Coptotermes formosanus Shiraki is the most invasive termite species and has the largest economic impact of all the subterranean termites in the US. Although a lot of work has been done on the determination of chemical composition of frontal gland secretion from different families of termites, the protein component of those secretions was relatively overlooked. This study started with the observation that proteins are present in the frontal gland secretion of Coptotermes formosanus Shiraki soldiers. The goal was to characterize one of these proteins by identifying its DNA sequence which would lead to protein sequence, function, structure and role it plays in the defense of Coptotermes formosanus Shiraki. The protein cloned named TFP4 and analysis of its sequence revealed a molecular weight of 6853 Daltons. The sequence was compared with those of known proteins in NCBIs data base and similarities with other proteins analyzed. Two functions were assigned to TFP4, lysozyme and serine protease inhibitor.

A previously undescribed anaerobic, heterofermentative, non-spore-forming, Gram-positive rod was isolated from the hindgut of the Formosan subterranean termite Coptotermes formosanus Shiraki. The DNA G+C content of this bacterium was 37·8 mol%. Sequence analysis of the 16S rRNA gene revealed that this organism is related to, but distinct from, several genera of lactic acid bacteria, principally several species of the genus Enterococcus. Phenotypic traits that serve to separate this organism from related genera include high levels of the cellular fatty acid C18 : 1ω9c and the production of ethanol along with lactic acid as fermentation products. Based on the collected phylogenetic and phenotypic evidence, it is proposed that the unknown organism represents a novel species in a new genus, Pilibacter termitis gen. nov., sp. nov. The type strain is TI-1T (=ATCC BAA-1030T=CCUG 49613T).

In termites, the soldier caste, with its specialized defensive morphology, is one of the most important characteristics for sociality. Most of the basal termite species have both male and female soldiers, and the soldier sex ratio is almost equal or only slightly biased. However, in the apical lineages (especially family Termitidae), there are many species that have soldiers with strongly biased sex ratio. Generally in termites, since high juvenile hormone (JH) titer is required for soldier differentiation from a worker via a presoldier stage, it was hypothesized that the biased soldier-sex ratio was caused by differences in JH sensitivity and/or JH titer between male and female workers. A novel Gram-stain-positive bacterium designated CMU-NKS-70T was isolated from a subterranean termite nest and characterized using a polyphasic approach. The strain exhibited branching, pinkish-cream aerial mycelium and cream-brown substrate mycelium, and formed chains of rod-like spores. The 16S rRNA gene sequence analyses indicated that strain CMU-NKS-70T belonged to the genus Pseudonocardia , showing high similarity with Pseudonocardia oroxyli D10T (98.9 % 16S rRNA gene sequence similarity), Pseudonocardia xishanensis YIM 63638T (98.9 %) and Pseudonocardia kujensis A4038T (98.5 %). There are four major research foci in my laboratory. First, we are interested in elucidating and understanding the molecular mechanisms involved in the survival of pathogenic bacteria in nature and the contribution of these mechanisms to aid these pathogens in their ability to cause human diseases. The frontal glands of the soldier caste termites comprising one third of the body mass, contain a secretion expelled through a foramen in defense. The small molecule composition of the frontal gland secretion is well-characterized, but the proteins remain to be identified. Herein is reported the structure and function of one of several proteins found in the termite defense gland secretion. TFP4 is a 6.9 kDa, non-classical group 1 Kazal-type serine protease inhibitor with activity towards chymotrypsin and elastase, but not trypsin. The 3-dimensional solution structure of TFP4 was solved with nuclear magnetic resonance spectroscopy, and represents the first structure from the taxonomic family, Rhinotermitidae. Based on the structure of TFP4, the protease inhibitor active loop (Cys8 to Cys16) was identified.

Our results show that colonies of R. virginicus undergo AQS as described previously in the Japanese subterranean termite, R. speratus [4]. As is typical of subterranean termites, colonies of both R. speratus and R. virginicus are founded by monogamous pairs of primary reproductives [1,19,20], but at some point, relatively early in the colony life cycle, the primary queen is replaced by her parthenogenetically produced neotenic daughters. This study was carried out to find the effect of some commonly present plant products on termite species (Heterotermis indicola) which are a major threat worldwide. Three different concentrations (20%, 10%, 5%) of various solutions consisting of garlic, Neem and tobacco was used in hot and cold water. Hot and cold water results showed that a high concentration treatment of the garlic (20%) solution caused 100% mortality after 48 hours, but in the case of the tobacco concentration (20%) of the solution in only cold water resulted in 100% mortality. As the concentration become lower, the efficiency becomes slower. 100% mortality in the 10% & 5% solutions of garlic and tobacco occurred in different days. In case of Neem, both hot and cold water was found less effective as compared to garlic and tobacco. The mortality occurred using Neem was significantly less than garlic and tobacco.
