PC4 and SFRS1 interacting protein 1, also known as lens epithelium-derived growth factor (LEDGF/p75), dense fine speckles 70kD protein (DFS 70) or transcriptional coactivator p75/p52, is a protein that in humans is encoded by the PSIP1 gene. PSIP1 has not been clearly linked to a specific cellular mechanism. The term LEDGF/p75 (Lens epithelium-derived growth factor) has entered common usage based on the initial characterization of PSIP1, however this is a misnomer, as the protein is present in most tissues and has no direct role in the development of lens epithelium. LEDGF/p75, a transcription coactivator, gained prominence as a host factor that assists HIV integration and is probably the only integrase interactor whose knock-down severely affects the HIV integration levels. The interaction between HIV integrase and human LEDGF/p75 is a promising target for anti-HIV drug discovery. LEDGF/p75 recruits MLL complexes to HOX genes to regulate their expression. LEDGF/p52 is shown to recruit splicing ...
TY - JOUR. T1 - Effects of miR-335-5p in modulating osteogenic differentiation by specifically downregulating Wnt antagonist DKK1. AU - Zhang, Jin. AU - Tu, Qisheng. AU - Bonewald, Lynda F.. AU - He, Xi. AU - Stein, Gary. AU - Lian, Jane. AU - Chen, Jake. PY - 2011/8/1. Y1 - 2011/8/1. N2 - Dickkopf-related protein 1 (DKK1) is essential to maintain skeletal homeostasis as an inhibitor of Wnt signaling and osteogenic differentiation. The purpose of this study was to investigate the molecular mechanisms underlying the developmental stage-specific regulation of the DKK1 protein level. We performed a series of studies including luciferase reporter assays, micro-RNA microarray, site-specific mutations, and gain- and loss-of-function analyses. We found that the DKK1 protein level was regulated via DKK1 3 UTR by miRNA control, which was restricted to osteoblast-lineage cells. As a result of decreased DKK1 protein level by miR-335-5p, Wnt signaling was enhanced, as indicated by elevated GSK-3β ...
Granulin is a protein that in humans is encoded by the GRN gene. Granulins are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. The 88 kDa precursor protein, progranulin, is also called proepithelin and prostate cancer (PC) cell-derived growth factor. Cleavage of the signal peptide produces mature granulin which can be further cleaved into a variety of active, 6 kDa peptides. These smaller cleavage products are named granulin A, granulin B, granulin C, etc. Epithelins 1 and 2 are synonymous with granulins A and B, respectively. Both the peptides and intact granulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. Granulin family members are important in normal development, wound healing, and tumorigenesis. The human liver fluke (Opisthorchis viverrini) contributes to ...
Background The Wnt signalling pathway is an important regulator of adult tissue maintenance and homeostasis. Disorders in Wnt signaling cause human degenerative diseases as well as cancer. Dickkopf-1 (DKK-1) is negative regulator protein of the Wnt pathway and is associated with a variety of organic diseases. Under the hypothesis that DKK-1 as a regulator protein is involved in ventricular remodelling after myocardial infarction we aimed to investigate the prognostic value of serum measurement of DKK-1 in patients with acute coronary syndromes.. Methods and results From April 2003 until April 2005 1136 consecutive patients (age 64±12 years; 347 females) with an ACS within the last 48 hours, who were referred for early invasive diagnositc were included. Follow up data were available for 1128 (99.3%) patients. Serum samples on admission were available for 1092 (97%) patients and from 820 (72%) patients a second sample the day following admission. Values are expressed as median. Values of DKK-1 on ...

