A cell line designated MES-SA has been developed from a uterine sarcoma. Cells from the surgical tumor specimen were grown in a soft-agar clonogenic assay, with a relatively high plating efficiency of 0.5% and sensitivity to multiple drugs. Indian childhood cirrhosis (ICC) is an almost uniformly fatal disease whose outcome may be modified with penicillamine if given at a sufficiently early stage. Twenty nine children with ICC seen in Pune, India, in 1980-7, who had survived at least five years from onset of penicillamine treatment, were reviewed aged 6.3 to 13 years. They were assessed clinically, biochemically, histologically, and by duplex Doppler ultrasound examination. None had symptoms suggestive of liver disease. There were no toxic effects of penicillamine other than asymptomatic proteinuria. Hepatosplenomegaly reduced significantly and liver function tests returned to normal in all. In four children, significant hepatosplenomegaly was associated with an abnormal duplex Doppler hepatic vein flow pattern and micronodular cirrhosis on biopsy. Unless you take pains to ensure reproducibility, you may encounter unpredictable staining results. PTA functions as a dye excluder, so that the operative acid dyes can stain differentially. If not enough PTA is present, the acid dyes will perform as though no PTA is present and NOT stain differentially. PTA is hygroscopic. It naturally absorbs moisture from the atmosphere. In the worst case, it appears grossly to be like school paste of the kind provided to me in the dark ages. Bottom line: when you weigh out PTA with much water and add it to a stain formulation, the stain will perform as though no PTA has been added. Ive seen the effect in Pap EA formulations, the origins of which are rooted in Mallorys trichrome stain. In such instances, the EA is awful.
