The function of quaternary structure, i.e. the arrangement of multiple subunits into an oligomer, may be to allow for cooperative effects, formation of novel active sites, provide additional stability, increase solubility or decrease osmotic pressure [2]. Am I the only one who had cPn test for IgG as positive (and HIGH ,1:512) for years, to suddenly after chemo begin to test negative for cPn IgG from now on?. Got second test done (ie second opinion) by same local Labcorp that has done my cPn tests in the past which had indicated positive in the past, and new test confirms negative results from my primary care physicians test.... I assume this means theres no cPn bacteria or antibodies in me anymore, including any cryptic state cPn hanging around - and thus, I no longer need to worry about cPn re-surfacing, and can stop CAP all together... and dont have to worry about any more Multiple Sclerosis relapses happening either. In Mycobacterium tuberculosis there are 2 distinct groEL homologues which encode the chaperonins Cpn60.1 and Cpn60.2, with the latter predicted to be the main house-keeping chaperonin. Phylogenetic analysis has revealed that the genes for the duplicated chaperonins diverged a long time ago. This implies that the duplicated chaperonins have evolved for different cellular functions. Interestingly, while most chaperonins occur as stable large complexes with a characteristic double ring structure of 14 subunits, the Mycobacterial chaperonins are very unstable and appear to form much smaller complexes. Given that the large structures formed by most chaperonins are vital to their mechanism of action, it is unclear why the oligomers are so much less stable in Mycobacteria. It was extremely tricky getting diagnosed with CPn in Australia a few years back.Australian Biologics mixed up my sample, and had to test 3 times (telling me a different result each time). Initially they told me my PCR test was negative. They thought I was so devoid of harmful microbes that they used my sample as a control when testing someone else. As a control my sample tested strongly positive to an array of nasties (even without the addition of an amplification agent). To their credit they called me back to get a fresh sample and retest. The retested with an amplification agent...and I again tested negative to CPn and other nasties.I had symptoms strongly suggestive of a CPn infection, so did the NAC test. The response was strong and jugular-centric (I felt them fill and stay under pressure for days). I obviously had a lot of Elementary Bodies [so I would have an immune response I guess??].
