Fig. 3. Development of stable STZ-induced diabetes requires coexisting diet-induced impaired glucose tolerance. (A) Diagrammatic representation of optimized STZ treatment to induce hyperglycemia in guinea pigs fed either normal or HFHC diet. STZ powder is dissolved in citrate buffer at 100 mg/ml, then incubated for 2 h to allow for α/β anomer equilibration. Twenty minutes before STZ treatment, guinea pigs are administered a 0.5 mg/kg dose of the α2 agonist yohimbine, by the intramuscular (IM) route. Guinea pigs are then given a single 200 mg/kg subcutaneous (SC) injection of anomer-equilibrated STZ. Diabetic hyperglycemia is determined by an oral glucose tolerance test (OGTT) at day 7, 14 and 21, based on a 2 h blood glucose ≥200 mg/dl. (B) Blood glucose concentrations were measured daily at random in nonfasted, STZ-treated guinea pigs fed a normal diet (n=5). Hyperglycemia, observable 24 h after STZ treatment, has a trend of steady decline over the course of 14 days. *P,0.05 compared with ...
Obesity Increases the Risk of Glucose Intolerance. Glucose intolerance, otherwise known as impaired glucose tolerance (IGT), is a condition that is often found to be in patients who eventually develop type 2 diabetes mellitus. (1) A patient with glucose intolerance is often also at high risk of cardiovascular disease as well as being resistant to insulin. The most significant characteristic of glucose intolerance is that it is often referred to as a gateway to developing type 2 diabetes. (2) Additionally, obesity has been shown to play a significant role in the development of both glucose intolerance and diabetes. (3) Furthermore, another study specifically found that upper-body obesity, that is an abundance of adipose body fat around the abdominal area as well as hypertension are all related to glucose intolerance as well. This is often referred to as the deadly quartet of upper-body obesity, glucose intolerance, hypertriglyceridemia, and hypertension. (4). Even though all these ...
Approach and Results-Diet-induced obese Id3Bcell KO mice demonstrated attenuated inflammation and insulin resistance in visceral adipose tissue (VAT), and improved systemic glucose tolerance. VAT in Id3Bcell KO mice had increased B-1b B cells and elevated IgM natural antibodies to oxidation-specific epitopes. B-1b B cells reduced cytokine production in VAT M1 macrophages, and adoptively transferred B-1b B cells trafficked to VAT and produced natural antibodies for the duration of 13-week studies. B-1b B cells null for Id3 demonstrated increased proliferation, established larger populations in Rag1−/− VAT, and attenuated diet-induced glucose intolerance and VAT insulin resistance in Rag1−/− hosts. However, transfer of B-1b B cells unable to secrete IgM had no effect on glucose tolerance. In an obese human population, results provided the first evidence that B-1 cells are enriched in human VAT and IgM antibodies to oxidation-specific epitopes inversely correlated with inflammation and ...

The body of a person with High Blood Sugar Levels either does not produce enough insulin or they cannot use their produced insulin properly, which results in glucose accumulation in the blood.. This high level of blood glucose can cause a range of symptoms, from exhaustion to heart disease and sometimes even heart attack.. Furthermore, sudden high blood sugar level could also be one of the answers to the question of what causes glucose intolerance.. High glucose levels can severely affect you from head to toe. In hypertension, an elevated blood pressure induces left ventricular hypertrophy to compensate for increased wall stress4 and also impairs endothelial function by increasing shear stress.15 In this context, an association between these 2 pathophysiological abnormalities has been reported.5 In subjects with abnormal glucose metabolism, hyperglycemia also impairs endothelial function.16 Thus, either elevation of blood pressure or impaired glucose metabolism can directly affect endothelial function. To our knowledge, this is the first study to examine whether left ventricular hypertrophy or abnormal glucose metabolism is more closely associated with endothelial dysfunction in hypertensive patients.. In the present study, multiple linear regression analysis demonstrated a significant relationship between endothelial dysfunction and 2-hour BG but no significant correlation between endothelial function and elevated blood pressure level or increased left ventricular mass. Hypertensive patients were ...