Primary cell cultures: human fetal material, oligodendrocyte isolation, enrichment, and maturation. Human fetal spinal cord tissue was obtained from the Einstein Human Fetal Tissue Repository (New York, NY) as approved by the Institutional Review Board. Tissues from abortuses of normal women were collected after elective pregnancy termination. Informed consent was obtained from all tissue donors. The ages of the abortuses were determined by multiple parameters, including the date of the last menstrual period by history, uterine size by bimanual and abdominal examination, ultrasonography by using predominantly the maximum biparietal diameter, and, postabortally, by measurement of fetal foot length (Streeter, 1920; Hern, 1984). After retrieval of tissue the spinal cords were stored in sterile medium with antibiotics on ice and were obtained from the neuropathologist within the hour. For cultures the oligodendrocytes were obtained from 21-23 gestational weeks (gw) human fetal spinal cords. The ...
Granulin-epithelin precursor (GEP) is a secretory growth factor, which has been demonstrated to control cancer growth, invasion, drug resistance and immune escape. Our previous studies and others also demonstrated its potential in targeted therapy. Comprehensive characterization of GEP partner on cancer cells are warranted. We have previously shown that GEP interacted with heparan sulfate on the surface of liver cancer cells and the interaction is crucial for GEP-mediated signaling transduction. This study aims to characterize GEP protein partner at the cell membrane with the co-immunoprecipitation and mass spectrometry approach. The membrane fraction from liver cancer model Hep3B was used for capturing binding partner with the specific monoclonal antibody against GEP. The precipitated proteins were analyzed by mass spectrometry. After identifying the GEP binding partner, this specific interaction was validated in additional liver cancer cell line HepG2 by co-immunoprecipitation using GRP78 and GEP

Granulin-epithelin precursor (GEP), a secretory growth factor, demonstrated overexpression in various human cancers, however, mechanism remain elusive. Primary liver cancer, hepatocellular carcinoma (HCC), ranks the second in cancer-related death globally. GEP controlled growth, invasion, metastasis and chemo-resistance in liver cancer. Noted that GEP gene locates at 17q21 and the region has been frequently reported to be amplified in subset of HCC. The study aims to investigate if copy number gain would associate with GEP overexpression. Quantitative Microsatellite Analysis (QuMA) was used to quantify the GEP DNA copy number, and fluorescent in situ hybridization (FISH) was performed to consolidate the amplification status. GEP gene copy number, mRNA expression level and clinico-pathological features were analyzed. GEP DNA copy number determined by QuMA corroborated well with the FISH data, and the gene copy number correlated with the expression levels (n = 60, r = 0.331, P = 0.010). Gain of GEP copy

In the present study, we have quantified levels of myotrophin concentration in the hearts of SHR and normal rats of different age groups. Myotrophin was quantified in normal and hypertrophied rat hearts for various age groups by using the specific antibody raised against MAP. The antibody was characterized by solid-phase radioimmunoassays and Western blot analysis. To define the specificity of the polyclonal antibody raised against myotrophin, the affinity constant was calculated from the Scatchard plot. The accuracy of the quantification depends on the specificity of the antibody raised against MAP and its affinity toward the antigen (myotrophin). The specificity of the antibody has been established by three methods. First, we determined the affinity constant of the myotrophin antibody from the Scatchard plot as described in Materials and Methods. The Scatchard plot was constructed by plotting the ratio of bound to free myotrophin against the concentration of bound myotrophin. The ...
A population of monocytes, known as Ly6C(lo) monocytes, patrol blood vessels by crawling along the vascular endothelium. Here we show that endothelial cells control their origin through Notch signalling. Using combinations of conditional genetic deletion strategies and cell-fate tracking experiments we show that Notch2 regulates conversion of Ly6C(hi) monocytes into Ly6C(lo) monocytes in vivo and in vitro, thereby regulating monocyte cell fate under steady-state conditions. This process is controlled by Notch ligand delta-like 1 (Dll1) expressed by a population of endothelial cells that constitute distinct vascular niches in the bone marrow and spleen in vivo, while culture on recombinant DLL1 induces monocyte conversion in vitro. Thus, blood vessels regulate monocyte conversion, a form of committed myeloid cell fate regulation. ...

Primary malignant lymphoma of the prostate (PMLP) is prone to occur in the elderly, and it has no significant correlation with lactate dehydrogenase (LDH) and prostate specific antigen (PSA). Clinical symptoms and imaging data of PMLP remain unspecific, and its prognosis is poor. A previous result showed that collapsin response mediator protein 4 (CRMP4) promotor methylation can be used as a predictor for lymph node metastases in prostate biopsies. However, the relationship between CRMP4 promotor methylation and PMLP has not been studied. We investigated the clinicopathological features of PMLP and the significance of CRMP4 methylation in PMLP. The clinical data and diagnosis information of 10 patients with PMLP were retrospectively analyzed. The CRMP4 promotor methylation level in paraffin-embedded tissues of the 10 patients with PMLP were determined and then compared to limited prostate cancer (LPCa) and its negative lymph node tissue [LPCa-LN (−) (10 cases)] and also to metastatic prostate

