TY - JOUR. T1 - The akt-endothelial nitric oxide synthase pathway in lipopolysaccharide preconditioning-induced hypoxic-ischemic tolerance in the neonatal rat brain. AU - Lin, Hsiang Yin. AU - Wu, Chao Liang. AU - Huang, Chao Ching. PY - 2010/7. Y1 - 2010/7. N2 - Background and purpose: Low-dose lipopolysaccharide (LPS) preconditioning provides neonatal rats long-term neuroprotection against hypoxic ischemia (HI). Upregulating endothelial nitric oxide synthase (eNOS) protects against cerebral ischemia; however, whether eNOS is required for LPS preconditioning-induced protection in neonatal rats is unknown. We hypothesized that Akt activation, which upregulates eNOS in neurons and endothelial cells, is required for LPS preconditioning-induced tolerance against HI in the neonatal brain. Methods: Six-day-old rat pups were intraperitoneally injected with LPS (0.05 mg/kg) or normal saline 24 hours before HI. Immunoblotting and immunohistochemistry were used to determine the phospho-Akt (pAkt Ser473), ...
Evidence from our group revealed that the benefits of preconditioning ischemia extend beyond infarct size reduction via up-regulation of cardioprotective signaling. We found that brief antecedent ischemia: i) attenuates molecular indices of platelet activation-aggregation (i.e., P-selectin expression, fibrinogen binding); and thus ii) improves arterial patency in models mimicking unstable angina. However, two issues limit potential translation of this endogenous anti-platelet effect: the conditioning stimulus was applied directly to the heart, as a pretreatment. Our current aim was to establish whether brief ischemia applied at a remote site (skeletal muscle), after the onset of recurrent coronary thrombosis, would improve subsequent coronary patency.. Methods: Recurrent platelet-mediated coronary thrombosis was initiated in anesthetized dogs by application of a stenosis at a site of vessel injury. At 1 h after injury + stenosis, dogs received: i) remote ischemic conditioning (RIC: 4 5-min ...
The impact of remote limb ischemic conditioning on post-stroke cognitive impairment was evaluated with 104 first-time patients of noncardiac ischemic stroke. During the acute phase the patients were randomized into control and remote limb ischemic conditioning groups. Both groups received standard treatment, while the remote limb ischemic conditioning group received additional remote limb ischemic conditioning treatment for 6 months. All participants underwent neuropsychological evaluation, transcranial Doppler detection, P300 event-related potential and brachial-ankle pulse wave velocity measurements, and determination of serum intercellular adhesion molecule-1 and endothelin-1 levels both at admission and 6 months poststroke. The number of cases with poststroke cognitive impairment in each group was evaluated 6 months poststroke. No statistically significant difference was found in demographic data or baseline detection indices at admission between the two groups. However, at 6 months ...

Intermittent cycles of remote ischemic preconditioning augment diabetic foot ulcer healing.. Can creating a transient ischemic event in an extremity lead to wound healing? Early data from colleagues in Israel (see link above) suggest that this may be the case. In a randomized, sham-controlled study, people receiving RIC had a significantly greater likelihood of healing, more rapid healing, and greater overall granulation tissue at the end of study.. The findings above follow well with other works that have supported findings in multiple organ systems. Heres a strong review on the subject and below we share some helpful diagrams.. ...
The present study provides evidence that low-dose LPS preconditioning in rat pups confers neuroprotection and vasculoprotection against HI injury by activating both Akt and eNOS in neurons and endothelial cells. We found that low-dose LPS increased eNOS, but not iNOS or nNOS, expression. The LPS-induced eNOS expression was localized primarily in neurons and endothelial cells. Inhibiting eNOS expression by N(ω)-nitro-l-arginine or antisense ODN showed that eNOS was required for LPS-preconditioning-induced HI tolerance. We also found that Akt activation was necessary to upregulate eNOS in neurons and endothelial cells as well as to provide protection against HI in LPS preconditioning. Furthermore, we showed that pharmacologically and genetically upregulating eNOS protected rat pups against HI. Taken together, these findings show that upregulating eNOS by activating Akt in the neonatal brain is critical for low-dose LPS preconditioning-induced survival.. The 3 isoforms of NOS that are distributed ...

N2 - Although ischemic preconditioning induces bioenergetic tolerance and thereby remodels energy metabolism that is crucial for postischemic recovery of the heart, the molecular components associated with preservation of cellular energy production, transfer, and utilization are not fully understood. Here myocardial bioenergetic dynamics were assessed by 18O-assisted 31P-NMR spectroscopy in control or preconditioned hearts from wild-type (WT) or Kir6.2-knockout (Kir6.2-KO) mice that lack metabolism-sensing sarcolemmal ATP-sensitive K+ (KATP) channels. In WT vs. Kir6.2-KO hearts, preconditioning induced a significantly higher total ATP turnover (232 ± 20 vs. ...

