneal endothelium. Changes in the distribution of keratan sulfate and chondroitin sulfate on endothelial cell surfaces were investigated on organ cultured bovine corneas during endothelial wound healing. We have prepared a monoclonal antibody, named MZ15, that specifically binds keratan sulfate. Immunofluorescence studies showed that the distribution of keratan sulfate in articular cartilage was not uniform: the amount of keratan sulfate increased with distance from the articular surface. Two subpopulations of chondrocytes could be distinguished after isolation from cartilage by the presence or absence of cell surface keratan sulfate. Keratan sulfate-negative chondrocytes were shown to come from the upper cartilage layers. There was therefore a direct correlation between biochemical heterogeneity of cartilage matrix and heterogeneity within the chondrocyte population. Keratan sulfate (KS), also called keratosulfate, is any of several sulfated glycosaminoglycans (structural carbohydrates) that have been found especially in the cornea, cartilage, and bone. To further understand the role of lumican as a myokine affecting adjacent bone metabolism, we investigated the effects of lumican on osteoblast biology. Mucopolysaccharidoses (MPS) are the group of lysosomal storage disorders caused by deficiencies of enzymes involved in the stepwise degradation of glycosaminoglycans. To identify brain pathology common for neurological MPS, we conducted a comprehensive analysis of brain cortex tissues from post-mortem autopsy materials of eight patients affected with MPS I, II, IIIA, IIIC, and IIID, and age-matched controls. Cornea plana congenita (CNA2) is a recessively inherited eye disease affecting development of the cornea, and leading to moderate impairment of visual capacity. The corneal changes are caused by structural abnormalities in keratocan, a leucine-rich protein component of a small proteoglycan, which together with other keratan sulfate proteoglycans is important for the transparency of the cornea. The cornea is thin and unusually flat, corneal limbus is hazy, opacities are frequent in the parenchyma, and arcus senilis developes at an early age. Hyperopia, up to +10 D, is the consequence of corneal changes.

Saline extracts of potato tubers contain a potent hemagglutininin that is not inhibited by monosaccharides. Carbohydrate inhibition studies have shown that the lectin has an extended binding site that can accommodate up to four carbohydrate units. The lectin binds oligosaccharides containing β(1,4)-linked GlcNAc. The tetrasaccharide is approximately 20 times more potent an inhibitor than the disaccharide 1 . STA also reacts with poly N-acetyllactosamine structures from N-linked oligosaccharides, albeit rather weakly 2 . Nevertheless, it is useful for histochemical staining of these structures 3 . Many lectins do not react with sulfated sugars, however, STA does bind to chitin sulfates and keratan sulfates 4 .