In this report, we show that the Wnt antagonist Dkk-1 suppressed canonical Wnt signaling in bone marrow endosteal cells, suggesting a role for Dkk-1 in the regulation of the bone marrow stem cell niche. Intriguingly, Dkk-1 mobilized vasculogenic progenitor cells without concomitant release of inflammatory cells as compared to G-CSF. These effects are reminiscent of the recently described effect of RANKL.16 Indeed, Dkk-1 induced expression of the osteoclast differentiation factor RANKL and the bone-resorbing protease cathepsin K. Subsequently, the vasculogenic progenitor mobilizing effect of systemically administered Dkk-1 resulted in enhanced neovascularization in the Matrigel plug assay, because higher numbers of GFP+ transgenic bone marrow-derived cells were recruited to newly formed vessels. Therefore, Dkk-1 appears to be a potent regulator of vasculogenic progenitors.. The mechanisms by which Dkk-1 stimulates RANKL expression and mobilization of vasculogenic progenitors may include a direct ...

Hepatocellular carcinoma (HCC) is the fifth most common cancer in the world. However, little is known regarding the molecular mechanism of HCC development and progression and effective therapeutic methods. Recently, the granulin-epithelin precursor (GEP) was reported as a novel growth factor that can control HCC cell proliferation. Using the CAPSID program, we designed three small interfering RNAs (siRNAs) targeting the GEP gene (GEP-siRNA1, 2 and 3) and examined their tumor regression and suppression effects on cell proliferation. GEP-siRNA1 exhibited the strongest anti-proliferative effect among the GEP-siRNAs, in a time-dependent manner. To increase the biostability of the siRNA, we also constructed a short hairpin RNA (shRNA) using an H1/TO promoter with the same sequence of GEP-siRNA1 (GEP-shRNA). GEP-shRNA decreased the expression levels of GEP and tumor cell growth via cell cycle arrest at the G2/M stage and down-regulation of the cell proliferation proteins cyclin D1 and α-tubulin. ...

The molecular factors that regulate cardiac differentiation have been extensively studied, yet, relatively little is known about how cardiomyocytes acquire atrial versus ventricular characteristics. Embryonic stem (ES) cells, which have the potential to differentiate to a wide array of distinct cell types, including most types of cardiovascular cells, offer a pertinent in vitro model to work out the molecular mechanisms of atrial specification and differentiation. We discovered that the secreted antagonist of BMP signaling, Protein Related to Dan and Cerberus (PRDC, also called Gremlin2) leads to a surge in cardiomyocytic differentiation when applied to mouse ES-derived cardiac progenitor cells. This property is unique to PRDC among tested BMP antagonists. Lineage expansion is restricted to cardiomyocytes, with the differentiation of endodermal, blood, endothelial and neuronal cells being unaffected. Using molecular and electrophysiological analyses, we show that PRDC-induced cardiomyocytes ...
Preadipocyte factor-1 (Pref-1) is a transmembrane protein highly expressed in preadipocytes. Pref-1 expression is, however, completely abolished in adipocytes. The extracellular domain of Pref-1 undergoes two proteolytic cleavage events that generate 50 and 25 kDa soluble products. To understand the function of Pref-1, we generated transgenic mice that express the full ectodomain corresponding to the large cleavage product of Pref-1 fused to human immunoglobulin-γ constant region. Mice expressing the Pref-1/hFc transgene in adipose tissue, driven by the adipocyte fatty acid-binding protein (aP2, also known as aFABP) promoter, showed a substantial decrease in total fat pad weight. Moreover, adipose tissue from transgenic mice showed reduced expression of adipocyte markers and adipocyte-secreted factors, including leptin and adiponectin, whereas the preadipocyte marker Pref-1 was increased. Pref-1 transgenic mice with a substantial, but not complete, loss of adipose tissue exhibited ...
Connective tissue growth factor (CTGF/CCN2) is axiomatically necessary for proper skeletal development and function. We need not look further than the studies that have been done to date utilizing mice genetically engineered to lack CTGF production. These CTGF null or knockout (KO) mice fail to form a normal murine skeleton and instead yield one littered with bony dysmorphisms, including incompetent craniofacial development, kinked limb bones, and misshapen ribs that are not conducive to proper respiratory function. As a result, the global lack of CTGF is incompatible with postnatal life. A closer look at several sites demonstrated defects in physiologic processes necessary for bone formation - angiogenesis, chondrogenesis, and osteogenesis. Therefore, the dogma in the CCN protein field to date has been that systemic ablation of CTGF production in vivo results in global defects in bone development. We believe this dogma is an oversimplification of the role of CTGF on skeletal development. Our ...

