The peptide hormone gastrin binds two ferric ions with high affinity and iron binding is essential for the biological activity of non-amidated gastrins and in the presence of Bi3+ ions the affinity of Fe3+ ions for Ggly was substantially reduced; the data was better fitted by a mixed rather than a competitive inhibition model. gastric mucosal damage induced by non-steroidal anti-inflammatory drugs aspirin or alcohol has also been noted with bismuth salts. In the colon treatment with bismuth reduced acid-induced colitis in rats [4 5 and microscopic [6] and ulcerative [7] colitis in humans. The bismuth salt most commonly used for treatment of gastrointestinal conditions in medical practice in Australia is currently colloidal bismuth subcitrate. Pharmacological studies have demonstrated that following absorption bismuth binds to plasma proteins [8] and is distributed through most tissues [9]. Gastrin is a gastrointestinal peptide hormone that was originally identified as a stimulant of acid ...
1. Serum gastrin concentrations before and after a standardized meal were determined in twenty-eight patients with duodenal ulcer and in ten normal control subjects. 2. In response to pentagastrin, thirteen of the duodenal ulcer subjects secreted acid within the limits of normal and fifteen secreted in excess. 3. The differences in the basal serum gastrin concentrations between the three groups, normal subjects, acid
Takaishi et al suggest that gastrin may serve as a rheostat for the stomach. Gastrin likely plays a central role in the safety network for the protection from mucosal damages caused by gastric acid secretion induced by gastrin itself, and thus either too much or too little gastrin can predispose to carcinogenesis. Therefore, clinicians in the future may need to be more circumspect when prescribing with acid suppressive drugs, such as proton-pump inhibitors, for a long-term use in patients infected with Helicobacter pylori. In future studies, Dr. Wang and colleagues plan to study host factors other than gastrin [that] are also ¦ important for Helicobacter-associated gastric carcinogenesis. These include specific cytokines and chemokines induced by Helicobacter infection, and modulated by gastrin, that link inflammation and cancer. In addition, they plan to study the role of other non-Helicobacter bacteria that colonize the stomach when acid secretion is suppressed, since bacterial overgrowth ...

Hypergastrinemia in INS-GAS mice leads to accelerated carcinogenesis of the stomach, but the mechanisms have not been well defined. We investigated the possible role of gastrin-induced gastric cell apoptosis in the development of gastric cancer. We examined apoptosis and the expression of Bcl-2 family proteins in INS-GAS mice of different ages, as well as in gastrin-deficient (GAS-KO) mice after gastrin-17 (G-17) infusion. In addition, we studied the effects of the gastrin/cholecystokinin-2 (CCK-2) receptor antagonist YF476 and/or histamine H2 (H-2) receptor antagonist loxtidine on apoptosis and atrophy in INS-GAS mice with or without Helicobacter felis (H. felis) infection. INS-GAS mice had age-associated increases in Bax protein expression and decreases in Bcl-2 protein expression, along with increased glandular and epithelial cell apoptosis. At 8-week gastrin infusions in GAS-KO mice resulted in a similar pattern of altered Bax and Bcl-2 expression, followed by gastric cell apoptosis. H. felis
OBJECTIVES: In addition to the acid-stimulatory gastrins, progastrin also release N-terminal fragments. In order to examine the cellular content, secretion and peripheral metabolism of these fragments, we developed an immunoassay specific for the N-terminal sequence of human progastrin.. RESULTS: The concentration of N-terminal progastrin fragments in human antral tissue was 6.7 nmol/g tissue (n=5), which was only half of that of acid-stimulatory gastrins (12 nmol/g tissue). Gel chromatography of antral extracts showed that the progastrin fragment 1-35 and 1-19 constitute the major part of the N-terminal progastrin fragments. The basal concentration of N-terminal fragments in normal human plasma was almost 30-fold higher than that of the amidated, acid-stimulatory gastrins (286 pmol/l versus 9.8 pmol/l, n=26, P,0.001). In contrast, the concentration of N-terminal fragments in hypergastrinemic plasma was only 2.7-fold higher than the concentration of amidated gastrins (540 pmol vs. 198 pmol/l, ...
Antibodies to the antral hormone gastrin have been induced in the rabbit and detected by passive hemagglutination. Specificity of the antibody, as determined by three methods, is directed to gastrins I and II, gastrin pentapeptide, and gastrin tetrapeptide, as well as to the stage-1 gastrin used for immunization.
article{39e7a8a0-7c43-4864-95ec-7e69e2f10374, abstract = {Background: The ECL cells are histamine-producing endocrine cells in the oxymic mucosa that synthesize and secrete proteins and peptides. They are the primary target for gastrin and mediate the control of gastrin on acid secretion and oxyntic mucosal growth. Knowledge of the molecular biology of the ECL cell is therefore important for understanding gastric physiology. Accordingly, we wanted to identify genes that are characteristically expressed in the ECL cells and controlled by gastrin. Methods: Using Affymetrix GeneChips((R)), RNA expression profiles were generated from ECL cells isolated by counterflow elutriation from hyper- or hypogastrinemic rats. Contamination from non-endocrine cells was eliminated by subtraction of the expression profiles of the fundic and antral mucosa. Results: The expression of 365 genes was ECL cell characteristic. Gastrin was found to control the expression of 120 which could be divided into two major ...

