The application of digital holographic microscopy offers quantitative phase contrast imaging of reflective and (partially) transparent samples. Low coherent light sources enable an increased resolution in phase shifting digital holography by the reduction of stray light and multiple reflections in the experimental interferometer setup. Therefore light emitting diodes (LED) are utilized. Here, the effects on the reconstruction quality by considering the whole spectrum of the light source are investigated. Furthermore, the propagation of the complex wave fronts, which are determined by digital holography, and used for re- and multifocusing, is modified to light sources with a spectral width wider than that of typical laser light sources. This paper present a novel approach to perform the tomography of biological specimen based on Digital Holographic Microscopy (DHM). A hologram results from the interference between a reference wave and an object wave reflected from or transmitted through a sample. In the hologram, both amplitude and phase of the field transmitted through the object are registered. In DHM, the object field is recovered when the hologram is processed by a digitally computed replica of the reference wave, allowing quantitative measurement of both phase and amplitude. Phase measurements provide high accuracy optical path length measurements across the specimen along the optical axis. We propose parallel phase-shifting digital holographic microscopy (PPSDHM) which has the ability of three-dimensional (3-D) motion measurement using space-division multiplexing technique. By the PPSDHM, instantaneous information of both the 3-D structure and the phase distributions of specimens can be simultaneously acquired with a single-shot exposure. We constructed a parallel phase-shifting digital holographic microscope consisting of an optical interferometer and an image sensor on which micro polarizers are attached pixel by pixel. We propose a fully automatic technique to obtain aberration free quantitative phase imaging in digital holographic microscopy (DHM) based on deep learning. The traditional DHM solves the phase aberration compensation problem by manually detecting the background for quantitative measurement. This would be a drawback in real time implementation and for dynamic processes such as cell migration phenomena. A recent automatic aberration compensation approach using principle component analysis (PCA) in DHM avoids human intervention regardless of the cells motion. However, it corrects spherical/elliptical aberration only and disregards the higher order aberrations. Traditional image segmentation techniques can be employed to spatially detect cell locations. Ideally, automatic image segmentation techniques make real time measurement possible. However, existing automatic unsupervised segmentation techniques have poor performance when applied to DHM phase images because of aberrations and speckle noise. ...
Red blood cell (RBC) membrane fluctuations provide important insights into cell states. We present a spatial analysis of red blood cell membrane fluctuations by using digital holographic microscopy (DHM). This interferometric and dye-free technique, possessing nanometric axial and microsecond temporal sensitivities enables to measure cell membrane fluctuations (CMF) on the whole cell surface. DHM acquisition is combined with a model which allows extracting the membrane fluctuation amplitude, while taking into account cell membrane topology. Uneven distribution of CMF amplitudes over the RBC surface is observed, showing maximal values in a ring corresponding to the highest points on the RBC torus as well as in some scattered areas in the inner region of the RBC. CMF amplitudes of 35.9+/-8.9 nm and 4.7+/-0.5 nm (averaged over the cell surface) were determined for normal and ethanol-fixed RBCs, respectively.

A new class of holograms, having the properties of full-color reconstruction from a single white light source, an extended vertical viewing aperture, and extended scene depth is disclosed. These advantageous properties are provided by a hologram composed of three intermeshed holograms, each of which reconstructs only one of the three color components of the scene. Each color component hologram consists of an array of noncontiguous small dots or thin stripes so that the three-color component holograms may be intermeshed without overlap of the dots or stripes. Associated with each color component hologram is a similar array of dot or stripe color filters which allow only the appropriate color of light to reconstruct each color component hologram. The component holograms themselves are vertically focused so that the composite hologram is white light viewable.
