N2 - Isoketals (IsoKs) are γ-ketoaldehydes formed via the isoprostane pathway of arachidonic acid peroxidation and are among the most reactive by-products of lipid peroxidation. IsoKs selectively adduct to protein lysine residues and are highly cytotoxic, but the targets and molecular events involved in IsoK-induced cell death are poorly defined. Our previous work established that physiologically relevant aldehydes induce mitochondrial dysfunction (Kristal et al., J. Biol. Chem. 271:6033-6038; 1996). We therefore examined whether IsoKs induced mitochondrial dysfunction. Incubation of mitochondria with synthetic IsoKs in the presence or absence of Ca2+ was associated with alterations in ...
Recent studies have focused on the role of oxidative stress in cardiovascular disease. In the present study, increased urinary 15-F2t-isoprostane concentrations in patients with CHF with acute exacerbation gradually decreased in proportion to the severity of CHF during hospitalisation. Urinary 15-F2t-isoprostane concentrations were significantly correlated with the concentrations of plasma BNP and serum IL-6, well established markers of CHF. These findings suggest that urinary 15-F2t-isoprostane may be a marker of morbidity, as well as of oxidative stress, in patients with CHF.. Recent studies have focused on 15-F2t-isoprostane, an index of lipid peroxidation, to investigate the involvement of oxidative stress in heart failure.19-22 Quantification of 15-F2t-isoprostane has been suggested to be a reliable measure of oxidative injury in vivo.8 Cracowski and colleagues19 investigated urinary 15-F2t-isoprostane formation in 25 patients suffering from severe heart failure and showed that ...
The present study reveals that 15-F2t-IsoP causes greater constriction of periventricular microvessels of the fetus than of newborn and juvenile animals due to greater thromboxane generation through a previously undescribed mechanism of action of 15-F2t-IsoP, which seems to involve activation of distinct calcium channels and an interaction between vascular and perivascular cells. Data indicate that 15-F2t-IsoP elicits vasoconstriction by increasing the release of thromboxane from brain astroglial and endothelial but not vascular smooth muscle cells; the increased thromboxane synthesis seems to result from increased entry of extracellular calcium into cells, possibly through N-type voltage-dependent calcium channels in astrocytes and non-voltage-dependent calcium channels in endothelial cells.. The effects of 15-F2t-IsoP have been found to be markedly inhibited by thromboxane receptor blockers7 22 23 24 ; however, ligand binding studies suggest that 15-F2t-IsoP does not directly interact with the ...
Previous studies of the effects of 8-isoprostanes on airway smooth muscle have been limited to the study of the excitatory effects of only one isoprostane, 8-iso-PGF2α, under the assumption that this isoprostane is the predominant form generated during free radical attack of cell membranes. In human large airways, the effects of 8-iso-PGF2α were shown to be mediated by TP receptors, although some heterogeneity in the receptor population involved could not be excluded (Kawikova et al., 1996).. In this study, we found several other isoprostanes can also cause contraction of airway smooth muscle. The 8-isoprostanes we tested generally had a higher contractile potency in human large airways compared with human small airways (Table 1). The two exceptions are 8-iso-PGE1, which had highly variable potency in both preparations, and 8-iso-PGF3α, which had powerful relaxant effects in human large airways that most likely masked any excitatory effects. The order of potency of the compounds in human ...
Our laboratory has been focused on understanding how inflammation, and in particular, the adaptive immune response contributes to hypertension. Several years ago, we found that T cells are essential for the development of hypertension. We have shown that various hypertensive stimuli, including angiotensin II, norepinephrine and DOCA-salt cause activation of T cells and leads to their accumulation in the perivascular fat and kidneys. Our data indicate that T cell-derived cytokines such as IL-17 and TNF-a enhance vasoconstriction and sodium retention, leading to the hypertensive phenotype. Central signals derived from the circumventricular organs contribute to T cell activation, and manipulation of signals from this region affect T cell activation and the eventual elevation in blood pressure caused by angiotensin II. We are attempting to understand mechanisms involved in T cell activation in response to hypertensive stimuli. We have recently shown that gamma-ketoaldehydes, or isoketals adduct to ...

