We have studied the regulation of the extracellular chymoelastase protease (Pr1) of Metarhizium anisopliae, an enzyme involved in the penetration of insect cuticle by Metarhizium and other entomopathogenic fungi. We report here the isolation and characterization of a Pr1 cDNA clone with a full length insert. Pr1 is synthesized as a large precursor (40.3 kDa) containing a signal peptide and a propeptide and the mature protein is predicted to have a relative molecular mass of 28.6 kDa. The primary structure of Pr1 shares extensive homology (30-60%) with enzymes of the subtilisin subclass of the serine endopeptidases and the serine, histidine and aspartyl components of the active site in subtilisins are preserved. The genes coding for chymoelastase or slightly altered versions thereof, can be used to transform various organisms (i.e. fungi, viruses, plants, bacteria, etc.) such that the transformed organisms are capable of producing chymoelastase in recoverable quantities. The dermal bone sculpture of early, basal tetrapods of the Permo-Carboniferous is unlike the bone surface of any living vertebrate, and its function has long been obscure. Drawing from physiological studies of extant tetrapods, where dermal bone or other calcified tissues aid in regulating acid-base balance relating to hypercapnia (excess blood carbon dioxide) and/or lactate acidosis, we propose a similar function for these sculptured dermal bones in early tetrapods. Unlike the condition in modern reptiles, which experience hypercapnia when submerged in water, these animals would have experienced hypercapnia on land, owing to likely inefficient means of eliminating carbon dioxide. The different patterns of dermal bone sculpture in these tetrapods largely correlates with levels of terrestriality: sculpture is reduced or lost in stem amniotes that likely had the more efficient lung ventilation mode of costal aspiration, and in small-sized stem amphibians that would have been able to use the skin for gas

As reproductive success is mediated through body condition, this study aimed to assess the body condition and physiological indicators of stress of southern right whales on the South Africa breeding ground at present, and compare it to historical data and other populations. The southern right whales (Eubalaena australis) were intensely exploited in the Southern Hemisphere between the 18th and 20th centuries. Although the species has been protected internationally from commercial whaling since 1935, some populations were further depleted by illegal catches until the early 1970s. In this work, an empirical model of nearest neighbor hopping parameter (γ0) in tight binding (TB) model of single wall carbon nanotubes is proposed in order to calculate first and second optical transition energies of semiconducting SWCNTs. In this model of γ0, observations from various optical spectroscopic experiments are incorporated. We investigated traveling fronts of the oxidation of sulfite by iodate and by bromate. Isothermal density changes (Delta rho(c)) and enthalpies were determined, the latter being negative in both cases. The Delta rho(c) is negative in the iodate-sulfite system but positive in the bromate-sulfite system. Double-diffusive convection was expected in the bromate system and was observed. The effect of tube diameter and the orientation of the tube on the wave velocity and wave shape were studied, and a gravitational anisotropy was found in both systems. According to the Pojman-Epstein model, only simple convection should have occurred in the iodate-sulfite system. In fact, curved descending fronts with velocities greater than the ascending ones and fingering were observed. Several explanations were considered, including the possibility that the current model must be expanded to explicitly treat the effect of a moving interface on the stability of the fluid.
A predatory Nematoctonus species (N90) isolated from farmyard soil in Ontario produced gill-forming basidiocarps of Hohenbuehelia on potato dextrose agar. In the presence of nematodes, discharged basidiospores germinated to produce short aerial stalks with terminal adhesive knobs. Germinated basidiospores adhered to the cuticle of nematodes to initiate monokaryotic infection cycles. Multiple basidiospore infections on a single host resulted in dikaryotization in the host body. Crossing single basidiospore cultures demonstrated bipolar sexuality for this species of Hohenbuehelia. The implications of these findings are discussed.
