Scientists at the John Innes Centre have found a way to produce industrial quantities of useful natural compounds efficiently, by growing them in tomatoes.. The compounds are phenylpropanoids like Resveratrol, the compound found in wine which has been reported to extend lifespan in animal studies, and Genistein, the compound found in soybean which has been suggested to play a role in prevention of steroid-hormone related cancers, particularly breast cancer. The diagnostic medical imaging technique of Positron Emission Tomography (PET) has been adapted at the University of Birmingham for use in imaging industrial processes. A particularly powerful technique, called Positron Emission Particle Tracking (PEPT) is used to study granular materials and fluid flows in a number of different applications. This thesis describes the most recent development of the detector systems which are used for PEPT studies at the Positron Imaging Centre. Primarily, the work has focussed upon developing a modular positron camera which can be arranged in custom geometries around the system under study. This camera is transportable and has been used to investigate a number of applications in situ. A new data acquisition system has been designed enabling the storage and analysis of the raw data produced by the detectors. Throughout the summer we worked on creating a prototype of the bioreactor. The process that was deemed most suitable, was a fed-batch system, where the reactors would be continually fed with more bacterial nutrients and fresh tailings in a continuous stir method. Product is removed at the same rate that biomass and nutrients are added. A continuous method would be more appropriate at an industrial scale (1000+ L tanks), where the bacteria will have enough time to convert toxins in such large tanks. Additionally, the process would have to occur within an enclosed system to ensure its containment.. Lastly, we needed a process to remove the hydrocarbons once they were produced. We decided to use a belt skimmer, similar to those used to help clean up oil spills. This method allows us to run the belt to pick up hydrocarbons without having to remove the entire solution of the batch. Proteolysis was shown to have a significant impact on the accumulation and the final yield of recombinant proteins. Much smaller intracellular concentrations of the proteins SpA and ZZT2 compared to their stable versions SpA-?galactosidase and ZZT0 were explained by their high proteolysis rate. The reduction of proteolysis rate in Clp protease-deficient strain enabled to increase the intracellular ZZT2 concentration two-fold, but impaired the cell growth. A method to calculate a hypothetical accumulation of the product assuming complete stabilization was described, which reveals a potential to increase the productivity by eliminating proteolysis. The effects of scale-up on the production of the ZZT2 protein were investigated in reactors of different configuration and scale: a well-mixed lab-scale bioreactor, an industrial scale 12-m3 bioreactor, and a two-compartment scale-down bioreactor.