Isoprostanes are stable products of arachidonic acid peroxidation and are regarded as the most reliable markers of oxidative stress in vivo. Here we describe the LC-MS/MS procedure enabling simultaneous determination of four regioisomers (8-iso prostaglandin F2α, 8-iso-15(R)-prostaglandin F2α, 11β-prostaglandin F2α, 15(R)-prostaglandin F2α) in plasma samples collected from mice. The four plasma isoprostanes are determined by LC-ESI-MS/MS with deuterated 8-iso-PGF2α-d4 as an internal standard (I.S.). For plasma samples spiked with the isoprostanes at a level of 200 pg/mL each, the method imprecision has been below 7.1% and mean inaccuracy equaled 8.7%. The applicability of the proposed approach has been verified by the assessment of changes in isoprostane levels in plasma samples derived from mice exposed to tert-butyl hydroperoxide (TBHP), a model inducer of oxidative stress, or to antitumor drug doxorubicin (DOX) known for potent stimulation of redox cycling. Compared to the control group of

Oxidative stress and inflammation are supposed to be the key players of several acute and chronic diseases, and also for progressive aging process. Eicosanoids, especially prostaglandin F-2 alpha (PGF(2 alpha)) and F-2-isoprostanes are endogenous compounds that are involved both in physiology and the above mentioned pathologies. These compounds are biosynthesized mainly from esterified arachidonic acid through both enzymatic and non-enzymatic free radical-catalysed reactions in vivo, respectively. They have shown to possess potent biological activities in addition to their application as biomarkers of oxidative stress and inflammation. Recent advancement of methodologies has made it possible to quantify these compounds more reliably and apply them in various in vivo studies successfully. Today, experimental and clinical studies have revealed that both PGF(2 alpha) and F-2-isoprostanes are involved in severe acute or chronic inflammatory conditions such as rheumatic diseases, asthma, risk factors ...
Oxidative stress is assumed to be the key underlying factor in the pathogenesis of many common diseases. This study describes the basal levels of 8-iso-PGF(2alpha ), a major F(2)-isoprostane and an in vivo oxidative stress biomarker in healthy subjects from three countries, namely Italy, Poland and Sweden, in relation to their smoking habits, age and gender. It studied urinary 8-iso-PGF(2alpha ) in 588 subjects from Sweden (n=220), Italy (n=203) and Poland (n=165). Polish subjects had the highest levels of F(2)-isoprostanes followed by the Swedish and Italians when adjusted for smoking, age, sex and creatinine and the inter-country differences were statistically significant. Smokers had significantly higher levels of 8-iso-PGF(2alpha ) compared to non-smokers in all countries and there was a moderate decrease with age. Women had only slightly lower 8-iso-PGF(2alpha ) than men. There is a difference in F(2)-isoprostane levels in vivo between countries. Smoking, age and gender affect isoprostane ...

Isoprostanes are a recently discovered group of prostaglandin isomers. Results of previous studies suggest that they can be used as oxidative stress markers, because in a number of cardiovascular, pulmonary and neurological diseases their levels in biological samples considerably increase. It...
Future studies are needed to substantiate some of the underlying assumptions made in this investigation. The evidence that F2-isoprostanes relate to hard cardiovascular disease endpoints, or that the notional potent proinflammatory effects of F2-IsoPs are implicated in cardiovascular pathophysiology, is very limited to date. No evidence for an association between increased F2-IsoP levels and increased fatal or non-fatal coronary heart disease incidence was identified in a prospective cohort study that specifically addressed the relationship between markers of oxidation and cardiovascular disease incidence. Rather, changes in F2-isoprostane levels in tissue fluids may relate better to the risk factors of cardiovascular disease, such as diabetes and smoking. The premise that F3-isoprostanes may have anti-inflammatory and protective effects in human tissues also needs to be evaluated, considering the yet poorly documented causal contributions of F2-isoprostanes to cardiovascular disease. This study ...
In the present study, daily supplementation for 2 months with vitamin C alone significantly decreased the plasma levels of F2-isoprostanes in smokers with BMI above the median when compared with those who received placebo. In the mixture group, the treatment effect was smaller and did not achieve statistical significance. Thus, we found that F2-isoprostanes are usable markers of oxidative damage in reasonably large-scale studies and that at least some antioxidants can reduce this biomarker in smokers. These data do not support the hypothesis that a combination of water- and lipid-soluble antioxidants is superior to a single antioxidant (vitamin C) alone in reducing F2-isoprostanes.. Strengths of the study include the randomized placebo-controlled design and the sample size. To our knowledge, few other intervention studies have examined the effect of antioxidants on the F2-isoprostane biomarker (39, 40, 41, 42, 43) ; the largest intervention group sample size was 22 (40) , and only two studies ...

