If you have a question about this talk, please contact Fiona Roby.. Clonality is a fundamental property of neoplasia and the evaluation of the clonal nature of a lymphocyte population within tissue samples is commonly used as an adjunctive diagnostic procedure in humans. In recent years, molecular techniques have become available for the diagnosis of canine, feline and equine lymphoproliferative disorders. Polymerase chain reaction (PCR) for antigen receptor rearrangement is a method used for the detection of clonal lymphocyte populations based on the assessment of diversity within the complementary determining region 3 of the immunoglobulin heavy chain variable region genes and the T cell receptor gamma genes. It represents a sensitive and flexible PCR assay system for the analysis of various source materials e.g. fine needle aspirates, whole blood, bone marrow, cerebrospinal fluid also allowing retrospective studies based on formaline fixed paraffin embedded material. In addition to the ...
Adaptative immune repertoire diversity in vertebrate species is generated by recombination of variable (V), diversity (D) and joining (J) genes in the immunoglobulin (IG) loci of B lymphocytes and in the T cell receptor (TR) loci of T lymphocytes. These V-J and V-D-J gene rearrangements at the DNA level involve recombination signal sequences (RSS). Whereas many data exist, they are scattered in non specialized resources with different nomenclatures (eg. flat files) and are difficult to extract. IMGT/GeneInfo is an online information system that provides, through a user-friendly interface, exhaustive information resulting from the complex mechanisms of T cell receptor V-J and V-D-J recombinations. T cells comprise two populations which express the αβ and γδ TR, respectively. The first version of the system dealt with the Homo sapiens and Mus musculus TRA and TRB loci whose gene rearrangements allow the synthesis of the αβ TR chains. In this paper, we present the second version of IMGT/GeneInfo

Interleukin 7 receptor (IL7R) also known as CD127 (Cluster of Differentiation 127) is a type I cytokine receptor. IL7R also denotes its human gene.. The protein encoded by this gene is a receptor for interleukine 7 (IL7). The function of this receptor requires the interleukin 2 receptor, gamma chain (IL2RG, or CD132), which is a common gamma chain shared by the receptors of various cytokines, including interleukine 2, 4, 7, 9, and 15. This protein has been shown to play a critical role in the V(D)J recombination during lymphocyte development. This protein is also found to control the accessibility of the TCR gamma locus by STAT5 and histone acetylation. Knockout studies in mice suggested that blocking apoptosis is an essential function of this protein during differentiation and activation of T lymphocytes. The functional defects in this protein may be associated with the pathogenesis of the severe combined immunodeficiency (SCID). [1] It has also recently been linked to the pathogenesis of ...

Aim: To compare clonal T cell receptor γ (TCRγ) gene rearrangements in frozen and formalin-fixed paraffin wax-embedded (FFPE) tissue, using capillary electrophoresis for use in diagnostics, as T cell lymphomas may be difficult to diagnose by conventional methods.. Methods: The DNA for PCR was extracted from frozen and FFPE tissue, cell lines and blood. PCR primers Vγ1-8, Vγ9, Vγ10 or Vγ11 (5′ end labelled) combined with a mixture of JγP1/JγP/JγP2/Jγ2 (unlabelled) were used. Monoclonal cases were sequenced and clonality, reproducibility, sensitivity and specificity analyses were carried out.. Results: In all cases the molecular test was found to be in agreement with the histological diagnosis. Discrepancies were found between frozen and FFPE tissue in 18 of 56 (32%) tests. The method was highly reproducible. The sensitivity was found to be 0.5% for cell lines and 1% for patient specimens and the specificity 100%. The junctional region between the Vγ and Jγ segments was specific for ...
The recognition that the majority of ACS evolve from only mildly to moderately obstructive atherosclerotic plaques led to an appreciation of nonmechanical factors contributing to the clinical manifestations of coronary artery disease.1 2 3 Our finding that an unusual subset of T lymphocytes, CD4+CD28null T cells, is expanded in patients with UA but not in patients with SA suggests that these cells are involved in the acute events complicating otherwise stable and slowly progressive coronary disease. This T-cell population breaks several rules that apply to classic CD4 helper T cells.21 CD4+CD28null T cells contain monoclonal T-cell populations that persist over time at a large clonal size. Sequence homologies of TCR β-chains isolated from clones of multiple patients support the notion that these clones are expanded in response to exogenous antigen(s) common to patients with UA. CD4+CD28null T-cell clonotypes are accumulated in lesions with plaque rupture, suggesting that they are involved in ...