ASN 2016 High Impact Clinical Trial. Kristin Veighey, Jennifer Nicholas, Tim Clayton, Raymond Macallister. Using 4 cycles of ischemia/reperfusion in the preoperative living donors, remote ischemic preconditioning (RIPC) was evaluated regarding its impact on post-tranplant outcomes. The primary endpoint was iohexal GFR at 12 months and secondary endpoints were change in eGFR, graft loss or death. There was no difference in the primary outcome. At 5 years post-transplantation, in the early RIPC group vs control, there was an overall improvement in eGFR by 4.8 ml/min (p = 0.003). There was no impact in the late RIPC or dual RIPC groups.. ...

Objectives: Heme-heme oxygenase (HO) system is one of the first line defenses against alterations in cellular redox, and, has been show to protect against vascular endothelial dysfunction in chronic diseased states, such as obesity and diabetes. We tested the hypothesis that pre-conditioning with L4F attenuates ischemia induced cardiac dysfunction in diabetic mice.. Methods: db/db mice were treated with either L-4F (an apo-lipoprotein mimetic inducer of HO) or vehicle for 8 weeks. Glucose, insulin, adiponectin, and pro-inflammatory cytokines (IL-1beta, TNF-alpha, MCP-1) were measured in plasma so as to confirm a diabetic, inflammatory state. Trans-thoracic echocardiography was performed at the completion of the experiment after which, isolated hearts were subjected to ischemia/reperfusion (IR) while mounted on a Langendorffs preparation. Isolated hearts were then analyzed for HO-1, pAMPK, peNOS, iNOS, adiponectin and superoxide levels.. Results: As shown before, db/db mice developed ...

TY - THES. T1 - Systemic response to local ischaemia: the evolving concept of remote ischaemic preconditioning. AU - Saxena, Pankaj. PY - 2009. Y1 - 2009. N2 - [Truncated abstract] Life on our planet is possible because of oxygen. All mammals, including humans, evolved with an innate mechanism by which organs and cells may protect themselves against lack or excess of oxygen to some extent. This lack and excess of oxygen that occurs during interruption and restoration of blood supply to an organ is referred to as ischaemia-reperfusion (IR). IR is a ubiquitous phenomenon that accounts for a number of pathophysiological conditions. A prolonged period of ischaemia followed by reperfusion causes IR injury and leads to significant damage to the affected organ. The activation of a systemic inflammatory response is associated with the most detrimental effects of IR injury. It has been noticed that brief episodes of intermittent ischaemia applied locally render significant protection against subsequent ...
Ischemic tolerance can protect the brain against cerebral ischemia and neurodegenerative diseases. However, several studies demonstrate ischemic tolerance by various methods, the exact mechanisms of ischemic tolerance has not been clearly understood. In this study, we first studied brain ischemia-related mechanisms and then evaluated the outcomes of mitochondrial pathophysiology of ischemic tolerance in focal and full stroke animal models. In this study, mitochondrial and peroxisomal reactions are considered critical in ischemic tolerance. In rats and Syrian mice, the middle cerebral artery occlusion (MCAO) model was employed to represent cerebrovascular stroke. Ischemic tolerance exhibits different types of adaptation responses associated with a number of sub-cellular changes. Changes in the cellular non-genomic pathways are usually short and reversible; while, the consequences of genes expression are a long-term process and can lead to permanent alternations in the genes expression pattern. ...

Ischemic tolerance is an endogenous neuroprotective phenomenon induced by sublethal ischemia. Ischemic preconditioning (IPC), the first discovered form of ischemic tolerance, is widely seen in many species and in various organs including the brain and the spinal cord. Ischemic tolerance of the spinal cord is less familiar among neurosurgeons, although it has been reported from the viewpoint of preventing ischemic spinal cord injury during aortic surgery. It is important for neurosurgeons to have opportunities to see patients with spinal cord ischemia, and to understand ischemic tolerance of the spinal cord as well as the brain. IPC has a strong neuroprotective effect in animal models of ischemia; however, clinical application of IPC for ischemic brain and spinal diseases is difficult because they cannot be predicted. In addition, one drawback of preconditioning stimuli is that they are also capable of producing injury with only minor changes to their intensity or duration. Numerous methods to ...

TY - JOUR. T1 - CREB activation in the rapid, intermediate, and delayed ischemic preconditioning against hypoxic-ischemia in neonatal rat. AU - Lin, Wan Ying. AU - Chang, Ying Chao. AU - Lee, Hsueh Te. AU - Huang, Chao Ching. PY - 2009/2. Y1 - 2009/2. N2 - Ischemic preconditioning (IP) is a defense program in which exposure to sublethal ischemia followed by a period of reperfusion results in subsequent resistance to severe ischemic insults. Very few in vivo IP models have been established for neonatal brain. We examined whether rapid, intermediate, and delayed IP against hypoxic-ischemia (HI) could be induced in neonatal brain, and if so, whether the IP involved phosphorylation of cAMP response element-binding protein (pCREB) after HI. Postnatal day 7 rat pups were subjected to HI at 2 h (2-h IP), 6 h (6-h IP), or 22 h (22-h IP) after IP. We found all three IP groups had significantly reduced neuronal damage and TUNEL-(+) cells 24 h post-HI than no-IP group. Compared with control, the no-IP ...