Connective tissue growth factor (CTGF) is a secreted protein that is strongly induced in human and experimental heart failure. CTGF is said to be profibrotic; however, the precise function of CTGF is unclear. We generated transgenic mice and rats with cardiomyocyte-specific CTGF overexpression (CTGF-TG). To investigate CTGF as a fibrosis inducer, we performed morphological and gene expression analyses of CTGF-TG mice and rat hearts under basal conditions and after stimulation with angiotensin II (Ang II) or isoproterenol, respectively. Surprisingly, cardiac tissues of both models did not show increased fibrosis or enhanced gene expression of fibrotic markers. In contrast to controls, Ang II treated CTGF-TG mice displayed preserved cardiac function. However, CTGF-TG mice developed age-dependent cardiac dysfunction at the age of 7 months. CTGF related heart failure was associated with Akt and JNK activation, but not with the induction of natriuretic peptides. Furthermore, cardiomyocytes from ...

EGFR a receptor tyrosine kinase. This is a receptor for epidermal growth factor (EGF) and related growth factors including TGF-alpha, amphiregulin, betacellulin, heparin-binding EGF-like growth factor, GP30, and vaccinia virus growth factor. EGFR is involved in the control of cell growth and differentiation. It is a single-pass transmembrane tyrosine kinase. Ligand binding to this receptor results in receptor dimerization, autophosphorylation (in trans), activation of various downstream signaling molecules and lysosomal degradation. It can be phosphorylated and activated by Src. Activated EGFR binds the SH2 domain of phospholipase C-gamma (PLC-gamma), activating PLC-gamma-mediated downstream signaling. Phosphorylated EGFR binds Cbl, leading to its ubiquitination and degradation. Grb2 and SHC bind to phospho-EGFR and are involved in the activation of MAP kinase signaling pathways. Phosphorylation on Ser and Thr residues is thought to represent a mechanism for attenuation of EGFR kinase activity. ...

Plasma and urinary levels of CTGF were significantly elevated in nondiabetic proteinuric CKD patients. Antiproteinuric intervention was associated with a stepwise reduction in urinary CTGF in proportion to the reduction in proteinuria but did not affect the elevated plasma CTGF levels.. CTGF is strongly implicated in diabetic renal fibrosis and injury (4-12). The elevated levels of CTGF in plasma and urine in our patients suggest that CTGF may also play a role in the pathophysiology of nondiabetic CKD and its extrarenal complications, as a biomarker and/or as a pathogenic factor.. The source of the urinary CTGF is of interest. Because of their small size (≤38 kD), CTGF and the fragments thereof are predicted to be cleared from plasma by glomerular filtration (21,22). Consequently, glomerular filtration of elevated plasma CTGF may be one of the causes of the elevated urinary CTGF levels in our patients who had a relatively preserved renal function. Second, the elevated urinary CTGF in our ...

