Influence of seasonal variations in ambient temperatures on performance of immunochemical faecal occult blood test for...
Background Faecal occult blood testing (FOBT) in population screening has proved to be effective in reducing mortality from colorectal cancer. In Italy a latex agglutination FOBT has been adopted for a single-sample screening programme. The aim of this study was to examine the performance of FOBTs in the Florence screening programme over several seasons to evaluate the impact of variations in ambient temperature on the performance of the screening test.. Methods Measured haemoglobin (Hb) concentrations were aggregated into seasons with their average ambient temperature (AAT). Using logistic regression, the AAT over the period preceding the test measurement was analysed. This period included the time between faecal sampling and return of the test sample (mean 7 days) and the time in the laboratory refrigerator before analysis (mean 4 days). The AAT from days 5-11 before analysis of the test sample was considered a determinant of test positivity. The Kruskal-Wallis rank test was used to evaluate ...

Gender Differences in Fecal Immunochemical Test Performance for Early Detection of Colorectal Neoplasia
Background & Aims: Fecal immunochemical tests (FITs) are used widely in colorectal cancer screening. Programs use the same fecal hemoglobin threshold for colonoscopy referral for men and women, but it is unclear whether FIT performs equally in both sexes. We therefore assessed FIT performance in men and women. Methods: A prospective cohort study was performed, in which a total of 10,008 average-risk subjects (age, 50-74 y) were invited for first-round screening and 8316 average-risk subjects (age, 51-74 y) were invited for second-round screening with a single FIT. Subjects with a hemoglobin (Hb) level of 10 µg hemoglobin (Hb)/g (or =50 ng/mL) feces or higher were referred for colonoscopy. The test characteristics were assessed by sex for a range of FIT cut-off values. Results: In total, 59.8% of men and 64.6% of women participated in the first round (. P < .001). At a cut-off level of 10 µg Hb/g feces, the positivity rate was significantly higher among men (10.7%) compared with women (6.3%; ...

SMP-2 is an androgen-repressible protein. The negative regulation of the SMP-2 gene activity by androgen accounts for its increased expression during the androgen insensitive states of the prepubertal and senescent livers, and its constitutive expresion in the female liver. PBRN research has broadened to include a focus on improving practice quality through improved systems and processes. The next set of articles falls into this category.. The US Preventive Services Task Force includes stool tests among recommended tests used to screen for colorectal cancer.17 However, little research is available to guide practices in choosing between different types of fecal immunochemical tests (FITs). In addition to differences in costs and test accuracy-that is, sensitivity and specificity FITs differ in collection method, number of samples needed, and ease of labeling and processing of samples.18 These issues affect patient adherence and submission of adequately collected and processed samples. Pham et al19 assess patients experiences with and preferences for 6 different FITs. Their findings, particularly if they are ultimately shown to affect the return rates of usable samples, could inform a practices decision about which FITs to use.. The emergence of the ...

Be Like Dennis: Get Screened for Colon Cancer
Its a good thing Dennis Pella listened to his wife Pennie when she asked him to get their colon cancer screening tests from their family doctor because, little did Dennis know, he had colon cancer.. My wife received her colon cancer screening reminder letter in the mail, and I was going to see my doctor about another issue that week. So, my wife said to bring the letter in and get the requisition completed for both of our screening kits, said Dennis. Im not the kind of person who would run to see the doctor, so it felt out of character for me to ask to have my requisition completed too, but Im glad that I did.. The simple, at-home colon cancer screening test that the Pellas received in the mail, after having their requisition completed by their doctor, is called a fecal immunochemical test (FIT) kit. The FIT kit is Ontarios colon cancer screening test for average risk (no first-degree family history of colon cancer - e.g. parent, sibling, child) individuals between the ages of 50 and 74 ...

