Our results demonstrate that the heat shock response is a potent inhibitor of RANTES gene expression in vitro. Induction of the heat shock response inhibited TNF-α-mediated expression of RANTES mRNA and secretion of immunoreactive RANTES. These effects were not secondary to cytotoxicity, and are consistent with previous data demonstrating that the heat shock response inhibited proinflammatory responses of lung cells (23, 24, 25, 26, 27). We propose that the observed inhibition is not a generalized effect of the heat shock response. Rather, it appears that the heat shock response has a relatively specific inhibitory effect on lung proinflammatory responses. Protein AMPylation is a conserved posttranslational modification with emerging roles in endoplasmic reticulum homeostasis. However, the range of substrates and cell biological consequences of AMPylation remain poorly defined. We expressed human and Caenorhabditis elegans AMPylation enzymes-huntingtin yeast-interacting protein E (HYPE) and filamentation-induced by cyclic AMP (FIC)-1, respectively-in Saccharomyces cerevisiae, a eukaryote that lacks endogenous protein AMPylation. Campylobacter jejuni is a leading cause of bacterial gastroenteritis in the developed world. The role of a homologue of the negative transcriptional regulatory protein HspR, which in other organisms participates in the control of the heat-shock response, was investigated. Following inactivation of hspR in C. jejuni, members of the HspR regulon were identified by DNA microarray transcript profiling. In agreement with the predicted role of HspR as a negative regulator of genes involved in the heat-shock response, it was observed that the transcript amounts of 13 genes were increased in the hspR mutant, including the chaperone genes dnaK, grpE and clpB, and a gene encoding the heat-shock regulator HrcA. Proteomic analysis also revealed increased synthesis of the heat-shock proteins DnaK, GrpE, GroEL and GroES in the absence of HspR. Loss of the alternative sigma factor σI results in a coordinate decrease in expression of all three toxin subunits. We have previously reported that low-intensity microwave exposure (0.75-1.0 GHz CW at 0.5 W; SAR 4-40 mW kg-1) can induce an apparently non-thermal heat-shock response in Caenorhabditis elegans worms carrying hsp16-1::reporter genes. This study provides a novel insight into the immediate and long-term cellular responses to elevated temperatures experienced in bone tissue during surgical cutting. The effects of heat-shock on bone cell viability and regeneration capacity are shown to be dependent on the degree of temperature elevation, the duration of exposure and the phenotype of the cell. The biological response of bone cells to elevated temperatures is characterized by an immediate necrotic response followed by later-stage apoptosis. Furthermore, is it shown that these biological responses are minimized if heat-shock is maintained below a critical temperature of less than equal to 47°C for 1 min. This study reports that osteoblast-like cells are more susceptible to heat-induced cellular death than osteocyte-like cells. The Hsp 20 genes are associated with stress caused by HS and other abiotic factors, but have recently been found to be associated with the response to biotic stresses. These genes represent the most abundant class among the HSPs in plants, but little is known about this gene family in soybean. Because of their apparent multifunctionality, these proteins are promising targets for developing crop varieties that are better adapted to biotic and abiotic stresses. Thus, in the present study an in silico identification of GmHsp 20 gene family members was performed, and the genes were characterized and subjected to in vivo expression analysis under biotic and abiotic stresses. A search of the available soybean genome databases revealed 51 gene models as potential GmHsp 20 candidates. The 51 GmHsp 20 genes were distributed across a total of 15 subfamilies where a specific predicted secondary structure was identified. Based on in vivo analysis, only 47 soybean Hsp 20 genes were responsive to heat shock stress.

Natural populations are constantly exposed to changing environments and genetic threats. The Hsps buffer this environmental variation and are therefore important factors for the maintenance of homeostasis across environmental regimes. Given the fact that Hsps influence fitness under non-optimal environmental conditions, we argue that the regulation and expression levels of these proteins are of major evolutionary and ecological importance. Here, we have reviewed the recent evidence on the role of Hsp expression from an ecological perspective, including a characterization of factors that induce the heat shock response and a discussion of the costs associated with this response. We suggest that up-regulation of Hsps in addition to being an important part of the response to sudden extreme stress exposures is of ecological relevance on a much wider scale with respect to less severe but regular incidences of stress.