The cytokine mda-7/interleukin (IL)-24 has been suggested to function as a tumor suppressor in a number of solid tumor types and to promote cancer-specific cell death in a context-dependent manner. Li and colleagues reveal that mammary tumorigenesis in rats was suppressed by single administration of an adenovirus-expressing mda-7/IL-24. Mechanistically, mda-7/IL-24 induced the expression of growth arrest-specific gene 3 (GAS3/PMP22) to inhibit the attachment and proliferation of tumor cells in part by blocking the interaction of β1-integrin with fibronectin. Using primary human breast cancer specimens, they found a positive correlation between the expression of the IL-24 receptor (IL20RA) and GAS3. Moreover, IL20RA expression was correlated with metastasis-free survival in HER2+ patient samples. Thus, a novel GAS3-integrin/fibronectin inhibitory pathway has been identified downstream of mda-7/IL-24 that can be targeted for therapy.. ...
resistin-like molecule (RELM) hormone family; RELMs, secreted proteins with roles including insulin resistance and the activation of inflammatory processes, are also known as found in inflammatory zone (FIZZ), and include four members in mouse (RELM-alpha/FIZZ1/HIMF, RELM-beta/FIZZ2, Resistin/FIZZ3, and RELM-gamma/FIZZ4) and two members in human (resistin and RELM-beta). Little is yet known about the differences and similarities in function of the different isoforms. RELMs are potentially implicated in a wide range of physiological and pathological processes including obesity-associated diabetes, cardiovascular system function, cancer development and metastasis. There are significant differences between human and rodent RELMs with respect to gene and protein structure, differential gene regulation, different tissue distribution profiles, and insulin resistance induction. Resistin appears to convey insulin resistance in rodents, and to instigate inflammatory processes in humans. In the ...
Background: Gas6 is a product of growth arrest-specific gene 6 and is mostly expressed by the growth-arrested organs and is implicated in thrombosis, various inflammation, and age-related diseases. However, its role in chronic obstructive pulmonary disease has not been investigated yet. Methods: We developed lipopolysaccharide-induced mouse model of chronic lung inflammation to study its involvement. Therefore, male C57BL/6 mice (20-25 g) were grouped into control and LPS group; each group having 15 animals each. LPS group was nebulized with LPS (1 mg/ml) for 45 min, once a week for 8 weeks to induce chronic inflammatory conditions in the lungs by use of nebulizer. The mice model was validated by assessing infiltration of various immune cells (T-cells, B-cells, and neutrophils) into the lungs and pro-inflammatory and anti-inflammatory cytokine profile by cytokine bead array kit by flow cytometry. Lung deterioration was assessed by lung histology. The level of gas6 protein from the lung ...

This gene encodes a high-affinity dickkopf homolog 1 (DKK1) transmembrane receptor that functionally cooperates with DKK1 to block wingless (WNT)/beta-catenin signaling. The encoded protein is a component of a membrane complex that modulates canonical WNT signaling through lipoprotein receptor-related protein 6 (LRP6). It contains extracellular kringle, WSC, and CUB domains. Alternatively spliced transcript variants encoding distinct isoforms have been observed for this gene. [provided by RefSeq, Jul 2008 ...

Purpose: Progranulin is an evolutionarily conserved secreted growth factor well established for its role in embryogenesis, tumor development and wound repair. Although much is known about progranulin activity in non-neuronal tissues, the functions of progranulin within the central nervous system (CNS) are not well understood. In humans, mutations in the progranulin gene (GRN) cause neurodegeneration. In zebrafish, progranulin-a (Pgrn-a), syntenic to GRN, is expressed exclusively by peripheral macrophages and CNS microglia, and is strongly upregulated in retinal microglia after the selective death and regeneration of photoreceptors. There is a gap in our knowledge, however, regarding the functions of progranulin during developmental neurogenesis. Therefore, the purpose of this study was to determine how Pgrn-a regulates vertebrate nervous system development, using the zebrafish retina as a model.. Methods: The work presented here combined reverse genetics and quantitative morphometric approaches, ...

Supplementary MaterialsSupplementary Statistics. while treatment with particular inhibitors indicated that hypoxia upregulates HBEGF biosynthesis through anybody from the three analyzed MAPKs.2, 3 However, it had been unclear whether this MAPK pathway was functional or downstream of HBEGF shedding upstream. As a result, MMP2 was quantified in individual TB cells cultured at 2% O2 with particular inhibitors of ERK, jNK and p38. These inhibitors didnt impact the upregulation of MMP2 at low O2, recommending which the MAPKs function solely downstream of HBEGF signaling through the ERBB1/4 tyrosine kinases in individual TB cells, as indicated in Amount 9. Although both MMP2 and HBEGF post-transcriptionally are governed by O2, HBEGF upregulation by low O2 or CoCl2 was obstructed by reported that serum degrees of HSP70 are continuous throughout normal being pregnant, but upsurge in women with preeclampsia or preterm PRI-724 irreversible inhibition delivery significantly.49 Elevated circulating HSP70 ...
