Indirect haemagglutination test using gonococcal pilus antigen: how useful to diagnose gonorrhoea? | Sexually Transmitted...
In 1979 an indirect haemagglutination test (gonococcal antibody test) using gonococcal pilus antigen replaced the gonococcal complement fixation test as our routine procedure to show gonococcal antibodies. In the diagnosis of current gonorrhoea the sensitivity of the gonococcal antibody test was far superior to that of the gonococcal complement fixation test (about 55% versus 9% for first episode gonorrhoea). To evaluate the usefulness of the test result the following population groups were studied: 1376 patients undergoing medical examination for gonorrhoea (386 had gonorrhoea), 1384 healthy people aged 15-65, 54 patients with meningococcal disease, 30 children with respiratory tract infection, and 254 patients with evidence of various diseases other than neisserial infections that might be associated with symptoms of arthritis. These investigations showed that (1) non-specific positive gonococcal antibody test results occur rarely, (2) at least half the people who have had gonorrhoea remain ...
Evaluation of an indirect hemagglutination test for Legionella pneumophila serogroups 1 to 4. | Journal of Clinical Microbiology
Parallel testing of 895 sera by indirect hemagglutination and indirect fluorescent-antibody techniques showed 97.3% agreement. Although the indirect hemagglutination technique usually showed more cross-reactivity among serogroups than the indirect fluorescent-antibody technique with Formalin-fixed antigens and a conjugate which detected primarily immunoglobulin G antibodies, heterologous serogroup reactions were significantly lower than homologous serogroup titers and the etiological serogroup could be easily defined. The indirect hemagglutination techniques showed no cross-reactivity with a crude extract of Escherichia coli O13:K92:H4. Since the indirect hemagglutination technique was shown to detect both immunoglobulin M and immunoglobulin G antibodies and was found to be rapid, simple, and inexpensive, it appears to be an excellent alternative to the indirect fluorescent-antibody test for serodiagnosis of legionellosis. ...

Infection with Toxoplasma has been found to be widespread in the human population and, as a consequence, is receiving increasing attention. The clinical manifestations may range from inapparent infection or benign disease to severe systemic disease or a chronic stage with eye disease and encephalopathy and mental retardation, or both. Because of the variable nature of the disease and the high rate of inapparent infections, laboratory diagnostic methods are essential to proper diagnosis.
Serologic tests have received a great deal of emphasis as diagnostic aids in suspected infections with Toxoplasma, the methylene-blue dye test being the most widely used. In recent years the indirect hemagglutination (IH) test has been reported to be a sensitive and specific serologic test that yields titers comparable to or slightly higher than the dye test. Because of its simplicity and safety to the laboratory worker, it has been recommended as a useful test for the serologic diagnosis of toxoplasmosis.
Particle Gel Immuno Assay (ID-PaGIA) zum Nachweis von anti-IgA Antikörpern
Anti-IgA antibodies are thought to be responsible for non-hemolytic transfusion reactions in one in 17,000 to one in 770,000 number of cases. This incidence is mainly supported by case reports. Despite their relative frequency of one in 18 to one in 1,250, since their discovery approximately forty years ago, the true significance of these antibodies has not yet been determined. Several specificities of these antibodies resulting in different reaction patterns make diagnosis and categorization difficult. Until recently, the lack of a fast and reliable laboratory test was a drawback. This test needed to be easily performed, fast, accurate, reproducible and accessible to many practitioners in many laboratories. The Passive Hemagglutination Assay (PHA), developed in the late 1960s, is neither precise nor reliable but easy to perform and therefore has been the mainstay in diagnosis of anti-IgA. While newer methods, such as Radio Immuno Assay (RIA) and Enzyme Linked Immunosorbent Assay (ELISA), are ...