This dissertation combines various holographic techniques with application on the two- and three-dimensional imaging of ophthalmic tissue, fingerprints, and microsphere samples with micrometer resolution. Digital interference holography (DIH) uses scanned wavelengths to synthesize short-coherence interference tomographic images. We used DIH for in vitro imaging of human optic nerve head and retina. Tomographic images were produced by superposition of holograms. Holograms were obtained with a signal-to-noise ratio of approximately 50 dB. Optic nerve head characteristics (shape, diameter, cup depth, and cup width) were quantified with a few micron resolution (4.06 -4.8 microns). Multiple layers were distinguishable in cross-sectional images of the macula. To our knowledge, this is the first report of DIH use to image human macular and optic nerve tissue. The understanding of cell-surface interactions plays an important role for the biomaterials development and bioengineering. Although it is already known that amine groups increase the cell adhesion and proliferation, the influence of amine layers properties on cell viability is the subject of further investigation. In this work, amine-rich coatings were prepared by low pressure plasma polymerization of cyclopropylamine using radio frequency (RF) capacitively coupled discharge. Normal human dermal fibroblasts were chosen for the monitoring of biological response to the properties of amine layers. In this study, multi-planar Nearfield Acoustical Holography (NAH) is used to investigate noise radiated from the front, side and rear areas of single tires on a two-wheel trailer. Contributions to the radiated noise from the leading edge, trailing edge, and sidewall of the tire are identified. Two tires - an experimental monopitch tire and a production passenger car tire - are evaluated on a smooth asphalt pavement at 58 km/hr. From the measured complex pressure, acoustic intensity is reconstructed on three planes surrounding the tire using modified NAH procedures. When one looks at a scene, each eye captures a portion of the light scattered from the scene, and the lens of the eye forms an image of the scene on the retina, in which light from each angular position is focused to a specific angular position in the image plane. Since the hologram reconstructs the whole of the scattered light field that was incident on the hologram, the viewer sees the same image whether it is derived from the light field scattered from the object, or the reconstructed light field produced by the hologram, and is unable to tell whether he or she is looking at the real or the virtual object. A method is described whereby projected lithographic images can be formed with x-rays by means of a transmission hologram. The form of the hologram is computed by an algorithm which eliminates all the unwanted signals normally present as systematic errors in holographic images. An example calculation is shown in which the form of a hologram is derived that is supposed to generate a diffraction image of a given test pattern. The holographic image is shown to have excellent fidelity with respect to both resolution and contrast. Such an approach to projection x-ray lithography requires an x-ray beam with very little coherence and is thus compatible with high wafer throughput schemes while at the same time being relatively immune to contamination.*LITHOGRAPHY

Here, the mapping of electrostatic potentials and magnetic fields in liquids using electron holography has been considered to be unrealistic. Here, we show that hydrated cells of Magnetospirillum magneticum strain AMB-1 and assemblies of magnetic nanoparticles can be studied using off-axis electron holography in a fluid cell specimen holder within the transmission electron microscope. Considering that the holographic object and reference wave both pass through liquid, the recorded electron holograms show sufficient interference fringe contrast to permit reconstruction of the phase shift of the electron wave and mapping of the magnetic induction from bacterial magnetite nanocrystals. We present measurements of the full Jones matrix of individual pixels in a liquid-crystal display (LCD) panel. Employing a polarization-sensitive digital holographic microscopy based on Mach-Zehnder interferometry, the complex amplitudes of the light passing through individual LCD pixels are precisely measured with respect to orthogonal bases of polarization states, from which the full Jones matrix components of individual pixels are obtained. My previous work included using chlorophyll fluoresecence and radiolabelled carbon to investigate estuarine microphytobenthic photophysiology and primary production; a NERC funded project working with Prof. Graham Underwood at Essex University. Abstract: We consider U(1) gauge fields in a slice of AdS_5 with bulk and boundary mass parameters. The zero mode of a bulk U(1) gauge field can be localized either on the UV or IR brane. This leads to a simple model of millicharged particles in which fermions can have arbitrarily small electric charge. In the electroweak sector we also discuss phenomenological implications of a localized U(1)_Y gauge boson. Using the AdS/CFT correspondence we present the 4D holographic interpretation of the 5D model. In particular the photon is shown to be a composite particle when localized near the IR brane, whereas it is elementary when localized near the UV brane. Could one replace AdS/CFT correspondence with TGD version of holography? One can criticize AdS/CFT based holography because it has Minkowski space only as a rather non-unique conformal boundary resulting from conformal compactification. Situation gets worse as one starts to modify AdS by populating it with blackholes. And even this is not enough: one can imagine anything inside blackhole interiors: wormholes connecting them to other blackholes, anything. Entire mythology of mystic creatures filling the white (or actually black) areas of the map.
