Dengue continues to be a major international public health concern. Despite that, there is no clinically approved antiviral for treatment of dengue virus (DENV) infections. In this study, geraniin extracted from the rind of Nephelium lappaceum was shown to inhibit the replication of DENV-2 in both in vitro and in vivo experiments. The effect of geraniin on DENV-2 RNA synthesis in infected Vero cells was tested using quantitative RT-PCR. The in vivo efficacy of geraniin in inhibiting DENV-2 infection was then tested using BALB/c mice with geraniin administered at three different times. The differences in spleen to body weight ratio, DENV-2 RNA load and liver damage between the three treatment groups as compared to DENV-2 infected mice without geraniin administration were determined on day eight post-infection. Quantitative RT-PCR confirmed the decrease in viral RNA synthesis of infected Vero cells when treated with geraniin. Geraniin seemed to provide a protective effect on infected BALB/c mice liver

Ellagitannins are plant phenolics characterized by biaryl-coupled gallic acid moieties esterified to a D-glucose core. They are widely distributed through higher plants. In the case of oaks, ellagitannin concentrations in heartwood can reach up to 10% (dry wt. basis). These secondary metabolites are not only important physiologically but also influence the economic value and quality of wood products that contain them. Efforts were made to develop and validate the methods used to quantify both soluble and insoluble ellagitannins. First, the efficiencies of the two commonly used extraction solvents, aqueous acetone and aqueous methanol were evaluated. The results showed that aqueous acetone is superior to aqueous methanol in obtaining higher vescalagin and castalagin yields. In a separate study, the method used for determining insoluble ellagitannins was found to under-estimate the contents of insoluble ellagitannins in wood products. Anhydrous methanolic HCl was found to be an excellent reagent ...

Abstract: Recent in vitro and in vivo preclinical studies have suggested that the Oriental herbal compound penta-1, 2, 3, 4, 6-O-galloyl-beta-D-glucose (PGG) is a promising chemopreventive agent for prostate cancer. Little is known of its safety for chronic chemoprevention use and virtually nothing is known of ...
Human enterovirus 71 is one of the major causative agents of hand, foot and mouth disease in children under six years of age. Presently, no vaccines or antiviral drugs have been clinically available to employ against EV71. In this study, we demonstrate that treatment with chebulagic acid reduced the viral cytopathic effect on rhabdomyosarcoma cells with an IC50 of 12.5 μg/mL. The utilization of the chebulagic acid treatment on mice challenged with a lethal dose of enterovirus 71 was able to efficiently reduce mortality and relieve clinical symptoms through the inhibition of viral replication. Chebulagic acid may represent a potential therapeutic agent to control infections to enterovirus 71.

Walnuts have been gathering attention for their health-promoting properties. They are rich in polyphenols, mainly ellagitannins (ETs) that after consumption are hydrolyzed to release ellagic acid (EA). EA is further metabolized by microbiota to form urolithins, such as A and B, which are absorbed. ETs, EA and urolithins have shown to slow the proliferation and growth of different types of cancer cells but the mechanisms remain unclear. We investigate the role of urolithins in the regulatory mechanisms in prostate cancer, specifically those related to the androgen receptor (AR), which have been linked to the development of this type of cancer. In our study, urolithins down-regulated the mRNA and protein levels of both prostate specific antigen (PSA) and AR in LNCaP cells. The luciferase assay performed with a construct containing three androgen response elements (AREs) showed that urolithins inhibit AR-mediated PSA expression at the transcriptional level. Electrophoretic mobility shift assays ...
True polyphenols from the tannin and other WBSSH types are routinely biosynthesized in the natural sources from which they derive; their chemical syntheses (using standard "bench" organic chemical methods) were somewhat limited until the first decade of the new millennium because these syntheses involve challenging regioselectivity and stereoselectivity issues.[11] Early work focused on the achiral synthesis of phenolic-related components of polyphenols in the late 70s,[12] and the Nelson and Meyers synthesis of the permethyled derivative of the ubiquitous diphenic acid core of ellagitannins in 1994[13] followed by stereoselective synthesis of more complex permethylated structures such as a (+)-tellimagrandin II derivative by Lipshutz and coworkers in the same year,[14] and Itoh and coworkers synthesis of a permethylated pedunculagin with particular attention to axial symmetry issues in 1996.[15] The total synthesis of a fully unmasked polyphenol, that of the ellagitannin tellimagrandin I, ...
Polyphenol interactions with both cellulose and collagen in the solid state have been studied by using chromatography on cellulose and by evaluating the hydrothermal stability of the polyphenol treated sheepskin collagen. Twenty-four polyphenolic compounds were studied, including seven glucose-based gallotannins, five polyalcohol-based gallotannins, and twelve ellagitannins. In the cellulose-polyphenols systems, the polyphenols affinity to cellulose is positively correlated with their molecular masses, the number of galloyl groups, and their hydrophobicity (logP). The polyphenol treatment increased the hydrothermal stability of collagen samples, and such effects are also positively correlated with the molecular masses, total number of galloyl groups and the hydrophobicity of polyphenols. Ellagitannins showed much weaker interactions with both biopolymers than gallotannins having similar molecular mass, the same number of galloyl groups, and the same number of phenolic hydroxyl groups. It is ...