The human TCR-gamma delta occurs in three biochemically distinct forms (forms 1, 2bc, and 2abc). A 40-kDa TCR gamma-chain is disulfide-linked to the TCR delta-chain in form 1, whereas 40-kDa or 55-kDa TCR-gamma polypeptides are noncovalently associated with the TCR delta-chain in forms 2bc and 2abc, respectively. Sequence analysis of TCR-gamma cDNA clones indicates that form 1 utilizes the C gamma 1 gene segment, whereas forms 2bc and 2abc appear to use allelic C gamma 2 gene segments containing either two copies (b and c) or three copies (a, b, and c) of the CII exon, respectively. We transfected TCR-gamma cDNA encoding form 1 or form 2abc into the MOLT-13 cell line that expresses form 2bc. The transfected TCR gamma-chains associate with the resident MOLT-13 TCR-delta, normally part of form 2bc, to yield CD3-associated TCR-gamma delta heterodimers identical to those seen on the donor cell lines (form 1 or 2abc). These transfection experiments show directly that, 1) when a single TCR-delta ...

I wouldnt worry about that statement too much. I think it was targetted to help motivate entrants to complete the judging so they can boost their own chances of winning. Its simply mentioning that as an entrant, you are welcomed to be a judge (for all entries including your own). Since you are your own judge, you have the benefit of being able to give yourself the highest marks/grade when scoring your entry. That doesnt mean you *HAVE* to give yourself a perfect score. But everyone else may be doing so, its more of a potential for an extra bonus point for your score if you take the time to complete the judging ...
Cytogenetic and molecular investigations of Acute Lymphoblastic Leukaemia (ALL) have identified the existence of distinct clinical subgroups. Molecular monitoring of clonal Immunoglobulin and T cell receptor (IG/TR) gene rearrangements has become an important tool in stratification of therapy of ALL. In order to determine whether certain features of the patient-specific rearrangements could hold further prognostic clues or provide information on the cell of origin of ALL, a comprehensive analysis of structural and biological features (V gene usage, coding frame and mutational status and complementarity-determining region -III length) of 473 IG/TR rearrangements identified in 229 adults with ALL was carried out. Distinct variable-gene usage profiles were identified between ALL subgroups, particularly for patients positive for BCR-ABL1 compared to MLL-AFF1 positive leukaemias; suggesting that the former is derived from a more mature B progenitor. Interestingly, occurrence of TRGV1-TRGV8 was prognostic for
We examined the developmental profile of TCR-gamma/delta+ cells with respect to CD45RO expression. Although total TCR-gamma/delta+ cells were negligible in the neonatal blood and increased with advancing age, most blood TCR-gamma/delta+ cells markedly expressed CD45RO without a distinction of age, probably reflecting a different CD45RO expression of two subsets defined by BB3 and delta TCS1 mAbs. The vast majority of BB3+ cells expressed CD45RO, whereas expression of CD45RO was virtually absent in the delta TCS1+ population. Functional studies revealed that, while both TCR-gamma/delta+ cell subsets showed CD3-mediated activation, only BB3+ (or Ti gamma A+) cells, but not delta TCS1+ cells, appeared to proliferate in response to PPD in PPD-reactive individuals. The results suggested that the CD45RO+ (BB3+ or Ti gamma A+) subset among blood TCR-gamma/delta+ cells may be mainly involved in the memory or primed component of the immune system responding to some foreign antigens. ...

Gastrointestinal lymphomas represent 5–20% of extranodal lymphomas and mainly occur in the stomach and small intestine. Clinical findings are not specific, thus often determining a delay in the diagnosis. Imaging features at conventional and cross-sectional imaging must be known by the radiologist since he/she plays a pivotal role in the diagnosis and disease assessment, thus assisting in the choice of the optimal treatment to patients. This review focuses on the wide variety of imaging presentation of esophageal, gastric, and small and large bowel lymphoma presenting their main imaging appearances at conventional and cross-sectional imaging, mainly focusing on computed tomography and magnetic resonance, helping in the choice of the best imaging technique for the disease characterization and assessment and the recognition of potential complications.
T-Cell Receptors composed of CD3-associated gamma and delta polypeptide chains and expressed primarily in CD4-/CD8- T-Cells. The receptors appear to be preferentially located in epithelial sites and probably play a Role in the recognition of Bacterial Antigens. The T-Cell Receptor gamma/delta chains are separate and not related to the gamma and delta chains which are subunits of CD3 (see Antigens, CD3 ...
Hallo, Im a 43 y.o. male and Ive been diagnosed of t-cell LGL leukemia... or maybe not! I cant still understand the mechanism of this illness, and doctors cant give exact answers, maybe someone of you can help me. I write maybe not because actually I do not have any kind of cytopenia, neutrophils are higher than 2000, lymphocites low then 3000, hemoglobin is 13. But I have 22% lymphocites with LGL caratheristics and t-cell receptor analysis defined a monoclonal T-cell proliferation. Parameters of immunogram blood tests are now the same than 12 years ago (even better). In 2006 I tried to understand with an hematologist why I had always in the analys an higher % of lymphocites compared to neutrophils, a low CD4/CD8, very low B lymphocites and moderate high t-killers. Last year in september, after recurrent infections I tried again to understand what was the deal, and only now after several months and doctors, looks like that someone understood what is the problem. Since my clinical picture ...

