Cellulases have diversity of industrial applications and their cost effective production using agroindustrial wastes by solid state fermentation poses an efficient method. Actinomycetes are considered highly valuable due to their secondary metabolites production and in this study, an attempt was made to optimize the use of ragi husk and refine the process of cellulase production by temperature mediated solid state fermentation. Actinomycetes were isolated from paper mill industry soil and cellulase producing |i|Streptomyces|/i| sp was selected for the experiments. Ragi husk was used as substrate for solid state fermentation of cellulase and varying incubation temperatures (20°C, 25°C, 30°C, 35°C and 40°C) was considered to determine its effect on enzyme activity after 6|sup|th|/sup|, 9|sup|th|/sup| and 12|sup|th|/sup| day of fermentation. The carboxymethyl cellualse (CMC-ase) activity was measured and the observations obtained were compared with the standard glucose curve to determine the amount of

Development of industrial yeast strains with high tolerance towards the inhibitors released during biomass pretreatment is critical for bioethanol production. Combining this trait with increased thermotolerance would result in a more efficient production via Simultaneous Saccharification and Fermentation (SSF) as well as reduced cooling costs. The aim of the present work was to develop a yeast strain combining these traits. Using a long-term adaptation strategy a stable Saccharomyces cerevisiae isolate (ISO12) was evolved from the industrial strain Ethanol Red (ER). ISO12, contrary to the parental strain, is capable of growing and fermenting the liquid fraction of non-detoxified spruce hydrolysate at 39°C with an ethanol yield of 0.38 g ethanol . g hexoses-1. In contrast with previous studies, the superior phenotype of ISO12 does not rely on higher reductase activities for furaldehyde inhibitor conversion, but rather on a higher thermotolerance. ISO12 shows a higher capacity to ferment hydrolysate at
Enhanced oil recovery (EOR) methods are used in oil industry to increase the ultimate recovery of crude oil. This normally involves application of an EOR method (sometimes called tertiary recovery method) to a specific underground oil bearing reservoir. Significant volumes of oil remaining in the depleted oil reservoirs (as much as 70%) could be produced by EOR methods. One of such method is microbial enhanced oil recovery (MEOR). Microbial enhanced oil recovery involves using microbes in the subsurface to recover additional hydrocarbon from reservoirs. The microbial enhanced oil recovery takes advantage of various microbial processes to increase hydrocarbon yield from residual oil by improving oil flow and flood water sweep in a reservoir during tertiary recovery. Microbial enhanced oil recovery strategies can be one or a mixture of processes which alters the lithology of the reservoir matrix, the viscosity of the oil in place, the chemical characteristics of the oil in place or the in situ ...

The Industrial Internet of Things (IIoT) is of strategic importance in the new era of industrial big data, creating a brand-new industrial ecosystem. Considering the unknown parameters in the IIoT-based industrial process control systems, this paper combines the artificial fish swarm algorithm (AFSA) and the particle filtering (PF) algorithm into the AFSA-PF algorithm based on the self-organizing state space (SOSS) model. The AFSA-PF algorithm not only can estimates the system state but also can make the sampling distribution of the unknown parameter to move the true parameter distribution. Ultimately, the true values of the unknown parameters are identified. In this way, the system model can gradually approximate the actual IIoT-based industrial process control system.

The Application of Solid State Fermentation for Obtaining Substances Useful in Healthcare. By Łukasz Wajda and Magdalena Januszek. In the current review we summarised the research involving solid state fermentation (SSF) for the production of compounds that could be used in healthcare (terpenoids, polyphenols, fibrinolytic enzymes, mycophenolic acid and others). We described several groups of obtained agents which hold various activity: antimicrobial, anti-inflammatory, immunosuppressive, anticoagulant and others (e.g. anticancer or anti-diabetic). It seems that especially terpenoids and polyphenols could be useful in that field, however, other substances such as enzymes and fatty acids play important role as well. We described main groups of microorganisms that are applied in SSF of those compounds, particularly Bacillus genus and fungi, and where possible provided information regarding genes involved in those processes. We also compared various approaches toward optimisation of SSF.. Part of ...
TY - JOUR. T1 - Combined discrete particle and continuum model predicting solid-state fermentation in a drum fermentor. AU - Schutyser, M.A.I.. AU - Briels, W.J.. AU - Boom, R.M.. AU - Rinzema, A.. PY - 2004. Y1 - 2004. N2 - The development of mathematical models facilitates industrial (large-scale) application of solid-state fermentation (SSF). In this study, a two-phase model of a drum fermentor is developed that consists of a discrete particle model (solid phase) and a continuum model (gas phase). The continuum model describes the distribution of air in the bed injected via an aeration pipe. The discrete particle model describes the solid phase. In previous work, mixing during SSF was predicted with the discrete particle model, although mixing simulations were not carried out in the current work. Heat and mass transfer between the two phases and biomass growth were implemented in the two phase model. Validation experiments were conducted in a 28-dm(3) drum fermentor. In this fermentor, ...