N2 - F2-isoprostanes (F2-iPs) are prostaglandin (PG)-like products of non-enzymatic free radical-catalyzed peroxidation of arachidonic acid that are now widely used as indices of lipid peroxidation in vivo. Knowledge of the metabolic fate of F2-iPs in vivo is still scant, despite its importance for defining their overall formation and biological effects in vivo. Type III F2-iPs, which are diastereoisomers of cyclooxygenase-derived PGF2α, may be metabolized through the pathways of PG metabolism. We therefore studied the in vitro metabolism of eight synthetic Type III F2-iP diastereoisomers in comparison with PGF2α. We used gas chromatography-mass spectrometry and ...
Local production of endothelial NO plays a pivotal role in governing the overall redox state of the vascular wall. A reduction in endothelial NO production would be expected to result in an increase in vascular oxidative stress. Arachidonic acid is a prime target of oxidants and is peroxidized to yield metabolites including F2-isoprostanes. APE1/ref-1+/− mice have been reported to have elevations in circulating markers of oxidative stress such as F2-isoprostanes.19 F2-isoprostanes themselves lead to vasoconstriction by direct action on vascular smooth muscle cells via thromboxane receptors.20 Therefore, in addition to a decrease in NO-mediated smooth muscle relaxation, the hypertension seen in APE1/ref-1+/− mice may also be attributable to the paracrine vasoconstricting effects of F2-isoprostanes and other oxidative stress-induced metabolites of arachidonic acid.. In addition to eNOS-derived NO production, the governance of H-ras expression by APE1/ref-1 could also contribute to some of the ...

All the water supplied in America is potable. Raw water undergoes treatment according to its source: it can be clorination to kill bacteria, sedimentation (with addition of alum) to reduce turbidity, filtration to separate particles. Estrogen compounds average size is in the order of 3 ?m. Only ultrafiltration or reverse osmosis can filter out parfticles of such a small size and those are very expensive processes not normally used in urban water supply plants. Ultrapure water - without virus and hormones - is used in pharma and vaccine production, but not for drinking. Regarding hormones in the environment, there are everywhere. The most commonly known sources are hormones supplied to domestic animals such a milch cows to increase milk production, pork and so on. Then treated urban wastewater is disposed in natural water bodies which all have high levels of estradiol like compounds (from female anticonceptives) and those are absorbed by the fish and by the animals feeding on fish and so on. Then ...

We investigated whether an EPA-enriched high-fat meal could improve postprandial vascular function. Seventeen healthy men ingested 2 test meals (51 g fat), 1 wk apart, in random order: 5 g EPA plus high-oleic sunflower oil (HOS) vs. HOS only. A second high-fat meal (44 g fat), the same on both study days, was provided 4 h later. Blood pressure and arterial function were measured using digital volume pulse (DVP) to derive a stiffness index (DVP-SI) and reflection index in fasting subjects at 3 and 6 h following the test meal. Blood samples were taken following the test meal for plasma 8-isoprostane F2alpha, nitric oxide (NO) metabolites (NOx), glucose, insulin, triacylglycerol, and fatty acid analysis ...

