Nanoparticle drug delivery (NDDS) is a novel system in which the drugs are delivered to the site of action by small particles in the nanometer range. Natural or synthetic polymers are used as vectors in NDDS, as they provide targeted, sustained release and biodegradability. Here, we used the chitosan and hepatoma cell-specific binding molecule, glycyrrhetinic acid (GA), to synthesize glycyrrhetinic acid-modified chitosan (GA-CTS). The synthetic product was confirmed by Fourier transformed infrared spectroscopy (FT-IR) and 1H-nuclear magnetic resonance (1H-NMR). By combining GA-CTS and 5-FU (5-fluorouracil), we obtained a GA-CTS/5-FU nanoparticle, with a particle size of 217.2 nm, a drug loading of 1.56% and a polydispersity index of 0.003. Objectives: To investigate the in vitro activity of Extractum liquiritiae (EL), glycyrrhizic acid (GL), glycyrrhetinic acid (GA) and a novel lipophilic derivative of glycyrrhetinic acid monoglucuronide (GAMG), acetylated GAMG (aGAMG), against 29 Helicobacter pylori strains.. Methods: The MIC of each compound was determined by the agar dilution method, and the killing kinetics were monitored in brain heart infusion broth (∼106-107 cfu/mL) at 0, 4, 24, 48, 72 and 96 h.. Glycyrrhizin (GA) analogs in the form of 3-glucuronides and 18-epimers were synthesized and their anticancer activities were evaluated. Alkaline isomerization of monoglucuronides is reported. In vitro and in vivo studies showed that glycyrrhetinic acid monoglucuronides (GAMGs) displayed higher anticancer activities than those of bisglucuronide GA analogs, while anticancer activity of the 18α-epimer was superior to that of the 18β-epimer. 18α-GAMG was firstly nicely bound to epidermal growth factor receptor (EGFR) via six hydrogen bonds and one charge interaction, and the docking calculation proved the correlation between anticancer activities and EGFR inhibitory activities. Highly active 18α-GAMG is thus of interest for the further studies as a potential anticancer agent.

BACKGROUND: Glycyrrhetinic acid (GA) is the most important ingredient in licorice due to its outstanding anti-inflammatory activity and wide application in the medicine and cosmetics industries. Contemporary industrial production of GA by acid hydrolysis of glycyrrhizin which was extracted from Glycyrrhiza plants, is not environment-friendly and devastates farmland since the Glycyrrhiza rhizomes grow up to 10 m underground. RESULTS: In this study, GA was produced through metabolically engineering Saccharomyces cerevisiae by introducing the entire heterogeneous biosynthetic pathway of GA. Codon optimized CYP88D6 and CYP72A154, combined with ß-AS (ß-amyrin synthase encoding gene) and the NADPH-cytochrome P450 reductase gene of Arabidopsis thaliana were introduced into S. cerevisiae. Retention of sodium and water was observed in both intact and adrenalectomized rats during 4 hour minerosteroid assays, the degree of retention being a function of log dose. In intact rats, the antidiuretic effect of glycyrrhizin was immediate and cumulative, while retention of sodium appeared only after a latent period and did not increase with time. Adrenalectomized rats, 3 days after the operation, showed a more marked antidiuretic effect than intact rats, but no greater retention of sodium.. A unique approach for extracting the thermal conductivity along the lateral and transverse heat flow directions was used in order to determine the thermal conductivity along different crystallographic directions. The data analysis at room temperature confirmed the expected anisotropy of the thermal conductivity of beta-Ga2O3, revealing the highest value of similar to 29 W/m K in the [010] direction. The thermal conductivity of the Sn-doped and Fe-doped beta-Ga2O3 samples was found to be lower than that of the undoped samples due to the enhanced phonon-impurity scattering contribution, which reduces the thermal conductivity.