Recombinant DNA technology is yielding organisms with virtually a complete range of genetic stability, i.e. from organisms that appear to be entirely stable over the course of many fermentations to those that are so unstable that their half-life as novel species may be appreciably less than a conventional fermentation time. As such, a novel time-dependent mixed culture community is created, containing at a minimum the original recombinant microbe and a second strain, which may be denoted as a revertant in some partial or full sense. These two strains will differ in one or more important characteristics such as (desired) product formation rates, substrate utiliza- tion abilities, morphology, biomass growth rates and oxygen demand. In this paper, I consider batch and continuous cultivation, and I use some plausible biological rate forms to explore implications in fermentor behaviour and product formation occa- sioned by the existence of this particular form of mixed culture. ...

The current commercial practice of gas-based enhanced oil recovery processes involves either continuous gas injection (CGI) or Water-Alternating-Gas (WAG) injection. Over 60 commercial projects in West Texas and other parts of the world have amply demonstrated that these CGI and WAG processes have been technically successful and commercially profitable. However, the oil recoveries from the CGI and WAG processes fall in the range of 5-15% of the remaining oil. The Gas-Assisted Gravity Drainage (GAGD) process, invented and patented at LSU, has yielded oil recoveries in the range of 65 - 95% in laboratory experiments conducted at realistic reservoir conditions. The GAGD process involves utilizing several vertical wells for injection of CO2 in addition to drilling long horizontal wells for production. Injected CO2 accumulates at the top of the payzone due to gravity segregation and displaces oil, which drains to the horizontal producer. This maximizes the volumetric sweep efficiency. The gravity segregation

DuPont has created a key genetic manipulation that makes an oleaginous yeast (Mortierella alpina) produce large quantities of the enriched fatty acids EPA and DHA in pure form. The genes cause the yeast to produce long-chain omega-3 and omega-6 polyunsaturated fatty acids (PUFAs).. Currently, omega fatty acids are used as dietary supplements for humans and fish (the fish are then processed to extract the oil for humans and other fish). The technology was developed as part of a larger program at DuPont to discover renewable biological feedstocks. The initial application for the technology was as fish food, which is commonly created by capturing and chopping up wild salmon to feed to farmed salmon. This process is unsustainable in the long run. A yeast-fermentation approach should be more cost-effective and much more sustainable.. Unlike EPA and DHA processed from fish meal, the DuPont fermentation technology produces pure fatty acids without the further refining and processing that is necessary ...

Microalgae belong to a genetically and physiologically highly divergent group of eukaryotic microorganisms possessing unique biochemical pathways. Their biodiversity offers an as yet unexploited natural source of high-value bioactive molecules of potential use in pharmaceuticals, food/feed additives, cosmetics, flavors, and high-performance chemicals. Although the term microalga is typically used as a synonym for a photoautotrophic, eukaryotic, unicellular organism, it is possible to achieve efficient biomass production and biosynthesis of certain metabolites when the cells are grown with organic carbon and without light. Consequently, microalgae can be cultivated similarly to bacteria or yeasts, in multipurpose stirred-tank bioreactors made from steel. Opting for such well-controlled cultivation processes is justified when regulatory approval of a pharmaceutical or nutraceutical product is required, or where manufacturing productivity and robustness can be significantly enhanced. However, ...

