Inositol levels, maintained by the biosynthetic enzyme inositol-3-phosphate synthase (Ino1), are altered in a range of disorders, including bipolar disorder and Alzheimers disease. To date, most inositol studies have focused on the molecular and cellular effects of inositol depletion without considering Ino1 levels. Here we employ a simple eukaryote, Dictyostelium discoideum, to demonstrate distinct effects of loss of Ino1 and inositol depletion. We show that loss of Ino1 results in an inositol auxotrophy that can be rescued only partially by exogenous inositol. Removal of inositol supplementation from the ino1(-) mutant resulted in a rapid 56% reduction in inositol levels, triggering the induction of autophagy, reduced cytokinesis, and substrate adhesion. Inositol depletion also caused a dramatic generalized decrease in phosphoinositide levels that was rescued by inositol supplementation. One hypothesis for the effectiveness of current treatments of bipolar disorder is the reduction of cellular myo-inositol. Lithium and valproate are known to reduce cellular inositol levels, however these treatments often have notable side effects. Currently, there is a need to find a treatment for bipolar disorder that reduces the adverse side effects of these drugs. Previous studies, have found success in using omega-3 fatty acids for the treatment of bipolar affective disorder. The long-term purpose of this study is to determine what effect omega-3 fatty acids have on myo-inositol concentration. This study has worked toward this purpose by comparing the growth effects of 0.8mM DHA to 2.5mM VPA. We have found that similar to VPA, cell growth is reduced at 0.8mM.

Mullerian inhibiting substance (MIS) is a glycoprotein hormone produced in Sertoli cells of the fetal and postnatal testis, and granulosa cells of the pubertal ovary. We examined MIS expression in a nonhuman primate, the cynomolgus macaque monkey (Macaca fascicularis), to define an animal model for studying MIS gene regulation. Changes in testicular MIS mRNA with age were assessed by in situ hybridization of prepubertal to adult testes, Northern analysis of pubertal and adult specimens, and determination of serum MIS concentrations from infancy to adulthood. We found that MIS expression was highest in the youngest animals and decreased progressively with increasing age. Serum MIS concentrations correlated inversely with increasing age (r = -0.74), body weight (r = -0.79), and testicular volume (r = -0.73), but not with testosterone levels (r = -0.35). The mean MIS concentrations +/- SEM for the four developmental age groups were 270.6 +/- 23.8 (infants), 195.5 +/- 18.5 (juveniles), 102.7 +/- 28.4

A homogeneous population of [3H]Ins(1,4,5)P3-binding sites was present (KD 23.1 +/- 3.6 nM) at high density (Bmax. 11.9 +/- 1.8 pmol/mg of protein); whereas data obtained for [32P]Ins(1,3,4,5)P4 specific binding were best fitted to a two-site model, the high-affinity binding component (KD 2.6 +/- 0.7 nM) constituted 64.2 +/- 4.3% of the total population and was present at relatively low density (Bmax. 187 +/- 27 fmol/mg of protein). 3.