Immune adherence hemagglutination: alternative to complement-fixation serology. - Semantic Scholar
Immune adherence hemagglutination (IAHA) was compared to complement fixation (CF), using standard procedures, for serological testing of human sera with a number of commercially available antigens. The antigens included herpes simplex, measles, cytomegalo-, and influenza (type B) viruses, as well as Mycoplasma pneumoniae and Chlamydia psittaci (Chlamydia group). The IAHA test was found to be as specific as the CF test, but 4 to 20 times as sensitive with all antigens tested. Antigen titers were also higher with the IAHA method, and the time required to complete the test was only 4 h for the IAHA method, compared with 20 h for the CF method. The increased sensitivity of the IAHA test should permit its use for determination of immunity, as well as for serodiagnosis of recent infections.
Aluminum-Magnesium Silicate enhances release of virions of cultured |i|Fowl Pox Virus|/i| and inhibits the virus
ABSTRACT. Antiviral effects of a synthetic Aluminum-Magnesium Silicate (AMS) were tested on Fowl Pox Virus (FPV). Five batches of the Nigerian brand of FPV vaccine were used as sources of the virus. The reconstituted vaccines were mixed with The Synthetic AMS on equal volume to weight basis and incubated at room temperature for one hour. They were centrifuged for 10 minutes at 2000 revolutions per minute. The incubation and centrifugation were repeated on a portion of each vaccine supernatant. The two sets of supernatants were tested by the Modified Passive Haemagglutination test, for FPV titres. Portions of the vaccines, not incubated with the AMS, were served as controls. Fowl Pox Virus titres of the vaccines increased from a mean of 2.8 ± 1.10 to 11.2 ± 4.38 when incubated with the AMS once. When incubation with the AMS was repeated, the titres reduced (P< 0.05) to zero in each sample.. 1. INTRODUCTION. Fowl Pox Virus belongs to the Avipoxviridae viral family [1]. Hemagglutination Inhibition Test (HAI): Principle, procedure, result and interpretations - Microbeonline
Hemagglutination occurs when measles viruses and red blood cells are mixed (See image a). But, if the serum of a person infected with measles virus is mixed with RBC and measles virus, there wont be any agglutination of RBC. This phenomenon is known as hemagglutination inhibition. This arises because antibodies present in the serum of that infected person reacts with the measles viruses and neutralized them (positive result).. If the patients serum do not contain antibodies against surface proteins of test virus, there will be presence of hemagglutination as surface molecules are free to hemaaglutinate RBCs (negative result).. ...

Serological screening is an important step in influenza monitoring and vaccine development. The hemagglutination inhibition assay (HAI) is widely used to evaluate vaccine-induced antibody responses as well as to antigenically characterize influenza viruses. HAI results are based on an endpoint titration where the titers are typically manually interpreted and recorded by a trained expert. For serological applications, the lack of a standardized endpoint interpretation and presence of interfering non-specific inhibitors can translate into a high degree of variability in the results. This variability along with the lack of a digital record ...

The rate of positive results for hepatitis B surface antigen was 0.7% in our pregnant population; the higher prevalence in the first two years of screening may be explained by the selective inclusion of additional women from high risk groups already in the third trimester of pregnancy. The rate of 0.7% should not be extrapolated to the whole population. Urban hospitals with high prevalences (1.4-1.8%) serve a population that includes high risk ethnic groups, low socioeconomic groups, or both, whereas the Twente-Gelderse-Achterhoek region (prevalence 0.3%) serves a suburban, socioeconomically more privileged population.. The use of the haemagglutination assay in the Twente-Gelderse-Achterhoek region instead of a third generation assay (radioimmunoassay, ELISA) as in the other centres cannot account for the low prevalence of 0.3% in the region: the haemagglutination test detected all but two of a panel of 100 samples positive for the surface antigen according to the third generation test. The two ...

THE IMMUNE RESPONSE TO A HYBRID PROTEIN MOLECULE | JEM
Upon immunization with LDH-III (subunit composition AABB) rabbits produce anti-A and anti-B antibodies in comparable amounts. These antibodies fit equally well to the hybrid enzyme and to LDH-V (AAAA) or LDH-I (BBBB) respectively, as tested by passive hemagglutination inhibition. No antibodies reacting with both LDH-I and LDH-V were detected. A minority of hybrid-specific antibodies was, however, present in the sera.. Animals primed with LDH-III respond regularly to a boosting injection of LDH-V with the production of large amounts of anti-A (but not anti-B) antibodies. A similar injection of LDH-I stimulates (if it has any effect at all) the production of anti-B antibodies only. Stimulation with one of the pure types does not impair a subsequent response to the other.. The majority of the animals primed with LDH-III responded not at all or weakly to a boosting injection of LDH-I, though antibodies to LDH-I were present in the sera at the time of stimulation. This effect can hardly be explained ...

External Quality Assessment for the Determination of Diphtheria Antitoxin in Human Serum | Clinical and Vaccine Immunology
Accurate determination of diphtheria toxin antibodies is of value in determining the rates of immunity within broad populations or the immune status of individuals who may be at risk of infection, by assessing responses to vaccination and immunization schedule efficacy. Here we report the results of an external quality assessment (EQA) study for diphtheria serology, performed within the dedicated surveillance network DIPNET. Twelve national laboratories from 11 European countries participated by testing a standard panel of 150 sera using their current routine method: Vero cell neutralization test (NT), double-antigen enzyme-linked immunosorbent assay (ELISA; DAE), dual double-antigen time-resolved fluorescence immunoassay (dDA-DELFIA), passive hemagglutination assay (PHA), toxin binding inhibition assay (ToBI), and in-house or commercial ELISAs. The objective of the study was not to identify the best assay, as the advantages and drawbacks of methods used were known, but to verify if laboratories ...
Construction and comparison of different source neuraminidase candidate vaccine strains for human infection with Eurasian avian...
Human infections with Eurasian avian-like swine influenza H1N1 viruses have been reported in China in past years. One case resulted in death and others were mild case. In 2016, the World Health Organization recommended the use of A/Hunan/42443/2015(H1N1) virus to construct the first candidate vaccine strain for Eurasian avian-like swine influenza H1N1 viruses. Previous reports showed that the neuraminidase of A/Puerto Rico/8/34(H1N1) might improve the viral yield of reassortant viruses. Therefore, we constructed two reassortant candidate vaccine viruses of A/Hunan/42443/2015(H1N1) by reverse genetic technology, with (6+2) and (7+1) gene constitution, respectively. The (6+2) virus had hemagglutinin and neuraminidase from A/Hunan/42443/2015, and the (7+1) one had hemagglutinin from A/Hunan/42443/2015, while all the other genes were from A/Puerto Rico/8/34. Our data revealed that although the neuraminidase of the (7+1) virus was from high yield A/Puerto Rico/8/34, the hemagglutination titer and the ...

