TY - JOUR. T1 - Teratogenicity of bifenox and nitrofen in rodents. AU - Francis, B. Magnus. PY - 1986/1/1. Y1 - 1986/1/1. N2 - The teratogenicity of the diphenyl ether herbicide bifenox [2,4-dichlorophenyl 3-carboxymethyl-4-nitrophenyl ether] was compared to that of nitrofen [2,4-dichlorophenyl 4-nitrophenyl ether] in rats and in mice. Neither compound increased prenatal mortality in mice. Because nitrofen causes both malformations that are compatible with survival to weaning and a high incidence of perinatal (but not of fetal) mortality, emphasis was placed on postnatal parameters of bifenox toxicity. In rats, bifenox caused a low incidence of bloody tears, but it did not decrease survival to term or to weaning in rats or mice, and did not reduce Harderian gland weight in mice. Because the weight of the Harderian glands is a more objective measure of their status than is the presence of an eye discharge, it is concluded that bifenox is not teratogenic at the levels administered. Nitrofen ...
4,4-Diaminodiphenyl ether was tested in mice and rats by oral administration and by subcutaneous injection. In two studies in rats, it produced benign and malignant liver-cell tumours following oral or subcutaneous administration; and following its oral administration in one study, benign and malignant follicular-cell tumours of the thyroid were produced. Other studies in rats by oral and subcutaneous administration were not adequate for evaluation. In a study in mice by oral administration it produced benign and malignant liver-cell tumours in females given the high dose and in males given the low dose and adenomas of the Harderian gland in animals of both sexes.. 4,4-Diaminodiphenyl ether is mutagenic to Salmonella typhimurium with metabolic activation.. No data were available to assess its teratogenicity to experimental animals. ...

We consider parabolic operators of the form $$\partial_t+\mathcal{L},\ \mathcal{L}:=-\mbox{div}\, A(X,t)\nabla,$$ in$\mathbb R_+^{n+2}:=\{(X,t)=(x,x_{n+1},t)\in \mathbb R^{n}\times \mathbb R\times \mathbb R:\ x_{n+1},0\}$, $n\geq 1$. We assume that $A$ is an $(n+1)\times (n+1)$-dimensional matrix which is bounded, measurable, uniformly elliptic and complex, and we assume, in addition, that the entries of A are independent of the spatial coordinate $x_{n+1}$ as well as of the time coordinate $t$. We prove that the boundedness of associated single layer potentials, with data in $L^2$, can be reduced to two crucial estimates (Theorem \ref{th0}), one being a square function estimate involving the single layer potential. By establishing a local parabolic Tb-theorem for square functions we are then able to verify the two crucial estimates in the case of real, symmetric operators (Theorem \ref{th2}). Our results are crucial when addressing the solvability of the classical Dirichlet, Neumann and ...

Purpose: To establish a rabbit model for keratoconjunctivitis sicca (KCS) to study autologous submandibular gland transfer for treating severe KCS.. Methods: In 2 groups of 10 rabbits, left eyes were operated and right eyes were controls. In the trichloroacetic acid-treated group, the lacrimal and harderian glands and nictitating membrane were removed surgically; the palpebral and bulbar conjunctiva were swabbed with 50% trichloroacetic acid. In the non-trichloroacetic acid-treated group, the lacrimal and harderian glands and nictitating membrane were surgically removed. The Schirmer test was performed preoperatively and 1, 2, 3, and 4 months postoperatively. Corneal densities of rose bengal and fluorescein staining were scored every month postoperatively. At 4 months, the cornea and bulbar conjunctiva were removed from operated and control eyes for histopathology. The upper bulbar conjunctiva was used to determine goblet cell density.. Results: Compared with preoperative conditions, tear ...