Detecting glucose intolerance after gestational diabetes: inadequacy of fasting glucose alone and risk associated with gestational diabetes and second trimester waist-hip ratio
Introduction: Patients with cystic fibrosis (CF) and normal glucose tolerance (NGT) may exhibit diabetic glucose tolerance during pulmonary exacerbations [1].. Aims: We examined glucose tolerance during exacerbations in children with CF and compared continuous glucose monitoring (CGM) with the gold standard oral glucose tolerance test (OGTT).. Methods: Children with CF aged at least 10 years hospitalised with an exacerbation were recruited. Those with diabetes and those on corticosteroids were excluded. On admission, patients had an OGTT and were fitted with a CGM for 3 days. Six weeks post discharge both tests were repeated.. Results: Ten patients (3 males), mean age 13.9 years, have completed the study. Two patients had CGM alone, 4 patients had OGTT alone and 4 patients had both tests. By OGTT criteria, 3 were diabetic, 3 had NGT and 2 had impaired glucose tolerance (IGT) during exacerbations. Glucose tolerance status did not change between exacerbation and follow-up although mean 2-hour ...

We have generated skeletal muscle-specific Gsα-knockout (KO) mice (MGsKO) by mating Gsα-floxed mice with muscle creatine kinase-cre transgenic mice. MGsKO mice had normal body weight and composition, and their serum glucose, insulin, free fatty acid, and triglyceride levels were similar to that of controls. However, MGsKO mice were glucose intolerant despite the fact that insulin ...
Introduction Hepatic steatosis is a major risk factor for the development of severe liver damage, including fibrosis, cirrhosis and hepatocellular carcinoma. It often exists as a co-morbidity factor with diabetes type I/II and with other manifestations of the metabolic syndrome. Recent studies highlight the importance of an epigenetic basis for the development of steatosis based on macroH2A1. MacroH2A1 is a histone variant of histone H2A, which possesses an additional protein domain called macro. When incorporated into the chromatin of hepatocytes, macroH2A1 regulates gene expression. Two alternatively spliced isoforms of macroH2A1 exist, which have been shown to be markers of breast, skin and lung cancer. Whole-body knock out of macroH2A1 in mice induces glucose intolerance and changes in genes regulating hepatic lipid metabolism. However, overt hepatic steatosis was not observed and the significance of these findings is unclear. We hypothesised that macroH2A1 could be involved in the ...
Recent studies have demonstrated subset-specific roles for B cells in both aggravating and regulating the perturbations associated with obesity. Winer et al4 demonstrated that B-2 B cells promote insulin resistance and glucose intolerance through production of pathogenic antibodies. Additional studies have shown that sorted splenic B-2 B cells from obese mice secrete more IL-6 and MIP-2 and less IL-10 compared with lean controls,48 and circulating B cells (of which the vast majority are B-2) from patients with type 2 diabetes mellitus are also skewed toward a proinflammatory phenotype after toll-like receptor stimulation.49 Our adoptive transfers of B-2 B cells provide further evidence that B-2 B cells promote metabolic dysfunction during DIO. In contrast, B-1a B cells were recently shown to protect against the inflammatory and metabolic consequences of DIO through a combination of IgM and IL-10 production,50 and B-cell-derived IL-10 reduces adipose tissue inflammation and insulin resistance ...
We chose the criteria of the World Health Organization to define gestational glucose intolerance on the basis of their worldwide use. As well, we set out to study metabolism by means of intermediate glucose intolerance instead of gestational diabetes. 14,15 We believe this choice to be a strength of our study. A large cohort study found that women without gestational diabetes and only mildly elevated glucose concentrations during pregnancy had an increased risk of postpartum diabetes and may benefit from increased (and early) monitoring of their glucose metabolism during pregnancy. 16 Our results suggest a way to estimate individual risk to improve this kind of close monitoring.. One limitation of our study was our modest sample size. It did not allow for the various subdivisions (e.g., by parity) that would help to better document the usefulness of the hyper-triglyceridemic-waist phenotype in the identification of women with a higher risk of gestational glucose intolerance or diabetes. ...
Introduction: Malignant neoplasm is a major cause of death in developed countries, and its incidence continues to grow, placing a heavy burden on the community. Diabetes mellitus is a serious and leading health problem. Recent studies demonstrated that glucose intolerance was associated with a higher risk of oral cancer death, beginning in the prediabetic range of glucose intolerance. However, few population‑based studies, especially in Asian populations, have addressed these issues or have estimated glucose intolerance status. Aim: We undertook this study with the aim of finding out an association between impaired glucose tolerance (IGT) and oral cancer along with finding out prevalence of other risk factors for oral cancer. Materials and Methods: Forty‑five cases and 45 controls were selected for the study. Oral glucose tolerance was performed on subjects who satisfied inclusion criteria and were willing to sign informed consent form. Results: Fifty‑three percent of the cases had ...