Background: Populations differ with respect to their cancer risk and screening preferences, which may influence the performance of colorectal cancer (CRC) screening programs. This review aims to systematically compare the mortality effect of CRC screening across European regions. Methods: Six databases including Embase, Medline, Web of Science, PubMed publisher, Google Scholar and Cochrane Library were searched for relevant studies published before March 2018. Bibliographic searches were conducted to select studies assessing the effect of various screening tests (guaiac fecal occult blood test [gFOBT]; flexible sigmoidoscopy [FS]; fecal immunochemical test [FIT] and colonoscopy) on CRC mortality in Europe (PROSPERO protocol: CRD42016042433). Abstract reviewing, data extraction and risk of bias assessment were conducted independently by two reviewers. Results: A total of 18 studies were included; of which, 11 were related to gFOBT, 4 to FS, 2 to FIT and 1 to colonoscopy; 8 were randomised ...
Faecal immunochemical test is superior to symptoms in predicting pathology in patients with suspected colorectal cancer...
To assess whether a faecal immunochemical test (FIT) could be used to select patients with suspected colorectal cancer (CRC) symptoms for urgent investigation.Multicentre, double-blinded diagnostic accuracy study in 50 National Health Service (NHS) hospitals across England between October 2017 and December 2019. Patients referred to secondary care with suspected CRC symptoms meeting NHS England criteria for urgent 2 weeks wait referral and triaged to investigation with colonoscopy were invited to perform a quantitative FIT. The sensitivity of FIT for CRC, and effect of relevant variables on its diagnostic accuracy was assessed.9822 patients were included in the final analysis. The prevalence of CRC at colonoscopy was 3.3%. The FIT positivity decreased from 37.2% to 19.0% and 7.6%, respectively, at cut-offs of 2, 10 and 150 µg haemoglobin/g faeces (µg/g). The positive predictive values of FIT for CRC at these cut-offs were 8.7% (95% CI, 7.8% to 9.7%), 16.1% (95% CI 14.4% to 17.8%) and 31.1% ...
Immunochemical characterization and toxicological significance of P-450HFLb purified from human fetal livers | Meta
Immunochemical properties of P-450HFLb purified from human fetal livers were investigated. P-450HFLb cross-reacted with antibodies to rat P-4501A1 but not with antibodies to CYP2A6, CYP2C9, CYP3A7 (P-450HFLa) and rat CYP2B1. In addition, P-450HFLb also cross-reacted with both monospecific antibodies to rat CYP1A1 and CYP1A2. However, P-450HFLb was shown to be an immunochemically distinct form of cytochrome P-450 from P-450PA (human CYP1A2). Immunoblot analysis of human fetal livers with the antibodies to P-450HFLb showed that P-450HFLb was expressed in all fetal livers studied although there appeared to be individual differences in the amounts of P-450HFLb expressed in fetal livers. The formation of mutagens from IQ (but not from AFB1) in fetal liver homogenates was inhibited by the antibodies to P-450HFLb in a dose dependent manner. These results suggest that P-450HFLb may be a form of human cytochrome P-450 classified into CYP1 gene family, and that the cytochrome P-450 is, in part, ...
UK receives $3.75M to promote screening, prevent colon cancer deaths in rural Kentucky - KyForward.com | KyForward.com
Over the next five years, Crosby and the RCPC team will develop, implement and disseminate an intervention to promote a simple, at-home screening test called FIT (fecal immunochemical test) that could drastically increase rates of annual colorectal cancer screening in rural areas. FIT tests use a new technology for detecting antibodies to polyps (potentially cancerous clumps of cells) in the stool. With FIT, you simply brush the surface of the stool with a brush included in the kit and then dab the end of the brush onto the test card, which is mailed off for testing. The test is quick, painless, low-cost, and doesnt require a trip to the doctor - still somewhat unpleasant, but likely preferable to an unnecessary colonoscopy or, of course, cancer. In the case of a positive FIT test result, RCPC staff will help individuals navigate the healthcare system to get further testing and treatment as needed ...