In this study we report that 8-iso-PGF2α, a major and stable product of lipid peroxidation, was potent in causing cell death of cultured brain microvascular endothelial cells as well as of ex vivo preparation of microvessels but not of human umbilical vein endothelium, smooth muscle, or astroglia; another related isoprostane, 8-iso-PGE2, did not trigger endothelial cell death. 8-Iso-PGF2α-induced cytotoxicity was TXA2 dependent and mostly of oncotic nature. Associated with these effects, intraventricular injection of 8-iso-PGF2α caused cystlike formations in the periventricular area.. The nature of cell death induced by 8-iso-PGF2α seems to be primarily oncotic rather than apoptotic necrosis (Figure 2). This inference is supported by the data that 8-iso-PGF2α caused ,8% of cell DNA fragmentation, a time-dependent increase in PI incorporation and LDH release, indicative of membrane disruption (Figure 2), and its effects, as well as those of its mediator TXA2, were unaltered by caspase ...

eNOS is constitutively expressed by endothelial cells and produces NO under basal conditions and in response to shear stress and agonists such as acetylcholine and bradykinin.25 Using eNOS-deficient mice, we investigated the role of eNOS in the development of fatty streaks in a dietary model. The results demonstrated that the absence of eNOS decreased fatty-streak formation in the mice. Plasma lipid levels were unlikely to explain the reduction in lesion formation, because eNOS-deficient mice did not show significant differences from control mice in the lipid levels.. Plasma 8-isoprostane was somewhat higher in eNOS-deficient mice. Isoprostanes result from oxidative modification of arachidonic acid through a free radical-catalyzed mechanism.26 Formation of these compounds in vivo is considered to be a reliable marker for lipid peroxidation. The present finding suggests that eNOS-derived NO protects against lipid peroxidation, probably by reacting with free oxygen radicals. Obviously, this ...

Much of the healing properties of calendula are because of its high levels of carotenoids (vitamin A like compounds). Because these compounds are oil soluble, an infused oil is a good way to go. Calendula also contains oil soluble sterols that help plump the skin and keep it thick. An infused oil is easily made by filling a jar with dried calendula flowers and covering the flowers with a carrier oil of some type; almond oil, olive oil, etc. You can get more out of the flowers by macerating the mixture in a blender. Let this oil set (infuse) for two weeks or more shaking it periodically to help extract the properties from the flowers. When ready to use filter the oil through cheesecloth. You can use this oil directly in a balm or as part of the oil portion of a cream or lotion. This infused oil can help with skin regeneration, diaper rash, sunburn, bed sores and various inflammatory conditions. Do make sure that your calendula is dry and that the oil completely covers the plant material to ...

This study deals with the important phenomenon of oxidative stress induced by the chronic inflammatory state in rheumatic patients of various categories and stages of disease. It investigates the local (knee joint) and systemic inflammatory response index and oxidative injury by measuring a major PGF2α metabolite, 15-keto-dihydro-PGF2α, and F2-isoprostane in the synovial fluid and blood, respectively. To our knowledge this study is the first to report a high concentration of 8-iso-PGF2α and moderately high levels of 15-keto-dihydro-PGF2α in various types of rheumatic patients in both synovial fluid and peripheral blood collected from the same patients. These results indicate that both non-enzymatic free radical and enzymatic cyclo-oxygenase catalysed oxidation of arachidonic acid occur in various types of rheumatic diseases, such as RA, OA, ReA, and PsA. In addition to the findings of this study, we have seen high levels of F2-isoprostanes and PGF2α metabolite in both synovial fluid and ...
In this study, we showed that pioglitazone treatment prevented the development of hypertension and renal oxidative stress in a rat model of diet-induced obesity. Also, our results indicate that compared with vitamin E, a well-known antioxidant, pioglitazone is more efficient in reducing BP and increasing sodium excretion in obese versus lean rats, while being equally efficient in reducing urinary isoprostanes, kidney TBARS, and HNE adducts. One possible explanation for these different effects is the ability of pioglitazone, as opposed to vitamin E, to increase renal NOS expression and NOx excretion in treated OP rats. In addition, pioglitazone, but not vitamin E treatment, significantly reduced expression of p47phox and gp91phox in OP rats, suggesting that both a reduction in oxidative stress and an improvement in NO production/bioavailability are important in efficiently reducing BP in this model.. Thiazolidinediones are known to exert pleiotropic actions both in vivo and in vitro, some of ...