The most relevant pomegranate phenolics (ellagitannins and ellagic acid) are extensively metabolized by the human gut microbiota to yield a number of metabolites called urolithins (mainly Uro-A). Urolithins have been reported to regulate in vivo the expression of genes involved in inflammation and cancer. Our hypothesis is that urolithins can be detected in the human colon mucosa where these metabolites can exert anti-inflammatory and anti-cancer activities. After colonoscopy and diagnosis, colorectal cancer patients will consume capsules containing three different pomegranate extract formulations until surgery. The aims of this trial are:. ...

The strange-looking leathery exterior of the pomegranate doesnt even hint at the deliciously juicy, sparkling little jewels inside. Beneath the rind, the fruit is densely packed with small white seeds, each enveloped in its own bright, clear packet of juice, somewhat like orange pulp inside an orange segment. As much as I am delighted by the flavour and visual appeal, what most impresses me about the pomegranate is what it can do for our health. Heres what research has found. Scientists took some cancer cells from men with highly aggressive prostate cancer. The cells were allowed to grow in tissue cultures in the lab. When extracts from pomegranate were added, the growth of the cancer slowed. The researchers then took those aggressive cancer cells and injected mice with them. The mice that got pomegranate juice in their drinking water developed significantly smaller tumors than mice that didnt get pomegranate juice.. Pomegranate juice is also beneficial for those with coronary heart disease. ...

Pomegranate May Reduce Stomach Pain and Inflammation Pomegranates are one of the trendiest health foods on the market today, celebrated for their high antioxidant content and potential therapeutic benefits. The fruit has been used for centuries to treat inflammatory diseases, prompting researchers from Mexico, Italy and Spain to explore the science behind pomegranate as a potential remedy for stomach pain and inflammation. Results from the 2015 study are published in the International Journal of Food Sciences and Nutrition.. Using extract from whole fruit, researchers tested how the polyphenols in pomegranates may alleviate stomach pain and inflammation in an animal model. Not only was the pomegranate extract effective in reducing pain and inflammation, it worked better than the anti-inflammatory drugs often given to people (indomethacin and diclofenac, both NSAIDs). Pomegranate extract also offered protection to the gastric mucosal barrier, the outer lining of the stomach that prevents stomach ...

Research has shown that the antioxidant content of pomegranate juice can reverse atherosclerosis, lowering ones risk of heart disease. Lets take this one step further. Instead of drinking a blood-sugar-spiking 8oz. glass of pomegranate juice, cut open the cheery red fruit and eat its arils (the red pods nestled within the white pulp.) Youll still reap the full benefit of the antioxidants, plus youll get an added dose of insoluble fiber. Not to mention the gustatory experience you will get to enjoy when you crunch into those arils, and their juice bursts out into your mouth. Some people are shy about preparing and eating a pomegranate because it looks confusing and theyre not sure which parts are edible. Never fear, because there is an easy way to prepare this superfood of a fruit. Fill a medium sized bowl with cold water. After rinsing your pomegranate, set it "tuft" side up on a glass (or any other material that wont stain) cutting board. Make one cut through the tuft from top to bottom, ...

The least creative messages left on pomegranates Home page were deposited there by a would-be hacker who realized that we had a security hole in the script which allowed a person to save and modify the form to take any input desired. Before we plugged up the hole with some server-side validation, messages such as Linux eats windows for breakfast, Linux eats Windows95 for breakfast, and Baby, oh, baby. Damn, Im good! were displayed on pomegranates Home page for short periods of time. The latter message was displayed twice (perhaps because of a lack of ideas for further comments), and after the hacker realized how to do it the messages were accompanied by a small graphic from some porn site which no longer seems to exist. That image, no more innovative than the Baby, oh, baby mantra and less revealing than some of our art photography, displayed in a closely cropped composition spanning from belly-button to mid-thigh the voluptuous hips of a woman clad in high-cut purple underwear. A remarkable study, published in the journal Clinical Nutrition, took 10 patients with atherosclerosis and measured their carotid arteries before and after one year of supplementing their diet with pomegranate juice. After one year, the plaque clogging their carotid arteries was reduced by up to 30%, while the control groups blockages increased by 9% - effectively reversing the disease process by up to 39% within one year. Additional "side benefits" included a 130% increase in their antioxidant status, with LDL susceptibility to oxidation (it is far more important to maintain the quality of the LDL, i.e. keep it from getting damaged, than to nit-pick about the "right number") being reduced 59%. Finally, systolic blood pressure was reduced 21% within the first year. Wow, right? But it only gets better ...
