Characterization of cathepsins in cartilage | Biochemical Journal
The presence of a cathepsin B-like enzyme in rabbit ear cartilage was established by the use of the synthetic substrates benzoyl-l-arginine amide and benzoyl-dl-arginine 2-naphthylamide. This was facilitated by using a technique that permits the incubation of a fixed weight of thin (18μ) cartilage sections with an appropriate exogenous substrate. The enzymic properties of cathepsin B in cartilage have been compared with an endogenous enzyme that liberates chondromucopeptide by degrading the cartilage matrix autocatalytically at pH5. Besides being maximally active at pH4·7, these cartilage enzymes are enhanced in activity by cysteine and inhibited by arginine analogues, iodoacetamide, chloroquine and mercuric chloride. They are not inhibited by EDTA, di-isopropyl phosphorofluoridate and diethyl p-nitrophenyl phosphate. An endopeptidase (designated RSIP, for root-starvation-induced protease) was purified to homogeneity from glucose-starved maize roots. The molecular mass of the enzyme was 59 kDa by SDS/PAGE under reducing conditions and 62 kDa by gel filtration on a Sephacryl S-200 column. The isoelectric point of RSIP was 4.55. The purified enzyme was stable, with no autoproteolytic activity. The enzyme activity was strongly inhibited by proteinaceous trypsin inhibitors, di-isopropylfluorophosphate, 3,4-dichloroisocoumarin and PMSF, suggesting that the enzyme is a serine protease. The maximum proteolytic activity against different protein substrates occurred at pH 6.5. With the exception of succinyl-Leu-Leu-Val-Tyr-4-methylcoumarin, no hydrolysis was detected with synthetic tryptic, chymotryptic or peptidylglutamate substrates. CD14, expressed on the surface of monocytes as a phospholipid-linked protein, is a receptor for serum LPS binding protein/LPS complex. It was specifically down-modulated after stimulation of monocytes by physiologic activating/differentiating agents such as bacterial LPS and IFN-gamma, by the pharmacologic agents PMA and calcium ionophore A23187, and by anti-CD14 antibodies. The down-modulation was almost totally blocked at 4 degrees C or at pH 4.5 and markedly inhibited by the protease inhibitors diisopropylfluorophosphate and PMSF. A soluble labeled CD14 was isolated from culture supernatant of surface iodinated monocytes after their activation, indicating that CD14 is shed from the cell surface rather than internalized. Tritiated acetylcholine was used to measure binding sites with characteristics of nicotinic cholinergic receptors in rat brain. Regulation of the binding sites in vivo was examined by administering two drugs that stimulate nicotinic receptors directly or indirectly. After 10 days of exposure to the cholinesterase inhibitor diisopropyl fluorophosphate, binding of tritiated acetylcholine in the cerebral cortex was decreased. However, after repeated administration of nicotine for 10 days, binding of tritiated acetylcholine in the cortex was increased. The formation of 1-alkylglycerol from 1-alkyl-sn-glycero-3-phosphoethanolamine in different cell fractions of rat brain is reported. The substrates used were labelled either with 14C or 3H in the alkyl residue or with 14C in the alkyl and 3H in the ethanolamine residue. The examination of the lipid- and water-soluble cleavage products showed that both ethanolamine and phosphoethanolamine are liberated from the substrate in the microsomal fraction of 14-day-old rat brain. The latter product is rapidly hydrolyzed. In comparison with other cell fractions, the microsomes contained the highest enzyme activities, which exhibited a pH optimum of 7.1--7.5. SH-group reagents are inhibitors, whereas diisopropylfluorophosphate has no effect. As the animals age, these enzyme activities decrease in brain homogenates. ...

Primary cultures of bovine adrenomedullary chromaffin cells were treated with the phosphorous acid ester, triphenyl phosphite (TPP), and the morphological changes assessed by transmission electron and scanning microscopy. Parallel studies were carried out with the cholinergic compound 0,0-diethyl-4 nitrophenyl phosphate (paraoxon) and the delayed neurotoxicant 0,0-diisopropylphosphorofluoridate (DFP). Transmission and scanning electron microscopy revealed that treatment with both of these organophosphorous compounds did not produce the ultrastructural effects that were seen with TPP. Treatment with 100 micromoles TPP for 4 or 24 hours caused a marked inhibition (90% relative to controls) of adenosine incorporation. Neither 100 micromoles paraoxon or 100 micromoles DFP had an inhibitory effect on incorporation. Epilepsy is the 3rd most common neurological disease, and it affects both humans and animals. Epileptogenesis is how an individual develops epilepsy and is typically caused by neuronal hyperexcitability. Antiepileptic drugs target ion channels to reduce neuronal hyperexcitability, but more than one-third of epileptic patients do not respond to treatments. There are around 30 antiepileptic drugs and about 45 other drugs that have failed clinical trials. The goal of this study was to find alternative pathways to aid in drug development. Neuroinflammation was the alternative target pathway studied. A rat model was used, and epileptogenesis was induced with the chemical nerve agent diisopropylfluorophosphate. Once epilepsy was established, one group was treated with the drug saracatinib and the other a control. Neutrophils, or polymorphonuclear leukocytes, comprise a crucial component of innate immunity, controlling bacterial and fungal infection through a combination of both oxidative and nonoxidative mechanisms. Indeed, neutrophils are believed to play an important role in controlling infection caused by the major human pathogen Streptococcus pneumoniae. However, the method by which neutrophils kill the pneumococcus as well as other Gram-positive bacteria, is not fully understood. We investigated human neutrophil killing of the pneumococcus in a complement-dependent opsonophagocytic assay. In contrast to other Gram-positive organisms, inhibition of the NADPH oxidase did not affect killing of S. pneumoniae. Supernatant from degranulated neutrophils killed the pneumococcus, suggesting a role for granular products.
