TY - JOUR. T1 - Pathogenicity and sequence analysis studies suggest potential role of gene 3 in virulence of swine enteric and respiratory coronaviruses. AU - Paul, Prem S. AU - Vaughn, Eric M.. AU - Halbur, Patrick G.. PY - 1997/7/4. Y1 - 1997/7/4. N2 - Coronaviruses have been commonly associated with enteric and respiratory diseases. Two of the swine coronaviruses, namely transmissible gastroenteritis virus (TGEV) and porcine respiratory coronavirus (PRCV) have been extensively studied. TGEV replicates in both the enteric and respiratory tracts and causes enteric disease, whereas, PRCV replicates in the respiratory tract with limited to no replication in the enteric tract. We have isolated PRCV from swine herds with respiratory disease and have reproduced moderate pneumonia in gnotobiotic and conventionally reared pigs with two of the PRCV isolates. We have also identified two PRCV isolates with low virulence. One consistent difference that we have observed between PRCV isolates of different ...
Michaela P. Trudeau, Harsha Verma, Pedro E. Urriola, Fernando Sampedro, Gerald C. Shurson, and Sagar M. Goyal The pork industry has recently experienced the devastating effects of swine corona viruses. The three known porcine corona viruses are transmissible gastroenteritis virus (TGEV), porcine delta corona virus (PDCoV), and porcine epidemic diarrhea virus (PEDV). Each of these viruses causes severe diarrhea in young pigs and increased mortality, with subsequent reductions in growth performance and increased cost to producers. These viruses are excreted through feces and can be spread by contaminated equipment, personnel, and other fomites.. After the 2013 outbreaks of PEDV in the United States, some evidence suggested that PEDV can be transmitted via contaminated feed and feed ingredients. In order to understand the role of virus-contaminated feed on its transmission, it is necessary to determine the risk of virus survival in various commonly used feed ingredients in swine diets. Our research ...

I think the English Sweating disease provides a particularly good lesson for the affluent since it killed more rich people than poor folks. It was an especially nasty thing to come down with. Most people were dead within one day of infection and almost certainly within two days. It spread so fast that doctors were never even able to figure out what virus was causing it. Well, cest la vie or, rather, cest la morte.. Heres the hard, cold facts: Even if the "developed" world doesnt care anything about the fact that over two million mostly people of color die of treatable diarrheic diseases every year, sooner or later that attitude will kill us all but when the next really nasty (read: quickly fatal, incurable) viruses and bacteria crop up--which we know will happen and probably soon, at that--theres a very big chance that its going to take out all of the constant-lysol-spraying, antibiotic-resistant folks (who could have done a lot more to make healthcare a right for everyone) pretty ...

Porcine epidemic diarrhea virus (PEDV) is a highly contagious disease characterized by vomiting, diarrhea and inappetence in pigs of all ages. Mortality in young piglets is generally less common than in transmissible gastroenteritis (TGE).
The recent emergence of a number of new viral diseases as well as the re-emergence of tuberculosis (TB), indicate an urgent need for new drugs against viral and bacterial infections.. Coronavirus nsp1 has been shown to induce suppression of host gene expression and interfere with host immune response. However, the mechanism behind this is currently unknown. Here we present the first nsp1 structure from an alphacoronavirus, Transmissible gastroenteritis virus (TGEV) nsp1. Contrary to previous speculation, the TGEV nsp1 structure clearly shows that alpha- and betacoronavirus nsp1s have a common evolutionary origin. However, differences in conservation, shape and surface electrostatics indicate that the mechanism for nsp1-induced suppression of host mRNA translation is likely to be different in the alpha- and betacoronavirus genera.. Preliminary results from studies on immunity, directed by the National Pork Board, confirm "immunity does appear not to be very long lived," said Lisa Becton, director of swine health information for the board. The board has collected more than $2 million for research on PEDv.. …. Veterinarians and others have been unable to predict the duration of immunity to PEDv in hogs following exposure, in part because the disease had never been in the United States before last year.. Ackerman had thought hogs would have a natural immunity to PEDv for three years after being infected because that is the case for a similar disease called Transmissible Gastroenteritis. "Just because a farm broke with PEDv last year doesnt mean that they are protected from re-breaking with it this year," he said in a telephone interview.. Ackerman said he did not know why the female pigs, or sows, on the Indiana farm were re-infected after being exposed to the virus during the original outbreak last year. At the time, they ...