Antigen-binding receptors on T lymphocytes and IgG antibodies with the same antigen-binding specificity as the T-cell receptors display shared or identical idiotypes. This was shown using a system where adult F1 hybrid rats between two inbred strains were inoculated with T lymphocytes from one parental strain. Such F1 hybrid rats produce antibodies directed against idiotypic determinants present on IgG alloantibodies, produced in the T donor genotype strain and with specificity for the alloantigens of the other parental strain. The idiotypic nature of the F1 antialloantibody serum against the parental alloantibodies was demonstrated both by indirect hemagglutination tests or by gel diffusion using alloantisera with different specificity as targets. Furthermore, the F1 anti-T-lymphocyte sera could be shown to contain antibodies against idiotypic parental T lymphocytes as well. This was shown by the capacity of the antisera, in the presence of complement, to wipe out the relevant parental T-cell ...

A comparative study of the indirect haemagglutination (IHA), immunofluorescence (IFAT) and immunoenzymatic (ELISA) tests was carried out to determine the prevalence of Toxoplasma gondii antibodies in goats. One hundred seventy-four serum samples were obtained from four goat herds from the region of Uberlândia, State of Minas Gerais. The distribution of the animals, according to their origin, was as follow: 71 from herd I; 39 from herd II; 37 from herd III; and 27 from herd IV. Serum samples were analyzed by IHA, IFAT and ELISA, considering the reactivity of the serum samples at dilution = 1:64 as cut off titer for the three tests. A global seroprevalence of 18.4% was observed, with significantly higher positivity rate in the herd II (66.7%) and older animals (} 36 months). A high and significant positive correlation was found between the titers obtained by the IHA versus IFAT, IHA versus ELISA, and ELISA versus IFAT. Therefore, it can be concluded that the three analyzed tests have shown to be ...

ABO Tolerance after Treatment of Infant or Adult Mice with MHC-Matched A-Expressing Blood Cells - ATC Abstracts
Purpose: ABO-incompatible heart transplantation (ABOi HTx) is safe during infancy and allows increased donor access. Post-ABOi HTx B cell tolerance develops to donor blood group antigen(s) by mechanisms not fully defined. We developed A-transgenic mice (A-Tg) that express A-antigen on vascular endothelium and erythrocytes and demonstrated A-antigen specific tolerance induced by HTx into 4 wk-old, MHC-identical, wild-type (WT) mice. Herein, we explored intentional tolerance induction in infant and adult WT mice using A-Tg blood cells.. Methods: WT BALB/c mice were injected ip (weekly[times]3) with intact A-Tg BALB/c blood cells (±40Gy irradiated), at age 7 days (neonates) or 5 months (adults; see Table). Two weeks after treatment, all mice were injected ip (weekly[times]5) with human A-erythrocytes ('A-sensitized') in an attempt to elicit anti-A antibody (Ab) production. Serum anti-A and 3rd-party (non-A anti-human) Ab were assessed by hemagglutination assay.. Results: In response to ...

The automated haemagglutination assay using Treponema pallidum antigen (AMHA-TP) and the Venereal Disease Research Laboratory (VDRL) test were used to examine 330 163 sera. Reactive results were checked by the fluorescent treponemal antibody-absorption (FTA-ABS) test. When isolated reactivity or non-reactivity in the AMHA-TP test was investigated an estimated margin of error of 0.7% probably wrongly non-reactive and 0.008% presumably false non-reactive results were found. These figures were confirmed by randomised FTA-ABS tests on 504 sera with repeat AMHA-TP tests. The latter is therefore still the most reliable and practicable method for mass screening for syphilis.. ...

Hemagglutination Reactions Involving Plasmas and Erythrocytes of Indiv by Elery R. Becker
The writer (Becker, 1952) has recently reported on the varied responses among both chicks and ducklings to blood-induced infection with Plasmodium lophurae. The present paper is concerned with hemagglutination reactions involving the plasmas of individual chickens and duck erythrocytes, on the one hand, and the plasmas of individual ducks and chicken erythrocytes, on the other. The agglutinations observed were the result of exposing the erythrocytes to diluted plasmas containing natural or innate agglutinins; i.e., agglutinins not formed as the result of active immunization through exposure to the antigens of the erythrocytes.
