Actin filaments are normally arranged as large stress fibres, even during early stages of infection, but become redistributed as densely staining regions near the centre of the syncytium in latent infection. Microtubules and vimentin filaments (easily visible in early infection) lose all visible organization during syncytium formation. Stabilization of microtubules with taxol results in the production of elongate syncytia with regularly organized microtubules. Conversely, disruption of microtubules with nocodazole inhibits cell fusion and causes a reduction in infectious virus production. This reduction upon treatment with nocodazole, and the subsequent rescue of virus production upon addition of taxol indicates that viral replication relies upon the presence of microtubules.

Microscopic examination of skin scrapes before treatment confirmed the presence of bacteria indicative of columnaris disease. In separate trials, walleyes (12 g) and channel catfish (2.3 g) were exposed to a total of three every-other-day hydrogen peroxide treatments of 0, 50, 75, or 100 mg/L for 60 min. Each treatment regimen was tested in triplicate, and each replicate contained 13 walleyes or 26 channel catfish. Hydrogen peroxide exposures of 50 mg/L for 60 min significantly increased walleye survival relative to that at other treatment concentrations, including the controls. While most Channel Catfish fisheries are managed under liberal harvest regulations, the Red River of the North (Red River) in Manitoba, Canada is managed with restrictive harvest regulations to promote a trophy fishery. Two barriers (dams) are present on the main stem of the Red River and may fragment the population to some degree. My objectives were to: 1) analyze population dynamics of the trophy Channel Catfish population on the lower Red River, 2) compare population characteristics of Channel Catfish in selected reaches throughout the Red River in Manitoba, and 3) determine movement characteristics of Channel Catfish and the permeability of a dam on the lower Red River. Interpretive Summary: Technical Abstract: Motile Aeromonas septicemia (MAS), caused by virulent clonal isolates of Aeromonas hydrophila (vAh), is emerging as a major disease in channel catfish (Ictalurus punctatus) aquaculture in the Southeastern United States. Predisposing conditions leading to vAh infection in catfish were however largely unknown. The objective of this study was to investigate factors that predispose catfish to vAh infection and to understand the pathogenesis of MAS. ABSTRACT: The adhesion dynamics of Flavobacterium columnare to fish tissues were evaluated in vivo by immersion challenge followed by bacterial plate count and confirmatory observations of gill-adhered bacterial cells using scanning electron microscopy. Adhesion of F. columnare genomovar I (ARS-1) and II (BGFS-27) strains to skin and gill of channel catfish Ictalurus punctactus and gill of zebrafish Danio rerio was compared. At 0.5 h post-challenge, both strains adhered to gill of channel catfish at comparable levels (106 colony forming units [CFU] g-1), but significant differences in adhesion were found later in the time course. The fumonisins represent a group of mycotoxins produced predominantly by Fusarium moniliforme species. The toxic effects in fish remains still poorly understood. In one study, channel catfish fed F. moniliforme culture material containing 313 ppm of fumonisin B1 (FB1) for 5 weeks revealed minimal adverse effects27. Conversely, another study28 reported that dietary levels of FB1 of 20 ppm or above are toxic to year-1 and year-2 channel catfish. After 10 and 14 weeks, respectively, year-1 and year-2 catfish fed 20 ppm or more of FB1 in the diet had lower weight gain compared to the control, and those fish fed diets with levels of 80 ppm and above showed significantly lower hematocrits and red and white blood cells than those fed lower doses29. Live-attenuated vaccines against Edwardsiella ictaluri or against Pasteurella piscicida are disclosed. Both vaccines are incapable of reversion to virulence, because both are made by deletion mutations in the aroA gene, the purA gene, or both. These vaccines may be used not only to vaccinate fish against Edwardsiella ictaluri or Pasteurella piscicida, but also to serve as vectors to present antigens from other pathogens to the fish, thereby serving as vaccines against other pathogens as well, with no risk of infection by reversion to the virulent form of the pathogen in which the antigen occurs naturally. ...

As IleRS can inadvertently accommodate and process structurally similar amino acids such as valine, to avoid such errors it has two additional distinct tRNA(Ile)-dependent editing activities. One activity is designated as pretransfer editing and involves the hydrolysis of activated Val-AMP. The other activity is designated posttransfer editing and involves deacylation of mischarged Val-tRNA(Ile).