We have investigated the role of common gamma chain (gamma c)-signaling pathways for the development of T cell receptor for antigen (TCR)-gamma/delta T cells. TCR-gamma/delta-bearing cells were absent from the adult thymus, spleen, and skin of gamma c-deficient (gamma c-) mice, whereas small numbers of thymocytes expressing low levels of TCR-gamma/delta were detected during fetal life. Recent reports have suggested that signaling via interleukin (IL)-7 plays a major role in facilitating TCR-gamma/delta development through induction of V-J (variable-joining) rearrangements at the TCR-gamma locus. In contrast, we detected clearly TCR-gamma rearrangements in fetal thymi from gamma c- mice (which fail to signal in response to IL-7) and reduced TCR-gamma rearrangements in adult gamma c thymi. No gross defects in TCR-delta or TCR-beta rearrangements were observed in gamma c- mice of any age. Introduction of productively rearranged TCR V gamma 1 or TCR V gamma 1/V delta 6 transgenes onto mice bearing ...

VetVine is an accredited Continuing Education provider for veterinary professionals and resource of expert-driven pet health information for pet owners. This is the forum topic view page. Feline small intestinal lymphoma is most commonly a mucosal T-cell lymphoma (SCL) and confident diagnosis remains challenging, as it presents histologically similar to inflammatory bowel disease (IBD). Distinguishing between SCL and IBD is important since > 50% of cats with SCL die within 1 year while > 30% of cats with IBD are still alive after 1 year, and treatment may be different for the 2 conditions. Diagnosis of SCL is most appropriately based on combination of histology, immunohistochemical phenotyping (CD3 – T cell, CD79a – B cell), and polymer chain reaction to assess antigen receptor gene rearrangements (PARR). PARR improves diagnosis by amplifying genes for T-cell receptor gamma (TCRG) in the case of T cell phenotype or genes for immunoglobulin heavy change (IGH) in case of B cell
Background Tyrosine kinase inhibitors (TKIs) have demonstrated success in the treatment of acute lymphoblastic leukemia (ALL) in patients that express BCR-ABL rearrangements (Philadelphia chromosome [Ph]). The current study aimed to assess the efficacy of TKIs and prognostic factors in the treatment of adults with Ph+-ALL. Methods In this multicenter retrospective study, the relationship between Ph+-ALL and treatment outcomes among Chinese patients receiving TKI-containing induction/consolidation chemotherapy was examined. A total of 86 Ph+-ALL patients were included and followed for 3.85 (0.43-9.30) years. Overall survival (OS) and event-free survival (EFS) were analyzed. Results A total of 86 Ph+-ALL patients (40 females and 46 males; median age: 34.0 years) were enrolled, including those with BCR/ABL transcripts 190 (n = 52), 210 (n = 25), and 230 (n = 2); BCR/ABL isoform determination was not available for 7 patients. Thank you to everyone who has replied. About Candida - yes, I did read the link - and find it very interesting. I had a yeast infection for 18 months - and Candida for who knows how long. Im still waiting to talk to my gastroenterologist about the results. I havent had the chance to talk to him for even a second after the procedure. And, obviously, I am very anxious. Hopefully I will get a call from him soon. I spoke with my general practitioner - after emailing her the results - and she said that, it looks like refractory sprue. Im not sure how familiar you guys are with that so Im going to quote the section on it from the book, Celiac Disease by Peter Green. Ive been really good about the diet - I watch everything I eat. I do eat out a lot, but only where I know Im safe... But my last biopsy was still totally flat. I dont know what Im doing wrong or what more I can do. Maybe stop eating.
