The present study was first aimed at a complete steady-state kinetic analysis of the reaction between guaiacol (2-methoxyphenol) and the myeloperoxidase (MPO)/H2O2 system, including a description of the isolation and purification of MPO from human polymorphonuclear neutrophil cells. Secondly, the overall reaction of the oxidation of NADPH, mediated by the reactive intermediates formed from the oxidation of guaiacol in the MPO/H2O2 system, was analysed kinetically. The presence of guaiacol stimulates the oxidation of NADPH by the MPO/H2O2 system in a concentration-dependent manner. Concomitantly, the accumulation of biphenoquinone (BQ), the final steady-state product of guaiacol oxidation, is lowered, and even inhibited completely, at high concentrations of NADPH. Adult polyglucosan body disease (APBD) is a late-onset disease caused by intracellular accumulation of polyglucosan bodies, formed due to glycogen-branching enzyme (GBE) deficiency. To find a treatment for APBD, we screened 1,700 FDA-approved compounds in fibroblasts derived from APBD-modeling GBE1-knockin mice. Capitalizing on fluorescent periodic acid-Schiff reagent, which interacts with polyglucosans in the cell, this screen discovered that the flavoring agent guaiacol can lower polyglucosans, a result also confirmed in APBD patient fibroblasts. Biochemical assays showed that guaiacol lowers basal and glucose 6-phosphate-stimulated glycogen synthase (GYS) activity. In this study an analytical assay for urinary methoxyphenols was developed as a potential biological marker of environmental wood smoke exposures. The assay was aimed at 11 marker methoxyphenols, which were guaiacol, methylguaiacol, ethyguaiacol, propylguaiacol, vanillin, eugenol, trans-isoeugenol, syringol, methylguaiacol, ethylguaiacol, and syringaldehyde. Methoxyphenol conjugates 10 ml urine samples were hydrolyzed in sulfuric acid solution and then loaded onto XAD-2 SPE cartridges. After separation from the urine matrix, analytes were eluted with ethyl acetate and quantified by GC-MS, using an isotope-dilution assay. The detection limits for methoxyphenols were approximately 8ng/ml in urine for most of the marker phenols. Malodourous compounds, including volatile phenols (VPs) are frequently found at concentrations below their odour thresholds in wine, and may therefore be considered to present no threat to wine quality. Most investigations into smoke taint quantify compounds by chemical/analytical means, or investigate sensory effects of supra- and peri-threshold contamination in model wine. In this project, twelve wines submitted by the South African industry as faulty and/or smoke tainted were screened for VPs using GC-MS, and characterized using Descriptive Analysis (DA) by a sensory panel highly trained in smoke taint evaluation. Results were compared statistically to elucidate relationships between chemical and sensory characteristics.