Anesthesia-associated mortality in horses is high when compared with other species, and may be related to development of hypoxemia during recumbency. To overcome this, high fractions of inspired oxygen (FiO2 ,95%) are used. However, this may lead to an increase in oxidative stress, free radical formation and tissue injury. Isoprostanes are a marker of oxidative stress and their quantification is a relatively new and reliable approach to evaluate oxidative stress status. The aims of this research were to evaluate changes in urine isoprostanes (IsoP), serum cardiac troponin I (cTnI), arterial blood parameters, and serum chemistry in horses anesthetized with FiO2 of 0.45, 0.75, or ,0.95, and also evaluate the quality of recover utilizing accelerometry. Twenty-four healthy adult horses were anesthetized with isoflurane for 120 minutes. Horses were randomly assigned to receive a FiO2 of 0.45 (n=8), or 0.75 (n=8), or ,0.95 (n=8) during anesthesia. Horses were anesthetized with a standard anesthesia ...
Lipid peroxidation products like malondialdehyde, 4-hydroxynonenal and F(2)-isoprostanes are widely used as markers of oxidative stress in vitro and in vivo. This study reports the results of a multi-laboratory validation study by COST Action B35 to assess inter-laboratory and intra-laboratory variation in the measurement of lipid peroxidation. Human plasma samples were exposed to UVA irradiation at different doses (0, 15 J, 20 J), encoded and shipped to 15 laboratories, where analyses of malondialdehyde, 4-hydroxynonenal and isoprostanes were conducted. The results demonstrate a low within-day-variation and a good correlation of results observed on two different days. However, high coefficients of variation were observed between the laboratories. Malondialdehyde determined by HPLC was found to be the most sensitive and reproducible lipid peroxidation product in plasma upon UVA treatment. It is concluded that measurement of malondialdehyde by HPLC has good analytical validity for inter-laboratory
Ferro D and colleagues from Sapienza University of Rome, Rome, Italy have come up with evidence [CIRCEP.111.968248 Published online before print February 23, 2012, doi: 10.1161/?CIRCEP.111.968248] that low serum vitamin E levels are associated with recurrence of atrial fibrillation (AF) after cardioversion. They hypothesise that vitamin E being an antioxidant, low vitamin E levels would correlate with oxidative stress which could have a role in recurrence of atrial fibrillation after cardioversion. The study involved one hundred and forty four patients who had undergone electrical cardioversion with biphasic shocks for non valvar persistent atrial fibrillation. During the three month follow up ninety four patients were in sinus rhythm while fifty had recurrence of atrial fibrillation. Serum vitamin E levels were lower in patients with recurrence of atrial fibrillation (p,0.001). Higher left atrial diameter, urinary isoprostanes, high sensitivity C reactive protein (hsCRP) and sNOX2-dp (soluble ...
It is established that peptidemimetic drugs, such as renin inhibitors, somatostatin analogs, and endothelin antagonists, are often efficiently excreted into the bile in an unchanged form, resulting in their low bioavailability (Bertrams et al., 1991a,b;Greenfield et al., 1989; Nakamura et al., 1996). However, the present study shows that both the CLtotal andCLbile, p differ between various endothelin antagonist. Compound A has a 4-fold lowerCLtotal than that of BQ-485 (Table 1).CLbile, p also differs between each compound, with an 8-fold difference between compound A and BQ-485 (Table 1). The inhibition constant of compound A for binding to ETA is in the nanomolar range, as in the case of BQ-485 and BQ-123 (22, 3.4, and 13 nM for BQ-123, BQ-485, and compound A, respectively) (Fukami et al., 1996; Itoh et al., 1993; Moreland et al., 1994). This means that it is possible to construct a small peptide, like compound A, that exhibits relatively lower hepatic extraction but still has potent antagonist ...