Carcinogenesis: Lifetime carcinogenicity bioassays were conducted in mice and rats. Mice were given daily oral doses of zoledronic acid of 0.1, 0.5, or 2 mg/kg/day for 2 years. There was an increased incidence of Harderian gland adenomas in males and females in all treatment groups (starting at doses equivalent to 0.002 times the human 5 mg intravenous dose, based on body surface area, mg/m2). Rats were given daily oral doses of zoledronic acid of 0.1, 0.5, or 2 mg/kg/day for 2 years. No increased incidence of tumors was observed at any dose (up to 0.1 times the human intravenous dose of 5 mg, based on body surface area, mg/m2).. Mutagenesis: Zoledronic acid was not genotoxic in the Ames bacterial mutagenicity assay, in the Chinese hamster ovary cell assay, or in the Chinese hamster gene mutation assay, with or without metabolic activation. Zoledronic acid was not genotoxic in the in vivo rat micronucleus assay.. Impairment of Fertility: Female rats were given daily subcutaneous doses of ...
The current study demonstrates the presence of megalin and cubilin in the lacrimal and Harderian glands, and provides evidence that these proteins may be involved in the secretion scheme of vitamin D into tear fluid. A recent study by Alsalem et al.2 confirmed previous results from our group demonstrating the presence of the metabolic machinery to process vitamin D and its metabolites within cells of the cornea, and importantly it also demonstrated that megalin and cubilin are present in several ocular barrier cell types. Megalin has also been detected in RPE and nonpigmented ciliary body epithelium, and a severe myopia phenotype as well as enlarged RPE melanosomes and abnormal ciliary body development were described in megalin-deficient mice.5 Previous work examining renal vitamin D uptake in megalin knockout mice demonstrated an increased urine concentration of 25(OH)D3 and decreased plasma levels of 25(OH)D3 and 1α,25(OH)D3, both resulting from decreased cellular uptake of the vitamin ...

Rats infected with rat coronavirus begin to show symptoms as early as 5 days, with respiratory involvement and cervical swelling by 7-8 days. The infection often presents as a characteristic inflammation of submaxillary and parotid salivary glands, which can result in tissue necrosis. Cervical lymph nodes and/or the harderian and intraorbital lacrimal glands behind the eyes may also be affected. As a result, bulging eyes, ocular lesions, facial swelling, lacerations, porphyrin discharge, bleeding or squinting due to photosensitivity may be observed. Sometimes symptoms may give the appearance of a shortened or swollen neck. In extreme cases, eyesight can be lost or compromised permanently due to the infection or from self-mutilation caused by scratching. Eye bulging may take several weeks to subside due to retro-orbital swelling ...
Please read the general principles page before attempting any blood sampling procedure. Technique Summary Resources and references Retro-orbital technique in other animals All blood sampling techniques in the mouse Technique Also referred to as peri-orbital, posterior-orbital and orbital venous sinus bleeding. Retro-orbital bleeding should be performed as a terminal procedure. It should only be used with recovery in rare circumstances with exceptional scientific justification (e.g. where a large blood volume is necessary or where peripheral veins are used for dosing), because of its potential impact on animal welfare. Adverse effects reported for this procedure are described in the summary below. Where its use is unavoidable, retro-orbital bleeding should only be used under terminal or general anaesthesia. Because of the severity of the adverse effects that can occur with this technique, even in skilled hands, it is essential that it is conducted only by staff members competent (practised) in the
Immortal unipotent porcine PICM-19H and PICM-19B stem cell lines - Two cell lines, PICM-19H and PICM-19B, were derived from the bipotent ARS-PICM-19 pig liver stem cell line and assessed for their potential application in artificial liver devices. The study included assessments of growth rate and cell density in culture, morphological features, and hepatocyte detoxification functions, i.e., inducible CYP450 activity, ammonia clearance, and urea production. The PICM-19H cells contain numerous mitochondria, Golgi apparatus, smooth and rough endoplasmic reticulum, vesicular bodies and occasional lipid vacuoles. PICM-19H cells display inducible CYP450 activity, clear ammonia, and produce urea in a glutamine-free medium. Ultrastructural analysis of the PICM-19B monolayers show that the roughly cuboidal cells display basal-apical polarization and are joined by tight junction-like complexes. Other ultrastructure features are similar to those of PICM-19H cells except that they possess numerous cell ...
Morphological, ultrastructural, and molecular- sequence data were used to assess the phylogenetic position of a tetraflagellate green alga isolated from soil samples of a saline dry basin near Fderick, Mauritania. This alga can grow as individual cells or form non- coenobial colonies of up to 12 individuals. It has a parietal chloroplast with an embedded pyrenoid covered by a starch sheath and traversed by single parallel thylakoids, and an eyespot located in a parietal position opposite to the flagellar insertion. Lipid vacuoles are present in the cytoplasm. Microspectroscopy indicated the presence of chlorophylls a and b, with lutein as the major carotenoid in the chloroplast, while the eyespot spectrum has a shape typical of green-algal eyespots. The cell has four flagella, two of them long and two considerably shorter. Sequence data from the 18S rRNA gene and ITS2 were obtained and compared with published sequences for green algae. Results from morphological and ultrastructural examinations ...

