Assembly of hemoglobin subunits into cooperative complexes produces a remarkable variety of architectures, ranging in oligomeric state from dimers to complexes containing 144 hemoglobin subunits. Diverse stereochemical mechanisms for modulating ligand affinity through intersubunit interactions have been revealed from studies of three distinct hemoglobin assemblages. This mechanistic diversity, which occurs between assemblies of subunits that have the same fold, provides insight into the range of regulatory strategies that are available to protein molecules.
The commercial feasibility of recombinant human Hb (rHb) as an O2 delivery pharmaceutical is limited by the production yield of holoprotein in E. coli. Currently the production ofrHb is not cost effective for use as a source in the development of third and fourth generation Hb-based oxygen carriers (HBOCs). The major problems appear to be aggregation and degradation of apoglobin at the nominal expression temperatures, 28-37° C, and the limited amount of free heme that is available for holohemoglobin assembly. The likelihood of developing OA increases with age. The prevalence of OA of the knee is higher among 70- to 74-year-old. Epidemiological studies have revealed that there are both endogenous and exogenous risk factors for OA. The pathogenesis of OA is characterized by the progressive destruction of articular cartilage. Many factors contribute to the overall degradation of cartilage observed in OA, either directly or indirectly by modulating anabolic factors [14]. The capacity of articular cartilage to repair is very limited, mainly because it is an avascular tissue [42].
