Flavobacterium columnare is the causative agent of columnaris disease, a disease affecting many freshwater fish species. Methods for the genetic manipulation for some of the species within the Bacteroidetes, including members of the genus Flavobacterium, have been described, but these methods were not adapted to work with F. columnare. As a first step toward developing a robust set of genetic tools for F. columnare, a protocol was developed to introduce the E. coli - Flavobacterium shuttle vector pCP29 into F. columnare strain C#2 by conjugal mating at an efficiency of 1.5 × 10-3 antibiotic-resistant transconjugants per recipient cell. Eight of eleven F. columnare strains tested were able to receive pCP29 using the protocol. pCP29 contains the cfxA and ermF genes, conferring both cefoxitin and erythromycin resistance to recipient cells. Selection for pCP29 introduction into F. columnare was dependent on cfxA, as ermF was found not to provide strong resistance to erythromycin. This is in contrast to

MDC staff assessed blue catfish and flathead catfish populations in Mark Twain Lake from 2012 through 2018. Findings indicate that a minimum length limit could improve catfish numbers in Mark Twain by increasing the abundance of preferred sized catfish (30-inch) and memorable sized catfish (35-inch) with minimal or no reductions in the yield, or pounds harvested by anglers.. MDC gathered public input online and through a mail survey to anglers returning tags from blue catfish caught in Mark Twain Lake. About two-thirds of the surveyed anglers support a minimum length limit for blue and flathead catfish at Mark Twain Lake. The Robertsonian-translocation hypothesis suggests a regular fusion of each ancestral acro- or telocentric chromosome to just one other by centromere fusions, thus halving the karyotype. An alternative genome-stirring hypothesis posits haphazard and repeated fusions, inversions, and reciprocal and nonreciprocal translocations. It was a fun filled memorial weekend at the lake and to top it off the fishing was great. The crappie were active around the 15ft depth. Minnows seemed to be the best method for catching crappie; most were caught around the docks and various brush piles on the lake. The channel catfish didnt take the weekend off as they were biting on most baits including, dough bait, chicken liver and minnows. The bigger channels were seen caught around brush piles in 18ft of water. The fighter jets of the under water environment, aka Sandbass, were active around the 6ft-10ft depth. Some very nice sandies were caught around the sand bar and south east shorelines with light colored crank-baits while trolling. A few saugeye were seen caught on shad colored crank-baits. I have started a new 29-gallon tank. So far I have 5 zebra danios and a dwarf gourami in there. In the future I would like to add a rainbow shark and some sort of catfish.