Common diseases - Gastroenteritis If your pet suddenly starts to vomit and has diarrhea, it may be suffering from gastroenteritis. It can occur following the ingestion of food your pet is not used to, of rotten food or of the contents of garbage cans. It is sometimes of infectious origin (viral such as parvovirus or bacterial such as salmonella for example).
Gastroenteritis due to a variety of organisms is one of the major causes of childhood mortality in the tropics. The substitution of the milk bottle for breast feeding is increasing rather than decreasing the problem. Mismanagement of artificial feeding is a serious cause, due to pathogenic coliform organisms.. Unwashed fruits contaminated with pathogenic organisms by flies are also a common cause of non-specific gastroenteritis in children and adults.. ...

Porcine epidemic diarrhea virus: current insights Chaochao Lv,1,* Yan Xiao,1,* Xiangdong Li,1 Kegong Tian,1,2 1National Research Center for Veterinary Medicine, Luoyang, People's Republic of China; 2College of Animal Science and Veterinary Medicine, Henan Agricultural University, Zhengzhou, People's Republic of China *These authors contributed equally to this review Abstract: Porcine epidemic diarrhea virus (PEDV), an enveloped, single-stranded, positive-sense RNA virus belonging to the genus Alphacoronavirus in the family Coronaviridae, can infect pigs of all ages and causes acute and watery diarrhea, vomiting, dehydration, and high mortality in neonatal piglets. This disease was first observed in England in 1971 and was subsequently reported in many swine-producing countries in Europe and Asia. In contrast to the situation in Europe, the disease has remained a major cause of diarrhea outbreaks on swine farms in Asia. Since late 2010, severe porcine epidemic diarrhea (PED) epizootics caused
Interest in porcine epidemic diarrhea has grown since the 2013 outbreak in the United States caused major losses, with mortality rates up to 100 % in suckling piglets. In Mexico, an outbreak of porcine epidemic diarrhea, characterized by 100 % mortality in piglets, began in March 2014 in the State of Mexico. The aim of this study was to confirm and identify porcine epidemic diarrhea virus (PEDV) in samples from piglets with suggestive clinical signs using virological, histological, and molecular techniques. Necropsy was performed on 13 piglets from two litters with initial and advanced clinical signs. Suggestive lesions of acute infection with PEDV were detected in histological sections of the small and large bowels; specifically, multiple virus particles with visible crown-shaped projections were observed using electron microscopy and negative staining. Viral isolation was performed in Vero cells with trypsin. Infection was monitored by observation of cytopathic effect, and titration was ...
In late 2013, outbreaks of porcine epidemic diarrhea virus (PEDV) infection recurred in South Korea. Genetic and phylogenetic analyses showed that isolates from the outbreaks were most closely related to emergent US strains of PEDV. These US strain-like PEDV variants are prevalent in South Korea and responsible for recent outbreaks in the country.
The USDA has reported another 296 cases of porcine epidemic diarrhea virus (PEDv) last week. That brings the number of cases in the last 10 months to 4,757. The disease was first reported in the U.S. hog population in May 2013, with animals in 27 states infected. Unfortunately, each case report can mean just one animal has been sickened, or an entire herd at one farm is sick. The virus, which is deadly for baby pigs, doesnt affect adult hogs. The disease has killed more than 4 million piglets in this country since May 2013. This is most likely going to cause dramatic price increases at the consumer level in the next year, since hog prices are at record highs. The USDAs estimate of hog slaughter on March 21, 2014 was 360,000, down 51,000 head from a week ago. Scientists say that the … [Read more...] ...

Results: In this study we constructed a recombinant baculovirus that was potent to express PEDV-S1 protein; PEDV-S1 protein (S1) which is essential to generate immunogenic VLPs in insect cells. A triplet cassette providing simultaneous and independent expression of S1 gene of PED virus was designed and subjected to synthesis. The cassette was then cloned into pFastBacHTb plasmid and then transformed in to competent Escherichia coli DH10Bac cells and the recombinant bacmids were produced following the homologous Tn7 site-specific transposition. This construction was verified by polymerase chain reaction (PCR) and then transfected into Sf9 insect cells to package new recombinant baculovirus expressing complex proteins of the interest. Restriction map of pFastBacHTb_PEDV-S1 plasmid confirmed the fidelity of the clone. The PCR carried out on the recombinant bacmids as template indicated that a proper homologous recombination has occurred between pFastBacHTb_PEDV-S1 donor plasmid and the bacmid. ...
