The role of cyclic AMP in the octopaminergic modulation of the dorsal longitudinal flight muscles of the locust Schistocerca gregaria has been investigated. Several techniques have been used to elevate cyclic AMP levels in this tissue by mechanisms that bypass the receptor activation stage. These include the use of phosphodiesterase inhibitors to block the metabolism of cyclic nucleotides, the use of forskolin, the diterpene activator of adenylate cyclase, and the direct application of permeable and phosphodiesterase-resistant analogues of cyclic AMP. All these approaches can be shown to mimic the modulatory effects of octopamine on the flight muscle. Surprisingly, the phosphodiesterase inhibitors used were not able to potentiate the actions of octopamine on this preparation. In this work, the distribution, chemical composition and water permeability of the cuticular lipids of the African migratory locust, Locusta migratoria migratoriodes and the desert locust, Schistocerca gregaria have been investigated. Scanning electron microscopy of the wings and terga of the two species showed that the epicuticular lipid formed a liquid coating over the surface. The lipid layer on the fore wing of L.migratoria migratoriodes was thinner and more crystalline than that of S.gregaria. The tergum of L.migratoria migratoriodes had a smooth epicuticular lipid layer while that of S.gregaria had a lipid layer with amorphous lipid masses. Transmission electron microscopy revealed that the wing and tergal cuticle of both species conformed to the basic structural plan of insect cuticle, namely an outer, thin epicuticle and an inner, thick procuticle divided into an outer exocuticle and an inner endocuticle, overlying a single layer of cells, the hypodermis. Cytochrome P450 superfamily proteins play important roles in detoxification of xenobiotics and during physiological and developmental processes. To contribute to our understanding of this large gene family in insects, we have investigated the function of the cytochrome P450 gene LmCYP4G102 in the migratory locust Locusta migratoria. Suppression of LmCYP4G102 expression by RNA interference (RNAi) does not interfere with moulting but causes rapid loss of body weight - probably due to massive loss of water, and death soon after moulting. Accordingly, maintaining these animals at 90% relative humidity prevented lethality. Consistently, RNAi against LmCYP4G102 provoked a decrease in the content of cuticular alkanes, which as an important fraction of cuticular hydrocarbons have been shown to confer desiccation resistance. The whole complement of synaptonemal complexes (SCs) and the axis of the X chromosome have been described and measured. The relative length of each SC is characteristic and constant and permits the construction of an idiogram. Relative lengths of SCs are almost equal to the relative lengths of mitotic chromosomes of spermatogonia (with the exception of the X chromosome), thus extending to an invertebrate the relationship between SCs and mitotic chromosomes that has been demonstrated in mammals. This is the case of Ronderosia bergii, in which further large pericentromeric inversion in the neo-Y also took place, making this species particularly interesting for investigating sex chromosome evolution. Here, we characterized the satellite DNAs (satDNAs) and transposable elements (TEs) of the species to investigate the quantitative differences in repeat composition between male and female genomes putatively associated with sex chromosomes. Madagascar has a long history of using Orthoptera as food and feed. Our understanding of the biological diversity of this resource, its contemporary use, and its future potentials in Madagascar is extremely limited. The present study contributes basic knowledge of the biological diversity and local uses of edible Orthoptera in Malagasy food cultures. Data was collected with key informants in 47 localities covering most of the ecoregions of Madagascar and corresponding to 12 of the 19 ethnic groups. Orthoptera are consumed throughout Madagascar. We report 37 edible Orthoptera species, of which 28 are new species records of edible Orthoptera in Madagascar and 24 are new species records of edible Orthoptera in the world. Most species are endemic and occur in farming zones. Children are the primary collectors and consumers of edible Orthoptera. The insects are eaten both as snacks and main meals. In all species songs of males from the Central Asiatic and European populations are almost identical in spite of the fact, that in some species localities in Kyrgyzstan are isolated from the main parts of their ranges by the deserts of the South Kazakhstan and Central Asia. Glyptobothrus maritimus is the only species from G. biguttulus group widely distributed over all the territory of Kyrgyzstan. Retrograde tracing of the nervi corporis cardiaci II (NCC II) in vitro with Lucifer yellow in combination with LomTK immunocytochemistry revealed that about half of the secretomotor neurons in the lateral part of the protocerebrum projecting into the glandular lobe of the corpora cardiaca (CCG) contain LomTK-immunoreactive material. Since the four LomTKs are present in the CCG, these four or five neurons in each hemisphere are likely to contain colocalized LomTK I-IV. Escherichia coli K1 infection is a major cause of neonatal meningitis, with high rates of mortality and disability. Despite years of research, only a small number of factors contributing to E. coli K1 virulence have been identified. The Tat (twin-arginine translocation) protein export system is found in the cytoplasmic membrane of E. coli and is involved in the transport of folded proteins. In vivo and ex vivo models using the African migratory locust, Locusta migratoria, were employed to explore the role of Tat pathway in E. coli K1 virulence using tat-deletion mutants. Groups of locusts were infected and mortality was recorded at 24-h intervals. The findings revealed that ?tatA, Delta tatAC and ?tat produced levels of mortality similar to wild-type E. coli K1, with |78% mortality recorded within 72 h. Laughter heals. Fortunately Im married to a very funny man! Throughout our marriage we have avoided and resolved many things through laughter. Life can be serious but it can also be quite funny - sometimes at the same time. I had a visit to the e.r. Monday morning with pain which resolved itself. While going through something terrible and being between two thin curtains I laughed at DHs comments and expressions related to the two people on opposite sides of me who really had created their own problems. (I also said a prayer for them) I thought it peculiar that I could laugh at a time like that.