Typically, the biotechnological basis for strain improvement to enhance recombinant protein production relies on the permanent implementation of desirable traits into the production strain to stimulate both cell growth and functional expression of the target gene during the cultivation. This chapter reviews the major technical issues associated with Escherichia coli strain engineering to enhance recombinant protein production and directs the reader to protocols appropriate for specific applications. Theoretically, strategies based on enhancing the limiting step can lead to an overall improvement in recombinant protein production. Stationary-phase genes encode proteins that may lead to a reduction in cellular and metabolic activity, which can negatively affect recombinant protein production, and as such, these genes are targets for strain improvement. The recently commercialized recombineering protocol from Gene Bridges, also based on λ Redmediated recombination, allows versatile chromosomal engineering
The renewability, biocompatibility, and mechanical properties of cellulose nanocrystals (CNCs) have made them an attractive material for numerous composite, biomedical, and rheological applications. However, for CNCs to shift from a laboratory curiosity to commercial applications, researchers must transition from CNCs extracted on the bench scale to material produced on an industrial scale. There are a number of companies currently producing kilogram to ton per day quantities of sulfuric acid-hydrolyzed CNCs as well as other nanocelluloses, as described herein. With the recent intensification of industrially produced CNCs and the variety of cellulose sources, hydrolysis methods, and purification procedures, the characterization of these materials becomes critical. This has further been justified by the past two decades of research that demonstrate that the CNC stability and behavior are highly dependent on the surface chemistry, surface charge density, and particle size. This work outlines key ...
Changes in pH, titrable acidity, protein, non-protein nitrogen, total soluble solids, protein fractions and in vitro protein digestibility were investigated during fermentation and/or supplementation of sorghum flour with whey protein. The pH of the fermenting material was decreased sharply with a concomitant increase in the titratable acidity. The total soluble solids increased with progressive fermentation time. The crude protein and. non-protein nitrogen were increased with fermentation time. The albumin plus globulin fraction was increased significantly (P = 0.05) during the first 8 h of fermentation. Other fractions contents were observed to fluctuate during fermentation time. Supplementation of the cultivar flour with whey protein greatly increased the protein content as well as albumin plus globulin fraction while other fractions were significantly decreased. The in vitro protein digestibility was significantly (P = 0.05) improved during fermentation time and even after supplementation. ...
Biotransformation of Steroids Using Different Microorganisms. By Arturo Cano-Flores, Javier Gómez and Rigoberto Ramos. The introduction of a hydroxyl group biohydroxylation in the steroid skeleton is an important step in the synthesis of new steroids used physiologically as hormones and active drugs. There are currently about 300 known steroid drugs whose production constitutes the second category within the pharmaceutical market after antibiotics. Several biotransformations at industrial scale have been applied in the production of steroid hormones and drugs, which have functionalized different types of raw materials by means of chemo-, regio-, and stereoselective reactions (hydroxylation, Baeyer-Villiger oxidation, oxidation reactions, reduction of group carbonyl, isomerization, and Michael additions, condensation reactions, among others). In Green Chemistry, biotransformations are an important chemical methodology toward more sustainable industrial processes.. Part of the book: Chemistry ...

Fermentation test in batch procedures for various biological processes (e.g. anaerobic digestion, anammox, dark fermentation, ethanol fermentation, etc.) are increasingly reconised as key experimental approaches for research studies and industrial applications. However, due to the nature of the microorganisms used in such studies, the batch tests often generate high variability between different laboratories, mainly due to a poor standardisation in experimental platform, experimental procedure, data interpretation, and presentation. As a consequence, much research is focused on standardisation of these tests; in this regard the automated batch fermentation system AMPTS II and Gas Endeavour are excellent experimental platforms as they minimise the human input and automatically measure, calculate, and present the data in a standardised way.. ...

Background: The bioethanol production system used in Brazil is based on the fermentation of sucrose from sugarcane feedstock by highly adapted strains of the yeast Saccharomyces cerevisiae. Bacterial contaminants present in the distillery environment often produce yeast-bacteria cellular co-aggregation particles that resemble yeast-yeast cell adhesion (flocculation). The formation of such particles is undesirable because it slows the fermentation kinetics and reduces the overall bioethanol yield. Results: In this study, we investigated the molecular physiology of one of the main S. cerevisiae strains used in Brazilian bioethanol production, PE-2, under two contrasting conditions: typical fermentation, when most yeast cells are in suspension, and co-aggregated fermentation. The transcriptional profile of PE-2 was assessed by RNA-seq during industrial scale fed-batch fermentation. Comparative analysis between the two conditions revealed transcriptional profiles that were differentiated primarily ...
Yellow water is a by-product of liquor in the solid state fermentation process, and contains a large amount of nutrients, such as acids, esters, alcohols and aldehydes produced by fermentation. The components in the yellow water reflect the fermentation information to a certain extent, so the fermentation process can be monitored by detecting the yellow water component online. A sensor array detection device is designed for detecting yellow water. In addition, chemical titration is used to obtain data such as acidity, reducing sugar and starch of yellow water. Principal component analysis and discriminant function analysis were performed on the data; and a multivariate linear regression was used to establish a prediction model for the data. The results showed that the prediction bias for acidity and alcohol was small, 0.39 and 0.43, respectively.

University of Helsinki, Faculty of Science, Department of Physical Sciences and The Finnish Forest Research Institute, Vantaa Research Unit and University of Oulu, Measurement and Sensor Laboratory. In recent years there has been growing interest in selecting suitable wood raw material to increase end product quality and to increase the efficiency of industrial processes. Genetic background and growing conditions are known to affect properties of growing trees, but only a few parameters reflecting wood quality, such as volume and density can be measured on an industrial scale. Therefore research on cellular level structures of trees grown in different conditions is needed to increase understanding of the growth process of trees leading to desired wood properties. In this work the cellular and cell wall structures of wood were studied. Parameters, such as the mean microfibril angle (MFA), the spiral grain angles, the fibre length, the tracheid cell wall thickness and the cross-sectional shape of ...

