Last Thursday was my birthday. I am now 52 years or 18,993 days old.. While there were some beautiful moments that day, Im not going to pretend like it was perfect because it wasnt. Even though it was most certainly a day to celebrate life, I felt thrown off by some conversations and relationships that historically trigger me. While I have worked hard to address these types of reactions this year, on my birthday I regressed a bit. In that tender humanness, I was reminded of how inner peace is a practice, a journey not a destination. The incidence of pulmonary non-tuberculous mycobacteria (NTM) disease in otherwise healthy adults is increasing as the population ages. The organisms are ubiquitous so susceptibility probably reflects a deficiency in a protective immune response. Here we investigate if singlenucleotide polymorphisms (SNP) affecting cytokines, chemokines and their receptors associate with pulmonary NTM disease. Samples from NTM patients (n=79) and healthy controls (n=188) were genotyped using TaqMan probes. Of the 16 SNPs assessed, IL28B-rs8099917*TG (rs8099917; P=0.01, OR=2.2), TNFA-1031*CC (rs1799964; p=0.02, OR=0.48) and IL10-1082*AA (rs1800896; P=0.001, OR=0.33) were significantly associated with NTM disease. IL28B-rs8099917 and IL10-1082 have been associated with perturbations of the Th1/Th2 balance, whilst TNFA-1031*CC associates with sensory neuropathy in HIV patients. The anti-tumor effect of syngeneic spleen cells, xenogeneic immune RNA (l-RNA) and tumor antigen (TA) was studied in a chemically induced (N-methyl-N-nitro-sourethane) colon carcinoma model in BALB/c mice. When the mice were treated, sequentially, with these three elements by local injection under the tumor, complete tumor regression was observed in 25% of the animals. Complete tumor regression was observed in 17% of animals treated by injection of spleen cells pre-incubated with l-RNA in vitro followed by an injection of TA. When spleen cells were treated first with l-RNA and then with mitomycin-C, therapeutic benefit was obtained and the survival rate of mice treated with these spleen cells and TA was significantly higher than that of the control group (no treatment). The anti-tumor effect of this treatment was abrogated completely when spleen cells were treated with mitomycin-C prior to l-RNA incubation. Direct hemoperfusion therapy with polymyxin B immobilized fiber cartridge (PMX-DHP) is an established strategy in the treatment of septic shock in Japan and parts of Western Europe. PMX-DHP is currently the subject of a pivotal North American randomized controlled trial (EUPHRATES) in patients with septic shock and confirmed endotoxemia, as measured by the endotoxin activity assay. The major mechanism of action of this therapy is the removal of circulating endotoxin. High affinity binding of circulating endotoxin by the PMX-DHP column may decrease circulating endotoxin levels by up to 90% after two standard treatments. Basic research has shown reductions in circulating cytokine levels and in renal tubular apoptosis. Clinical research has shown that PMX-DHP therapy results in hemodynamic improvements, improvements in oxygenation, renal function, and reductions in mortality. Further research is needed to further define additional patient populations with endotoxemia that may benefit from PMX-DHP therapy
Adult house flies, Musca domestica L., are mechanical vectors of more than 100 devastating diseases that have severe consequences for human and animal health. House fly larvae play a vital role as decomposers of animal wastes, and thus live in intimate association with many animal pathogens. We have sequenced and analyzed the genome of the house fly using DNA from female flies. The sequenced genome is 691 Mb. Compared with Drosophila melanogaster, the genome contains a rich resource of shared and novel protein coding genes, a significantly higher amount of repetitive elements, and substantial increases in copy number and diversity of both the recognition and effector components of the immune system, consistent with life in a pathogen-rich environment. There are 146 P450 genes, plus 11 pseudogenes, in M. domestica, representing a significant increase relative to D. melanogaster and suggesting the presence of enhanced detoxification in house flies. Physiological activation of the N-methyl-D-aspartate (NMDA) subtype of glutamate receptors has been proposed to play a key role in both neuronal cell function and dysfunction. In the present study, we used selective NMDA receptor antagonists to investigate the involvement of NR2A and NR2B subunits in the modulatory effect of basal NMDA receptor activity on the phosphorylation of Tau proteins. We observed, in acute hippocampal slice preparations, that blockade of NR2A-containing NMDA receptors by the NR2A antagonist NVP-AAM077 provoked the hyperphosphorylation of a residue located in the proline-rich domain of Tau (i.e., Ser199). This effect seemed to be Ser199 specific as there was no increase in phosphorylation at Ser262 and Ser409 residues located in the microtubule-binding and C-terminal domains of Tau proteins, respectively. The use of fetal bovine serum (FBS) as a cryopreservation supplement is not suitable for the banking of mesenchymal stem cells (MSCs) due to the risk of transmission of disease as well as xenogeneic immune reactions in the transplanted host. Here, we investigated if human serum albumin (HSA), human serum (HS), or knockout serum replacement (KSR) can replace FBS for the cryopreservation of MSCs. In addition, we examined the characteristics of MSCs after multiple rounds of cryopreservation. The neighbourhood of algae and fungi in an aquatic environment is obviously of advantage only for the fungi, but the situation in terrestrial biotopes is changed. Fungi are especially common in the two uppermost soil layers (horizons A and B). The colonization of soils may originally have occurred after the dying of organisms left a sediment at the ground of inshore waters where fungi and bacteria settled. These nutrient-rich layers reached the earths surface by drying up or silting up of the inshore water. They became the biotopes of the first terrestrial fungi. Algae had at first only limited chances to survive, since they are dependent both on water and light. Light was available only in the uppermost, only a few millimetre thick layer where also the danger of drying out was greatest.
