In taxonomy, Haloarcula is a genus of the Halobacteriaceae. Haloarcula species are extreme halophilic archaea. They are distinguished from other genera in the Halobacteriaceae family by the presence of specific derivatives of TGD-2 polar lipids. Currently, seven recognized species are in the genus: H. vallismortis, H. marismortui, H. hispanica, H. japonica, H. argentinensis, H. mukohataei, and H. quadrata. H. quadrata was first isolated when researchers were attempting to culture Haloquadratum walsbyi, a haloarchaeon that was thought to be unculturable until 2004. H. quadrata has predominantly flat, square-shaped, somewhat pleomorphic cells. Haloarcula species grow optimally at 40-45 °C. Growth appears in sheets of up to 65 cells often in the shape of a square or triangle. Haloarcula species are found in neutral saline environments such as salt lakes, marine salterns, and saline soils. Homologous recombination plays a central role in the repair of double-strand DNA breaks, the restart of stalled replication forks and the generation of genetic diversity. Regulation of recombination is essential since defects can lead to genome instability and chromosomal rearrangements. Strand exchange is a key step of recombination - it is catalysed by RecA in bacteria, Rad51/Dmc1 in eukaryotes and RadA in archaea. RadB, a paralogue of RadA, is present in many archaeal species. RadB has previously been proposed to function as a recombination mediator, assisting in RadA-mediated strand exchange. In this study, we use the archaeon Haloferax volcanii to provide evidence to support this hypothesis. We show that RadB is required for efficient recombination and survival following treatment with DNA-damaging agents, and we identify two point mutations in radA that suppress the ΔradB phenotype. The motile, predominantly square-shaped, red archaeon strain 801030/1T, isolated from a brine pool in the Sinai peninsula (Egypt), was characterized taxonomically. On the basis of its polar lipid composition, the nucleotide sequences of its two 16S rRNA genes, the DNA G+C content (60·1 mol%) and in growth characteristics, the isolate could be assigned to the genus Haloarcula. However, phylogenetic analysis of the two 16S rRNA genes detected in this organism and low DNA-DNA hybridization values with related Haloarcula species showed that strain 801030/1T is sufficiently different from the recognized Haloarcula species to warrant its designation as a new species. A new species, Haloarcula quadrata, is therefore proposed, with strain 801030/1T (= DSM 11927T) as the type strain.
Biotechnologically produced 2,3-butanediol (2,3-BDO) is a potential starting material for industrial bulk chemicals such as butadiene or methyl ethyl ketone which are currently produced from fossil feedstocks. Honesty compels me to add that I was a member of the committee that wrote the current (1983) IUPAC-IUB Recommendations on Nomenclature and Symbolism for Amino Acids and Peptides, in which you will find, in Table 1, the words The systematic names and formulas given refer to hypothetical forms in which amino groups are unprotonated and carboxyl groups are undissociated. This convention is useful to avoid various nomenclatural problems but should not be taken to imply that these structures represent an appreciable fraction of the amino-acid molecules. I tried but failed to convince the rest of the committee that hypothetical was a euphemism that should be replaced by fictitious or nonexistent. After more than 30 years its hard to be certain, but I think the second sentence had not originally been in the draft but was added in an effort to keep me quiet.