This study investigated whether or not there was an overall difference in the pulmonary oxidative marker, 8-isoprostane (PGF2-a), found in exhaled breath condensate, during pregnancy versus the non-pregnant state. The utility of this information was important secondary to the effect of maternal asthma on pregnancy and outcome, as it has been demonstrated in past studies that overall pregnant females with asthma have been shown to have an increased risk of exacerbation requiring medical intervention. The primary goal of this study sought to determine if there is a difference in PGF2a levels in the exhaled breath condensate of pregnant versus non-pregnant females. In order to achieve this goal a cross-sectional study was performed consisting of two groups and were compared to one another. A group of 16 healthy, non-pregnant females aged 18-35 years old was compared to a group of 6 healthy, pregnant females in their third trimester of pregnancy. Both groups exhaled breath condensate was collected and 8
The white willow is a long used medicinal plant, rich in aspirin like compounds. It was formerly used in many of the ways we use aspirin today, to reduce inflammation and to reduce fever. But, in addition to these simple actions, white willow inspires the body to heal itself.
Antifreeze proteins (AFPs) are often called ice-binding proteins and are thought to bind to ice or ice-like crystals exclusively to control their growth. Here molecular recognition repertoire of AFPs has been greatly expanded to other hydroxyl containing, non-ice like compounds. The inhibitory and habit-modifying effects of AFPs on the stable nuclei formation and on the crystals of nucleosides have been first demonstrated.
Oxidative stress has been linked to many obesity-related conditions among children including cardiovascular disease, diabetes mellitus and hypertension. Exposure to environmental chemicals such as phthalates, ubiquitously found in humans, may also generate reactive oxygen species and subsequent oxidative stress. We examined longitudinal changes of 8-isoprostane urinary concentrations, a validated biomarker of oxidative stress, and associations with maternal prenatal urinary concentrations of phthalate metabolites for 258 children at 5, 9 and 14 years of age participating in a birth cohort residing in an agricultural area in California. Phthalates are endocrine disruptors, and in utero exposure has been also linked to altered lipid metabolism, as well as adverse birth and neurodevelopmental outcomes. We found that median creatinine-corrected 8-isoprostane concentrations remained constant across all age groups and did not differ by sex. Total cholesterol, systolic and diastolic blood pressure were ...
In the present study, we examined whether particular urinary oxylipins (isoprostanes (IsoPs), leukotrienes (LTs), prostaglandins (PGs), and thromboxanes (TXs)) in 16 elite triathletes could alter during 145 days of training. Within this time span, 45 days were dedicated to examining the effects of the intake of a b
This latest edition has been comprehensively rewritten and updated (over 80% of the text is new), whilst maintaining the clarity of its predecessor. There is expanded coverage of isoprostanes and related compounds, mechanisms of oxidative damage to DNA and proteins (and the repair of such damage), the free radical theory of ageing and the roles played by reactive species in signal transduction, cell death, human reproduction, and other important biological events. Greater emphasis has also been placed on the methods available to measure reactive species and oxidative damage (and their potential pitfalls), as well as the importance of antioxidants in the human diet. This book is recommended as a comprehensive introduction to the field for students, clinicians and researchers, and an invaluable companion to all those interested in the role of free radicals in the life and medical sciences.--BOOK JACKET.
R. John Davenport http://sageke.sciencemag.org/cgi/content/abstract/sageke;2002/36/nw126 Key Words: NCX-4016 hypercholesterolemia malondialdehyde isoprostanes. Abstract: Modern twists on old favorites sometimes make them better. According to a new study, a redesigned aspirin molecule that delivers nitric oxide battles atherosclerosis, at least in rodents. The modified molecule keeps fatty plaques from accumulating in blood vessels and choking circulation. The study suggests that the compound, already being tested as a gut-sparing painkiller, might double as an effective plaque-buster.. The signaling molecule NO relaxes blood vessels and soothes inflammation. Previous work suggested that NO also protects blood vessels from fatty buildup: Atherosclerosis-prone mice accumulate more plaques when treated with compounds that block NO production. But whether adding NO could prevent atherosclerosis was unclear.. To further explore NOs plaque-preventing prowess, Napoli and colleagues used an old standby ...
