Klotho is a transmembrane protein with a wide spectrum of activity. The human Klotho gene shows 86% amino acid identity with the mouse protein. Many important pleiotropic functions of the Klotho protein have been revealed. Amongst them, there is a regulation of nitric oxide production, suppression of oxidative stress and inflammation, influence on the insulin-like growth factors and fibroblast growth factors signaling, modulation of calcium and phosphate metabolism, synthesis of vitamin D and other. Two forms of the Klotho protein are known. The secreted form strongly inhibits the oxidative stress, and, in humans, is more dominant than the membrane form. Studies on a mouse model resulted in the finding of the anti-aging effect of the Klotho protein. Mouse beta-glucuronidase has a dual intracellular localization, being present in both endoplasmic reticulum and lysosomes of several tissues. Previous studies demonstrated that the protein egasyn is complexed with microsomal but not lysosomal glucuronidase and that a mutant lacking egasyn is deficient in microsomal, but not lysosomal, glucuronidase. By means of a recently developed radioimmunoassay for egasyn, the relationship between microsomal glucuronidase levels and egasyn levels has been examined in various adult tissues, during postnatal development in liver, and after androgen induction of glucuronidase in kidney. The results indicate that the relative availability of egasyn determines the balance between glucuronidase incorporation into membranes and that into lysosomes.

Here, we demonstrate that COX2 inhibition reduces osteogenic gene induction and AoV calcification in a mouse model of CAVD. COX2 expression is increased in calcified AoVs of klotho-deficient mice and is localized to the VICs before the formation of AoV calcification, which occurs in the context of minimal inflammation. In human CAVD, COX2 expression is increased and is present in regions where the valve leaflet is calcified. In porcine VICs, COX2 mRNA expression is induced upon treatment with osteogenic media, and COX2 inhibition reduces osteogenic gene induction and calcification, supporting a direct role for COX2 in VIC mineralization. Furthermore, COX2 inhibition through genetic or pharmacological manipulation is sufficient to reduce AoV calcification and osteogenic gene expression in klotho-deficient mice. Several carboxypeptidase Y-/3-glucuronidase (CPY-GUS) plasmids were expressed in yeast. The CPY-GUS fusion proteins were inactivated during passage through the secretory pathway. This work was done in an attempt to recover GUS activity in yeast. CPY-GUS plasmids were transformed to protease-deficient yeast strains so as to test whether the low GUS activity was due to the degradation by intracellular proteases. The results failed to show this because of the problems associated with the western blots. Tunicamycin, a drug that inhibits glycosylation, was added to the yeast culture in order to recover GUS activity. The results show that a short term treatment could increase GUS activity by about 10%. In a long term treatment, tunicamycin was found to affect the normal growth of yeast adversely and reduce the GUS activity by 90%. A limited number of studies have checked heparanase levels in different sarcoma types, including a study by Shafat et al. [16], which examined the level of heparanase in pathological samples taken from children with Ewings sarcoma. Heparanase levels were evaluated using immunohistochemistry of 69 pathological samples utilizing methodology similar to that applied in this study. Lysosomal enzyme beta-glucuronidase (GUSB) was found as a key regulator of arthritis severity associated to Lyme disease that is caused by the spirochete Borrelia burgdorferi, the most prevalent arthropod-borne illness in the United States. Severely arthritic C3H mice possessed a naturally occurring hypomorphic Gusb allele and C3H mice expressing WT Gusb as a transgene were protected from severe Lyme arthritis. The hypomorph Gusb allele also exacerbated disease in a model of rheumatoid arthritis. Development of Lyme and rheumatoid arthritis in mice carrying the Gusb hypomorph allele was associated with heightened accumulation of GAGs in joint tissue. Pancreatic β-cells are essential to the regulation of glucose homeostasis. Substantial β-cell failure is now believed to occur at an early stage in the progression of T2DM (2). Thus, one of the goals in the treatment of T2DM is to preserve functional β-cells. Klotho, a recently discovered aging-suppressor gene, was believed to be expressed in kidneys (21). Our most recent study indicates that the Klotho gene and protein are expressed in pancreatic islets (11). Notably, Klotho protein expression in β-cells was decreased in both patients with T2DM and db/db mice, a mouse model of T2DM (Fig. 1A-F). The role of the fecal microflora in the induction of colon cancer was investigated in individuals believed to be genetically predisposed to colon cancer. Subjects were members of families with increased occurrence of colon and endometrial carcinomas characteristic of the cancer family syndrome. Group 1 consisted of 5 cancer family syndrome individuals previously diagnosed with colon cancer. Homology-dependent gene silencing contributes to variation between transgenic plants with respect to transgene and/or endogenous gene expression levels. Recent studies have linked post-transcriptional gene silencing and virus resistance in plants expressing virus-derived transgenes. Using a potato virus X vector, we present three examples in which silencing of nonviral transgenes prevented virus accumulation. This effect was dependent on sequence homology between the virus and the silenced transgene. Analysis of potato virus X derivatives carrying bacterial [beta]-glucuronidase (GUS) sequences showed that the 3[prime] region of the GUS coding sequence was a target of the silencing mechanism in two independent tobacco lines. Methylation of the silenced GUS transgenes in these lines was also concentrated in the 3[prime] region of the GUS coding sequence. When the mRNAs for these two proteins were translated in the presence of microsomal membranes, the in vitro synthesized polypeptides were cotranslationally glycosylated and transferred into the microsomal lumen. Polypeptides synthesized in the absence of microsomal membranes were approximately 2,000 daltons larger than the respective unglycosylated microsomal polypeptides found after short times of labeling in cultured rat liver cells treated with tunicamycin.