We compared antioxidant responses and regulation of porphyrin metabolism in rice plants treated with oxyfluorfen (OF) or methyl viologen (MV). Plants treated with MV exhibited not only greater increases in conductivity and malondialdehyde but also a greater decline in Fv/Fm, compared to plants treated with OF. MV-treated plants had greater increases in activities of superoxide dismutase (SOD) and catalase (CAT) as well as transcript levels of SODA and CATA than OF-treated plants after 28 h of the treatments, whereas increases in ascorbate peroxidase (APX) activity and transcript levels of APXA and APXB were greater in OF-treated plants. Both OF- and MV-treated plants resulted in not only down-regulation of most genes involved in porphyrin biosynthesis but also disappearance of Mg-porphyrins during the late stage of photooxidative stress. By contrast, up-regulation of heme oxygenase 2 (HO2) is possibly part of an efficient antioxidant response to compensate photooxidative damage in both treatments. Our
The thyroid gland is a highly vascular organ, situated at the front and sides of the neck; it consists of right and left lobes connected across the mid line by a narrow portion, the isthmus. Its weight is somewhat variable, but is usually about 30 grams. It is slightly heavier in the female, in whom it becomes enlarged during menstruation and pregnancy. The lobes are conical in shape, the apex of each being directed superior and lateral as far as the junction of the middle with the lower third of the thyroid cartilage; the base looks inferior, and is on a level with the fifth or sixth tracheal ring. Each lobe is about 5 cm. long; its greatest width is about 3 cm and its thickness about 2 cm. The lateral or superficial surface is convex, and covered by the skin, the superficial and deep fasciae, the Sternocleidomastoid, the superior belly of the Omohyoid, the Sternohyoid and Sternothyreoid, and beneath the last muscle by the pretracheal layer of the deep fascia, which forms a capsule for the ...
Dry eye is a common chronic disease that affects both humans and dogs and causes progressive damage to the cornea leading to corneal scars, corneal vascularization, corneal pigmentation, pain, and ultimately vision loss. Causes of dry eye include immune-mediated, congenital, age-related, post inflammatory, and drug-induced.. Treatments for dry eye include various topical medications such as tear supplements, steroids, cyclosporine or tacrolimus (immunomodulatory drugs), as well as surgical efforts such as parotid duct transposition. In order to maintain vision and comfort, dogs with dry eye are often managed with one or more of these topical medications twice daily lifelong.. While it may seem that there are many options for dry eye treatment, the majority of these topical medications are aimed at the immune-mediated component of dry eye and can be ineffective for dry eye caused by other etiologies. In addition, currently there is only one FDA approved product (0.2 % cyclosporine) on the market ...

