Persistent infection with hepatitis C virus (HCV) often causes chronic hepatitis, and then shows a high rate of progression to liver cirrhosis and hepatocellular carcinoma. To clarify the mechanism of the persistent HCV infection is considered to be important for the discovery of new target(s) for the development of anti-HCV strategies. In the present study, we found that the expression level of annexin A1 (ANXA1) in human hepatoma cell line Li23-derived D7 cells was remarkably lower than that in parental Li23 cells, whereas the susceptibility of D7 cells to HCV infection was much higher than that of Li23 cells. Therefore, we hypothesized that ANXA1 negatively regulates persistent HCV infection through the inhibition of viral RNA replication. The results revealed that HCV production was significantly inhibited without a concomitant reduction in the amount of lipid droplets in the D7 cells stably expressing exogenous ANXA1. Six chicken 7S immunoglobulin (Ig) allotypic specificities were demonstrated with antisera produced in highly inbred chickens. A radioimmunoassay which utilized intact, radiolabeled 7S Ig alloantigens was developed, and was used for the structural and genetic analysis of these specificities. The binding of dinitrophenylated alloantiserum (DAA) with radiolabeled alloantigen was demonstrated by the addition of rabbit anti-dinitrophenyl antiserum followed by precipitation of complexes with sheep anti-rabbit γ-globulin antiserum. Binding inhibition assays with the DAA method detected allotypes on 2 to 5 ng of 7S Ig alloantigen. Structural analyses of the six allotypic specificities were performed by direct binding and binding inhibition analysis with the DAA assay. Genetically controlled intraspecific differences between immunoglobulins (allotypes) provide valuable markers for the study of the quantitative expression of allelic and nonallelic alternative forms of immunoglobulins (Igs) during the normal development of rabbits. Heterozygous rabbits are mosaics of cells expressing different Ig-genes since fully differentiated productive cells generally secrete only one of alternative forms of Ig. The proportions of cells that differentiate to produce allelic forms of immunoglobulins during normal development depend on the particular heterozygous genotype. Hepatitis C Virus (HCV) infection causes liver disease, affecting approximately 200 million people worldwide. In most individuals acute HCV infection is asymptomatic prior to the development of persistent infection in approximately 80% of infected individuals leading to life-long infection. This results in active inflammation, fibrosis and cirrhosis of the liver, in some cases developing into hepatocellular carcinoma. In the remaining 20% of infected patients, spontaneous clearance of the virus is believed to be due to a broad, multi-specific CD4 and CD8 T cell response. Despite the advent of new direct acting antivirals which cure a high proportion of treated patients, many patients will not be cured or will not be treated due to the high cost of therapy. Therefore it is important to investigate immunotherapeutic strategies to combat persistent HCV infection. Previous studies show that antibody treatments that inhibit the protein, called programmed cell death 1 (PD-1), can shrink tumors in humans. This new work suggests that anti-PD-1 antibodies might be equally effective in treating hepatitis C and other persistent human viral infections, says Christopher Walker, PhD, a senior author on the study and director of the Center for Vaccines and Immunity at Nationwide Childrens.. PD-1 is a regulatory protein that helps keep the immune system in check. Normally, PD-1 acts as a switch to turn off immune responses when an infection is under control. Some viruses such as HCV manipulate the PD-1 off switch so that T cells lose their ability to fight the infection, a condition scientists call T-cell exhaustion. Among the disease-initiating mechanisms, host genetic factors are thought to be important, but there is a paucity of genetic studies and to date only a few probable susceptibility factors have been described1. Our aim is to spur further genetic research by postulating Ig GM allotypes - highly polymorphic hereditary antigenic determinants expressed on γ chains - as possible risk factors for IgG4-related disease. GM (IgG marker) allotypes are encoded by 3 very closely linked genes on chromosome 14q32. They are localized on the constant region of γ1, γ2, and γ3 chains2. Heterozygous rabbits of genotype a1n81f73g74/a2n82f71g75 were suppressed at birth for the VH region al allotype. At 8 weeks of age, quantitative analysis of serum IgG, IgM, and IgA molecules showed that the VHal specificity was effectively suppressed in the three classes of Ig and that the suppression was extended to the CH region n81 specificity on µ-chains as well as to the CH region f73 and g74 specificities on αf and αg chains. At 26 weeks of age, analysis of serum IgG and IgM molecules showed that a1 was still suppressed to approximately the same extent in both Ig classes and the suppression was still extended to the CH region n81 specificity. However, at 26 weeks, the percentage of molecules with a1 specificity had doubled among serum and colostral IgA molecules and this increase was extended to the CH region f73 and g74 specificities. Delayed and variable antibody responses to HCV make it difficult to diagnose acute HCV infection reliably. Immunoglobulin (Ig)M and IgG anti-HCV may be observed simultaneously as disease persists. IgM plays a key role in mixed cryoglobulinemia (MC), an immune complex disease strongly associated with persistent HCV infection. In MC, clonal or oligoclonal IgM rheumatoid factors facilitate the deposition of immune complexes in small blood vessels and tissue, leading to inflammation, complement activation and tissue damage. Clonally expanded IgM(+)κ(+) B-cells expressing rheumatoid factor-like IgM are abundant in many HCV patients with MC. The observation that identical or similar IgM antibodies are expressed in different patients clonally expanded B-cells supports the hypothesis that MC is driven by antigen-specific B-cell activation, rather than polyclonal B-cell activation or HCV replication in B-cells. More study is required to identify the antigens that drive the development of MC.

Rhesus macaques represent a valuable model in biomedical research and in development of vaccines and therapeutics. Due to the lack of reagents, the general properties of IgG and corresponding cellular receptors (FcγR) in this species are poorly characterized. We engineered recombinant IgGs containing each of the four rhesus macaque heavy constant region (CH) subclasses. To define FcγRs that mediate IgGs, we identified and characterized three FcγR classes, and generated recombinant cDNA constructs. cDNA IgH constructs were created by fusing - by sequence overlap extension PCRs - a gene segment encoding the murine variable heavy domain specific for the hapten NIP, an established specificity system for assessing antibody effector functions, with rhesus macaque CH fragments. The complete IgH constructs were transfected into J558L cells, a murine IgH-lost myeloma cell line expressing anti-NIP light chain.
