Proper gonad development is essential for successful reproduction. Gonad development has been studied but the mechanisms involved are not fully understood. I used C. elegans as a model to study how the gonad is formed, by disrupting genes that may be involved. One of those genes is cdk-1, a cyclin-dependent kinase that is essential for cell division, because cdk-1 is required for the initiation of gonad development. How does cdk-1 control the initiation of gonad development? What other proteins does CDK-1 interact with? Cyclin-dependent kinases require cyclins to function and CDK-1 is known to interact with several cyclins. I sought to determine which of these cyclins CDK-1 interacts with to control initiation of gonad development by knocking down each cyclin by RNAi and looking for cdk-1 absence of gonads phenotype. I will also look at cdk-1 inhibitors and targets and shed some light on the control of reproductive development.
In most organisms, primordial germ cells (PGCs) arise far from the region where somatic gonadal precursors (SGPs) are specified. Although PGCs in general originate as a single cluster of cells, the somatic parts of the gonad form on each site of the embryo. Thus, to reach the gonad, PGCs not only migrate from their site of origin but also split into two groups. The progress of gonadal recovery was followed by periodic determination of gonadosomatic index, condition index, digestive gland index and oocyte diameter. There was significant difference in the gonadosomatic index between fed and unfed clams at 23°C and 28°C. Quail embryos at 4 days of incubation were treated with diethylstilboestrol (DES), using the egg dipping method. At 10 days of incubation, embryos were removed and killed by decapitation. To establish stromal cell lines of human aorta-gonad-mesonephros (AGM) region and study the growth characteristics and hematopoiesis related molecules expression in these cells. Stromal cells of AGM region in human embryos of day 30-35 were isolated and cultured in vitro. Besides their growth characteristics were studied, hematopoiesis related molecules including surface and extracellular matrix proteins were analyzed respectively by flow cytometry and immunofluorescence methods. Stromal cells of human AGM region held the morphology of fibroblastoid cells. During logarithmic growth period, the cell population doubling time is about 16 h. By flow cytometry, surface markers CD31, CD34, CD45 were negatively expressed, while CD29, CD44, CD105, CD166 were positively detected on human AGM stromal cells.
