McArdle disease is a rare metabolic myopathy caused by a complete absence of the enzyme muscle glycogen phosphorylase. Affected people experience symptoms of fatigue and cramping within minutes of exercise and are at risk for acute muscle injury (rhabdomyolysis) and acute renal failure. One mutation that is common in North American and European populations is written as Arg50Ter or R50X. This mutation creates a premature stop signal in the instructions for making myophosphorylase, which decreases the production of the enzyme.