The present study has examined the ability of Akt1 and its oncogenic homologue, v-akt, to activate ECM invasion in mouse mammary epithelial cells. To achieve this objective, we developed a cell line that is amenable to transduction by one or more recombinant avian retroviruses (37) . This system is based on the introduction of the ALV subtype A receptor, tv-a, into mammalian cells making them permissive to infection by recombinant ALV (38) . The mouse mammary epithelial cell line, COMMA-1D, was used for this retrovirus delivery system because these cells have been shown to differentiate in vitro and to give rise to normal gland structures in the cleared fat pad of the BALB/c mouse (39 , 40) . Although these immortalized mammary epithelial cells contain one mutated p53 allele (41) , they do not exhibit a transformed phenotype either in vitro or in vivo (39 , 40) .. Transduction of COMMA-1D/tv-a cells with Akt1 or v-akt led to an interesting dichotomy between their ability to induce transformation ...

Lung cancer is the most common non-cutaneous cancer in the western Countries; in the United States, near to 30% of the cancer deaths are due to carcinoma of the lung. The age-adjusted cancer death rate is close to 30 × 105 women, and 75 × 105 men. In 1998 it has been estimated that there will be 67,000 lung cancer deaths among women and 93.000 deaths among men in the United States. In this Country, whilst the incidence in men shows a decline commencing in 1990s, since 1970 there has been a sharp increase in women. In 1987 lung cancer surpassed breast cancer as the greatest cause of female cancer death.

TY - JOUR. T1 - Overexpression of ras in mucus-secreting human colon carcinoma cells of low tumorigenicity. AU - Augenlicht, Leonard H.. AU - Augeron, C.. AU - Yander, G.. AU - Laboisse, C.. PY - 1987. Y1 - 1987. N2 - We have investigated the expression of the protooncogenes of the myc and ras family in HT29 cells and in three different clonal cell lines derived from this colon carcinoma cell line. In contrast to the decrease in myc expression seen when leukemia cells are induced to differentiate, we have found no changes in expression of the myc gene family in differentiated colon carcinoma cells. However, a greater than 5-fold increase in expression of sequences which hybridize to Ha-ras was observed in cells which secrete mucin, with a smaller increase seen in expression of Ki-ras in the same cells. This increase was not seen in cells which exhibit vectorial transport of water and ions, and which are not mucus-secreting. All differentiated lines were less tumorigenic in nude mice than the ...

Service of the PI3K/AKT signal pathway is a known driving force for the progression to castration-recurrent prostate cancer (CR-CaP), which constitutes the major lethal phenotype of CaP. RUNX2 binding to the PIP promoter is increased in FOXO4-KD cells. Indeed, the forced expression of FOXO4 reversed the increased invasiveness of LNCaP/shFOXO4 Rabbit Polyclonal to DECR2 cells; the forced expression of FOXO4 did not alter RUNX2 protein levels, yet it decreased RUNX2 binding to the PIP promoter, resulting in PIP downregulation. Finally, there was a correlation between FOXO4, but not FOXO1 or FOXO3, downregulation and decreased metastasis-free survival in human CaP patients. Our data strongly recommend that improved PI3E/AKT-mediated metastatic invasiveness in Cover can be connected with FOXO4 reduction, and that systems to induce FOXO4 re-expression might suppress Cover metastatic aggressiveness. Intro Prostate tumor (Cover) continues to be the most diagnosed non-cutaneous tumor and the second ...

This video tutorial will show you the process of drawing blood from rats through the saphenous vein and by cardiac puncture. Drawing blood from rodents is necessary for a large number of both in vitro and in vivo studies. Sites of blood draws are numerous in rodents: retro-orbital sinus, jugular vein, maxillary vein, saphenous vein, heart. Each technique has its advantages and disadvantages, and some are not approved any more in some countries (e.g. retro-orbital draws in Holland). Here we present two techniques for drawing blood from rats, each with its specific applications. Blood draw

