Fish gills represent a complex organ composed of several cell types that perform multiple physiological functions. Among these cells, ionocytes are implicated in the maintenance of ion homeostasis. However, because the ionocyte represents only a small percent of whole gill tissue, its specific transcriptome can be overlooked among the numerous cell types included in the gill. The objective of this study is to better understand ionocyte functions by comparing the RNA expression of this cell type in freshwater and seawater acclimated rainbow trout. To realize this objective, ionocytes were captured from gill cryosections using laser capture microdissection after immunohistochemistry. Then, transcriptome analyses were performed on an Agilent trout oligonucleotide microarray. Gene expression analysis identified 108 unique annotated genes differentially expressed between freshwater and seawater ionocytes, with a fold change higher than 3. Most of these genes were up-regulated in freshwater cells.

An examination is made of the hypothesis that endogenous 5-hydroxytryptamine (5-HT) serves as a local hormone regulating ciliary activity in the lamellibranch gill. These cilia are sensitive to exogenous 5-HT and respond to it by a prompt, sustained, and reversible rise in beat frequency; at the same time the carbohydrate metabolism is stimulated, as described elsewhere. Control gill contains small but definite amounts of endogenous 5-HT according to bioassay, fluorometry, and chromatography. The amount can be increased markedly by exposing the isolated gill to the precursor substance 5-hydroxytryptophan but not l-tryptophan. As the tissue level of 5-HT rises, the spontaneous beat frequency also rises. Both remain elevated for hours and perhaps for days. Brandi Carlile and Shooter Jennings co-produced the disc with Carlile having a hand in doing much of the writing. Gill goes back to his roots with OKIE, his first disc in three years. Gill gets personal throughout the album with thoughts about marriage, faith and sexual abuse included in the mix. He also paid tribute to the late songwriter Guy Clark. Copper (Cu) is an essential element required by all living organisms, since at least 30 enzymes are known to use Cu as a cofactor. Cu is also toxic in excess and liver and gills are known to be target organs for it. In the present study, 240 Siberian sturgeon juvenile (with initial weight 29.2 ± 3.1 g and initial length 21.8 ± 1.4 cm) were randomly distributed in 12 fiberglass tanks at 4 different copper nanoparticle (Cu-NPs) treatments with 3 replicates. Treatments included control (T0 = no added Cu-NPs), 50 (T50), 100 (T100), 200 (T200) µg.l-1 Cu-NPs (mean primary particle size of 2 - 6 nm) in a semi-static waterborne exposure regime. Water exchanged were 20% daily with re-dosing after each change. The experimental period lasted 28 days, 14 days exposure to Cu-NPs and 14 days as recovery time. In addition to tracking the pH in fishes under normal physiological conditions, we monitored the pH in comatose and dying individuals. Repeated anesthesia can be associated with intoxication, after which some fish cannot be awakened. The percentage of fish that fell into a coma is probably dependent on the natural variability in the health status of the subjects. The state is caused by oxygen deprivation in the brain, which generally results from poor gill ventilation and decreased heart rate. We applied the chosen anesthesia procedure intentionally to induce significant disturbances in metabolism and show the functionality of pH-sensitive MBMs. The pH values in dead fish were measured just once after death and were excluded from the experiment after the 100 min time point.. The median blood pH in the gills of a comatose fish (7.20) was lower than that in fish at normal state (7.47) when measured 100 min after the injection, but the difference was not statistically significant (P-value=0.072). ...

Juvenile and adult growth of Chasmagnathus granulatus was studied in the laboratory in terms of molt increment in size (MI) and the intermolt period (IP), comparing data obtained from short-term (STE) and long-term (LTE) laboratory experiments. Crabs in a premolt condition were collected for STE, including the entire size range of the species. Larger crabs remained in the laboratory no more than 14 days; the average time to molt was 5.8 ± 3.1 d. We registered the molt of 94 females, 64 males and 34 undifferentiated juveniles and calculated their MI. Moreover, 24 males and 4 females were reared in the laboratory over 3 years (LTE). Hiatt diagrams did not show sex-specific differences between juveniles of both sexes, but revealed differences between juveniles and adults in each sex as well as between adults of both sexes. Six consecutively hatched cohorts and one cohort of pre-hatch eggs of farmed barramundi (Lates calcarifer) from south Australia were examined for Chlamydia-like organisms associated with epitheliocystis. To identify and characterise the bacteria, 59 gill samples and three pre-hatch egg samples were processed for histology, in situ hybridisation and 16S rRNA amplification, sequencing and comprehensive phylogenetic analysis. Cases of epitheliocystis were observed microscopically and characterised by membrane-enclosed basophilic cysts filled with a granular material that caused hypertrophy of the epithelial cells. In situ hybridisation with a Chlamydiales-specific probe lead to specific labelling of the epitheliocystis inclusions within the gill epithelium. Two distinct but closely related 16S rRNA chlamydial sequences were amplified from gill DNA across the seven cohorts, including from pre-hatch eggs. This study surveyed freshwater fishes Cyprinus carpio and Carassius auratus from Chongqing, China, for infection with ectoparasitic trichodinids. Fish specimens were obtained from culture pools, smears of gills were prepared and used for silver nitrate impregnation. Microbiologic studies identified two species of trichodinids belonging toTrichodina Ehrenberg, 1838 including one new species from C. carpio that was namedTrichodina cyprinocola sp. nov. Comparisons with closely related species revealed that this new species T. cyprinocola has unique sturdy denticles including the sickle blade with bluntly round tangent point, well developed central part, robust, straight but short ray. The other known species Trichodina borokensis Arthur and Lom (1984) from C. auratus was described here as the first record in China. Our survey also yielded morphometric data and prevalence of infestation for these two species.
