Research has shown that the rate of habituation is inversely related to the amount of food consumed and slower habituation may be a factor that is relevant to obesity, as overweight youth and adults habituate slower and consume more energy than their peers. In patients with Crohns disease (CD) stress is believed to increase the incidence of disease relapse. The brain processes stressful stimuli and triggers the stress-evoked responses. Habituation to stress is an adaptive process that allows minimizing these responses. We hypothesized inadequate habituation to stress in CD patients. The aim of this study was to compare the neural habituation between CD patients and controls. Twenty CD patients and eighteen controls underwent a functional magnetic resonance imaging while performing two repeated runs of a stress-evoking task. The task elicited different neural activity between the groups across runs in (1) amygdala, hippocampus, (2) insula, putamen (3) cerebellar regions, suggesting altered habituation to stress in patients. The insect mushroom bodies are higher-order brain centers and critical for odor learning. We investigated experience dependent plasticity of their intrinsic neurons, the Kenyon cells (KCs). Using calcium imaging, we recorded KC responses and investigated non-associative plasticity by applying repeated odor stimuli. Associative plasticity was examined by performing appetitive odor learning experiments. Olfactory, gustatory and tactile antennal stimuli evoked phasic calcium transients in sparse ensembles of responding KCs. Repeated stimulation with an odor led to a decrease in KCs response strength. The pairing of an odor (conditioned stimulus, CS) with a sucrose reward (unconditioned stimulus) induced a prolongation of KC responses. After conditioning, KC responses to a rewarded odor (CS+) recovered from repetition-induced decrease, while the responses to a non-rewarded odor (CS−) decreased further. Non-associative memory contributes to filtering extraneous information from the environment. Additionally, behavioral state alterations such as stress have long been implicated in memory. Here, I determined how stress, through evoked changes in the noradrenergic system, affects early processing of sensory information during non-associative tasks. In chapter 1, I show that behavioral habituation to odor stimuli is correlated with adaption of neural responses in mitral/tufted (M/T) neurons of the main olfactory bulb (MOB) in anesthetized rats. I further show that both behavioral and neural habituation require functioning NMDA receptors in the MOB. This suggests a likely odor experience-dependent plasticity that occurs in the MOB. In chapter 2, I show that noradrenergic manipulations affect odor memory duration.