Prostatic adenocarcinoma (PCa) is the most frequently diagnosed non-cutaneous malignancy and second leading cause of cancer death in men in the US. Although organ-confined disease is manageable, treatment options for disseminated PCa are limited. First-line therapy for metastatic PCa targets the androgen receptor (AR) via androgen deprivation therapy (ADT); however, tumors often recur, displaying reactivation of AR signaling despite continued therapeutic targeting. There is currently no durable treatment for this advanced disease stage, termed castrate-resistant PCa (CRPC). While most localized PCa express wild-type p53, recent high-profile, genome-wide studies identified increased TP53 gene mutation rates in advanced disease (specifically CRPC). Using these data, we noted that the patient-derived, CRPC-specific p53 mutations observed are relatively unique to prostate cancer. These mutations occur in hotspot clusters altered in other tumor types, but represent distinct amino acid changes ...
All normal people have to some extent both male and female hormones. male chest occurs when male hormones (androgens) are relatively weak compared to female hormones (estrogen) present in the body. This may occur temporarily during normal development of boys and may result in gynecomastia in infants or during puberty. In these cases gyneco usually disappears automatically when hormone levels return to normal.. In other cases, an illness or medical treatment can create a hormonal environment in the body that allows the development of male gland.. Here are some examples of conditions that may be associated with gynecomastia: ...
The question of difference - between women and men and among women - is at the heart of feminist theory and the history of feminism. Feminists have long debated the meanings of sexual difference: is it an underlying truth of nature or the result of changing social belief? Are women the same as or different from men? Feminism and History argues that sexual difference, indeed that all forms of social differentation, cannot be understood apart from history. It brings together the best critical articles available to analyze the ways in which differences among women and men have been produced. The articles range across many countries and time periods (from the Middle Ages to the present) and they include analyses of western and non-western experiences. There are discussions of race in the United States and in colonial contexts. A variety of theoretical approaches to the question of difference is included; but in all cases, difference is the focus of the historians analysis. The analytic focus on difference

A Large-size engineering single layer fuel cell (SLFC) consisting of a nano-structured Li0.4Mg0.3Zn0.3O2-delta/Ce0.8Sm0.2O2-delta (LMZSDC) composite with an active area of 25 cm(2) (6 cm x 6 cm x 0.1 cm) is successfully fabricated. The SLFC is evaluated by testing the cell durability with a time-dependent degradation using an H-2 fuel and an air oxidant at 600 degrees C for over 120 h. A maximum power of 12.8 W (512 mW cm(-2)) is achieved at 600 degrees C. In the initial operation stage around 50 h, the cells performance decreases from 12.8 to 11.2 W; however, after this point, the performance was consistently stable, and no significant degradation is observed in the current density or the cell performance. The device performed excellently at low temperatures with a delivered power output of more than 250 mW cm(-2) at a temperature as low as 400 degrees C. By curve fitting the X-ray photoelectron spectroscopy (XPS) results, the ratio of Ce3+/(Ce3++Ce4+) before and after the long-time operation ...

Here is the molecular structure of erythromycin. Like many polyketides it is circular, which at first appears to be a bit of a headache to synthesise. The way its put together, though, is actually very clever. The backbone of the circular section is made up first, using a system of modular enzymes that pass the growing chain along like a conveyor belt, adding new residues at each stage. Then the straight chain is curled up into a ring, and finally the two side residues (the ones on the bottom right of the chemical structure, that look a bit like squashed rectangles with dents in them) are stuck on ...

As some of you know, I have been yammering for ages about hosting an aldehydic sample pass- and here it finally is! Most, more likely all, perfumes contain aldehydes but not all are aldehydics. I hope this pass will demonstrate the difference. Aldehydes provide lift and sparkle to a scent; without them, perfumes tend to sit flat and heavy on the skin. When we refer to scents that are aldehydic, we specifically mean those containing straight chain aliphatic aldehydes, typically C10-C12. No.5 is

At the heart of the transgender revolution is the notion that psychological identity trumps bodily identity. In this way of thinking, a person is whatever they think themselves to be. If a girl perceives herself to be a boy, then she is one even if her biology says otherwise. If a boy perceives himself to be a girl, then he is one even if his biology says otherwise. Gender is self-determined, not determined by the sexual differences that the Creator has embedded into every cell of our bodies.. Have Americans thought through the implications of believing that ones psychological identity should trump ones biological identity when the two seem to be out of sync? I am doubtful that we have..