Consistent with our hypothesis, deletion of SOD1 resulted in glucose intolerance. This suggests that oxidative damage plays a role in the glucose metabolic abnormalities in T2D subjects. Surprisingly, the glucose intolerance seen in SOD1-null mice was not the result of significantly reduced insulin sensitivity in periphery (muscle) tissues or the liver. This prompted us to examine whether abnormalities in β-cell function could explain the glucose intolerance. Indeed, the results from the hyperglycemic clamp demonstrated a significantly blunted insulin secretion response, a defect that was in part explained by a reduction in β-cell volume.. The effect of oxidative stress on insulin sensitivity and peripheral (muscle) glucose disposal is controversial (18-20). Elevations in plasma glucose and free fatty acids are thought to increase ROS levels (18,21), which in turn activate inflammation signaling pathways such as mitogen-activated protein kinases (22) and nuclear factor-κB (23). The activation ...
Aim: Physical exercise reduces obesity, insulin resistance and dyslipidemia. We previously found that maternal obesity alters central appetite circuits and contributes to increased adiposity, glucose intolerance and metabolic disease in offspring. Here we hypothesized that voluntary exercise would ameliorate the adverse metabolic effects of maternal obesity on offspring. Methods and Results: Sprague-Dawley females fed chow (C) or high-fat diet HFD (H) were mated. Female offspring from C dams were weaned onto chow (CC); those from H dams recieved chow (HC) or HFD (HH). Half of each group was provided with running wheels (CCEX, HCEX, HHEX; n=10-12).Maternal obesity increased body weight (12%), adiposity, plasma lipids and induced glucose intolerance (HC vs CC; P,0.05). These were exaggerated by postweaning HFD (HH vs HC; P,0.01), showed doubled energy intake, a 37% increase in body weight, insulin resistance and glucose intolerance (HH vs HC; P,0.01). Exercise reduced fat mass, plasma lipids, HOMA ...

In this study, we demonstrate a stepwise increase in the prevalence of dysglycemia at each of 3 months, 1 year, and 3 years postpartum, as gestational glucose tolerance worsens from normal (normal GCT NGT) to mildly abnormal (abnormal GCT NGT) to moderately abnormal (GIGT) to GDM. At each postpartum time point, there was an analogous gradient of β-cell dysfunction, insulin resistance, and increasing glycemia across these four groups. Most importantly, each degree of gestational glucose intolerance independently predicted distinct trajectories of β-cell function, insulin sensitivity, and glycemia during the first 3 years postpartum. It thus emerges that the spectrum of glucose intolerance in pregnancy may enable anticipation of the course of metabolic function in the first 3 years after delivery.. A series of previous studies have demonstrated that each degree of dysglycemia in pregnancy predicts a proportionately increased future risk of T2DM in the years thereafter, with GDM representing the ...
In 1963 a long-term incidence study of ischemic heart disease and hypertension was initiated in Israel. A total of 10,059 Jewish men over forty years of age in government service were surveyed. The initial examination included a medical history and determinations of blood sugar, cholesterol, and uric acid. There were 682 men designated as diabetic suspects, because of either a history of diabetes or a casual blood sugar value of at least 130 mg. or both.. Results of previous glucose tolerance tests, fasting blood sugar estimates, and information about treatment were obtained from physicians and clinics, or a glucose tolerance test was done at the survey laboratory. For forty-three men not enough information was obtained to decide whether they actually had diabetes. Seventy-nine men were not diabetic, and sixty-two others were not diabetic but had abnormal glucose tolerance tests. The remaining 498 men were classified as diabetic, and of these 296 had a history of the disease and 202 were newly ...