The design and construction of microbial cell factories is often focused on the engineering of intracellular enzymes and pathways, and the role of membrane-embedded proteins is often overlooked. It is nevertheless becoming increasingly clear that integral membrane proteins, in particular transporters, are critical for the performance and stability of microbial production strains [1-4]. Membrane-embedded transport proteins mediate the cellular uptake and extrusion of a wide diversity of solutes. As a consequence, adding or engineering a suitable uptake system into a production strain may greatly enhance substrate utilization and flux towards product [5-7]. Likewise, secretion systems increase flux, resolve toxicity-caused limitations, and facilitate product purification by secretion of the desired product to the extracellular environment [8-10].. Besides introducing and engineering known transport proteins for biotechnological applications, there is a critical need to identify novel transporters ...
article{8634556, abstract = {Thermal engineering of metal-organic frameworks for adsorption-based applications is very topical in view of their industrial potential, in particular, since heat management and thermal stability have been identified as important obstacles. Hence, a fundamental understanding of the structural and chemical features underpinning their intrinsic thermal properties is highly sought-after. Herein, we investigate the nanoscale behavior of a diverse set of frameworks using molecular simulation techniques and critically compare properties such as thermal conductivity, heat capacity, and thermal expansion with other classes of materials. Furthermore, we propose a hypothetical thermodynamic cycle to estimate the temperature rise associated with adsorption for the most important greenhouse and energy-related gases (CO2 and CH4). This macroscopic response on the heat of adsorption connects the intrinsic thermal properties with the adsorption properties and allows us to evaluate ...
In this article, an experiment was run in order to test nutritional and industrial potential of Moringa oleifera seed oil in different areas of Pakistan. It was found that in areas where the moringa tree was able to grow with a regular source of water, the yield of oil was much higher than in areas such as its native environment in mountainous areas. It was also observed that for many variables, the content of the oil was significantly higher than that of the M. oleifera oil native in Kenya. Overall, the moringa seeds had a much higher yield than conventional oil seeds, and also contained high amount of protein, fatty acids, and other variables that show potential for farm and industrial tasks, which makes M. oleifera seed oil a possibly successful crop for Pakistan, which would not only cut down costs of importing oils, but would also be a reasonable export ...

In some cases you may find it preferable to stop the fermentation process prior to it pulls up by itself. The most common factor for wanting to quit the fermentation procedure is that you have found the red wine currently has the exact quantity of sweet taste that you favor and you do not desire it to proceed any kind of even more.. By quiting the fermentation then, numerous winemakers believe that they can preserve the amount of sweetness that the white wine has actually currently generated. If you desire a truly pleasant a glass of wine, such as an after-dinner drink, this is absolutely understandable. The idea behind quiting the fermentation procedure is that if you enabled the red wine to continue fermenting it would certainly come to be much less pleasant as time went on. When the red wine became completely dry, the fermentation process would eventually stop by itself with no intervention from you.. wine on my time. As a result, there are a number of various methods that home winemakers ...

Oil production in fractured vuggy carbonate reservoir has unpredicted behaviors due to complex vug connectivity, variation of oil-water interface, and oil-water multiphase flow. Early water breakthrough and rapid increased of water cut as well as decreased of productivity are often encountered in this kind of reservoir. Experiments were conducted in vug-pore artificial cores with the size of 4.5 × 4.5 × 30 cm to study the influence of viscosity and different inclination angle on the oil recovery (OR). The influence of N2 and CO2 on the relative location of the water-oil/gas-water interface were compared. These results indicate that OR increases with the decrease of oil viscosity but not always with increase of inclination angle. Under CO2 conditions, however, influence of inclination angle on OR becomes more sensitive and gravity separation becomes more important factor at higher pressures.. Stainless steel tube with inner diameter (ID) of 1 mm was used to simulate the fracture. Time for CO2 ...

Due to climate change and plastic pollution, there is an increasing demand for bio-based materials with similar properties to those of common plastics yet biodegradable. In this respect, cellulose is a strong candidate that is already being refined on a large industrial scale, but the properties differ significantly from those of common plastics in terms of shapeability and water-resilience.. This thesis investigates how supramolecular interactions can be used to tailor the properties of cellulose-based materials by modifying cellulose surfaces or control the assembly of cellulose nanofibrils (CNFs). Most of the work is a fundamental study on interactions in aqueous environments, but some material concepts are presented and potential applications are discussed.. The first part deals with the modification of cellulose by the spontaneous adsorption of xyloglucan or polyelectrolytes. The results indicate that xyloglucan adsorbs to cellulose due to the increased entropy of water released from the ...
