Gentian (Gentiana sp.) is an important perennial flower in Japan. In terms of cut flower cultivation area size it occupies 2nd to 3rd place. Cucumber mosaic virus (CMV), broad bean wilt virus (BBWV), tobacco rattle virus and clover yellow vein virus have been reported to infect gentians. We have analyzed gentian plants collected from the largest production area in Japan, Ashiro-cho in the Iwate prefecture, and found that most plants with severe virus symptoms were infected either by CMV alone or by both CMV and BBWV. The association of CMV with severe symptomatolgy was also confirmed in greenhouse experiments in which CMV and BBWV isolates obtained from symptomatic gentian plants were inoculated to gentian seedlings. CMV induced severe symptoms, while BBWV did not cause any. Dual inoculation of CMV and BBWV slightly aggravated symptoms in comparison with those caused by CMV alone. Currently there are few flowers blooming in the Dunson Native Flora Garden but the curator, Joey Allen, is trying to plant some that will flower at this time of year. One recent planting is harvest bells, or soapwort gentian, Gentiana saponaria, that was in full bloom. Someone asked if its closed flower ever opened up, and the answer was that it does not. Bumble bees that pollinate it have to push their way in. Somehow we got to talking about the fringed gentian, Gentianopsis crinita, that grows on the road verges around Track Rock Gap near Young Harris. At least it used to do so. Common models for the evolution of duplicated genes after genome duplication are subfunctionalization, neofunctionalization, and pseudogenization. Although the crucial roles of cis-regulatory mutations in subfunctionalization are well-documented, their involvement in pseudogenization and/or neofunctionalization remains unclear. We addressed this issue by investigating the evolution of duplicated homeobox genes, six6.L and six6.S, in the allotetraploid frog Xenopus laevis. Based on a comparative expression analysis, we observed similar eye-specific expression patterns for the two loci and their single ortholog in the ancestral-type diploid species Xenopus tropicalis. However, we detected lower levels of six6.S expression than six6.L expression. The six6.S enhancer sequence was more highly diverged from the orthologous enhancer of X. tropicalis than the six6.L enhancer, and showed weaker activity in a transgenic reporter assay. We were in Boston over the weekend and I got back last night in time to take a good look around the changes in the garden wrought by a few more days of sunshine. The thing that most caught my attention (and sent me into the house for the camera) was this Chilean Blue Crocus. This was a new one for me. I had grown the Leichtlinii variant last year and was very much taken with the white face surrounded by sky blue edges, but I hadnt ever seen the standard Chilean Blue before. As a femme dun certain age (48, if you must know), I am assuming that my stomach acid is no longer as strong as it used to be (thats what you read, at least). In white-haired older male patients, we dyed the surrounding scalp skin purple with a gentian violet solution-stained toothpick. Our method has several advantages: surgeons can easily focus on the center of the follicular unit and rapidly perform punching, they can recognize the condition of the harvested follicular units during FUE, and the hair transplant team can secure a clear view for trimming and loading into the implanter. We developed and applied a mutagenesis protocol based on γ-irradiation of in-vitro cell cultures from which regenerants were obtained. Based on a radio-sensitivity test, the irradiation dose reducing to 50% the number of regenerants per callus (RD50) was estimated at 35 Gy. A large mutagenic experiment was carried out by irradiating a total of 3,120 calli with approx. 1x, 1.5x and 2x RD50. A total of 1,004 regenerants from irradiated calli were hardened in pots and transplanted to the field. Approx. 10% of field grown clones showed remarkable morphological aberrations including dwarfism, altered tillering, abnormal inflorescence, leaf variegation and others, which were tested for stability over generations.