Oral NaCl produces stronger natriuresis and diuresis as compared with venous infusion of same amount of NaCl, indicating the existence of renal-gastric axis. Although numerous hormones are secreted in gastrointestinal tract, gastrin is evident one due to its natriuretic effects and taken-up by the renal proximal tubule (RPT) cells. We hypothesize that there is an interaction between gastrin and dopamine receptor in kidney, which synergistically increases sodium excretion, the impaired interaction would be involved in the pathogenesis of hypertension. In WKY rats, infusion of gastrin, via renal artery, induced natriuresis and diuresis, which was blocked in the presence of CI988, a gastrin receptor blocker. Similarly, the natriuretic and diuretic effect of fenoldopam, a D1-like receptor agonist, was blocked by the D1-like receptor antagonist, SCH23390, indicating that gastrin and fenoldopam, via individual receptor, play natriuretic and diuretic effects. Our further study found that lower dosages ...

acetylcholine (ACh; M3 receptor), and histamine (H2 receptor). Acid is secreted across the parietal cell canalicular membrane by the H+/K+ ATPase proton pump into the gastric lumen. The gastrin that is secreted by antral G cells in response to intraluminal dietary peptides acts directly on parietal cells and also stimulates release of histamine from enterochromaffin-like (ECL) cells. The vagus nerve stimulates postganglionic neurons of the enteric nervous system to release acetylcholine ACh, which acts on parietal and ECL cells. In the antrum, release of gastrin-releasing peptide (GRP) from postganglionic neurons directly increases gastrin release, whereas release of ACh indirectly increases gastrin secretion by inhibiting release of somatostatin from antral D ... ...

Results:. Esophagitis recurred in 47% of the patients receiving 20 mg of omeprazole daily, but all rehealed after the dose was doubled. Seven of 40 patients (18%) had a second relapse after a mean follow-up time of 24 months (range, 9 to 36 months) that was successfully treated with a further 20-mg dose increment for a mean period of 36 months (range, 6 to 39 months). Median gastrin levels increased initially from 60 ng/L before study entry to 162 ng/L (P , 0.01) with treatment and reached a plateau during maintenance treatment. Very high gastrin levels (,500 ng/L) were observed in a subgroup (11%) of patients. The incidence of micronodular hyperplasia increased from 2.5% of the patients at first biopsy to 20% at the last biopsy (P = 0.001), with a corresponding progression of gastritis to subatrophic or atrophic gastritis from less than 1% to 25% (P , 0.001), which was more pronounced in patients with very high serum gastrin levels. ...

34 Part of the human digestive (gastrointestinal) tract. (a) The parietal cells and chief cells of the gastric glands secrete their products in response to the hormone gastrin. Pepsin begins the process of protein degradation in the stomach. (b) The cytoplasm of exocrine cells is completely filled with rough endoplasmic reticulum, the site of synthesis of the zymogens of many digestive enzymes. The collecting ducts ultimately lead to the pancreatic duct and thence to the small intestine. (c) Amino acids are absorbed through the epithelial cell layer (intestinal mucosa) of the villi and enter the capillaries. Recall that the products of lipid hydrolysis in the small intestine enter the lymphatic system after their absorption by the intestinal mucosa. ...

Fasting serum gastrin, secretin stimulation test, and gastric acid secretion (basal acid output) studies have been used for diagnosis of ZES. The gastric acid secretion test is no longer used routinely.. Fasting serum gastrin should be performed in any patient suspected of having ZES. The combination of a fasting serum gastrin concentration higher than 1000 pg/mL (about 10 times the upper limit of normal) and an intragastric pH below 2.5 is virtually diagnostic of the disease. Measurement of gastric pH on a single specimen is important to exclude secondary hypergastrinemia due to achlorhydria. About two-thirds of patients with ZES have serum gastrin concentrations between 150 and 1000 pg/mL (mildly to moderately elevated). The secretin stimulation test should be used to evaluate these patients.. If fasting serum gastrin is not diagnostic, the secretin stimulation test should be used to differentiate patients with ZES from those with other causes of hypergastrinemia (G-cell hyperplasia, gastric ...
Result Gastrin infusing WKY rats via renal artery induced natriuresis and diuresis, which was blocked in the presence of CI988, a gastrin receptor blocker. Similarly, effect hereinbefore of fenoldopam, a D1-like receptor agonist, was blocked by D1-like receptor antagonist, SCH23390, indicating gastrin and fenoldopam play natriuretic and diuretic effect through individual receptors. Lower dosages of gastrin or fenoldopam failed to induce natriuresis and diuresis alone, while putting together induced the effects. The above-mentioned effects were lost in SHRs. Natriuresis and diuresis was partially blocked by SCH23390 or CI988, indicating the interaction between gastrin and D1-like receptor. Stimulation of either receptor increased the expression of the other and inhibited Na+-K+-ATPase activity, while the inhibitory effect of Na+-K+-ATPase activity was partially blocked through its corresponding receptors due to respective existence of SCH23390 and CI988.. ...
Traditionally, gastric carcinoid type 1 (GCA1s) are endoscopically or surgically removed, depending on the number, appearance and size of the tumors. Antrectomy, with surgical excision of the majority of the G cells, is thought to facilitate regression of these tumors by removing the source of excessive gastrin secretion; however, the long-term benefits of antrectomy still remain uncertain. Although proton pump inhibitors are effective in reducing hypergastrinemia-induced gastric acid hypersecretion in GCA2, they do not affect ECL-cell hyperplasia, and therefore their role in GCA1 is limited. Moreover, in selected cases, significant reduction of hypergastrinemia does not prevent development of ECL carcinoid, suggesting that, in addition to hypergastrinemia, other pathogenic or genetic factors may be involved. Treatment with somatostatin analogues (SSA) might impede ECL-cell hyperplasia by suppressing gastrin secretion and/or by a direct anti-proliferative effect on ECL cells. Treatment with SSAs ...

Introduction: The diagnosis of Zollinger-Ellison syndrome is suspected in patients with symptoms of gastric acid hypersecretion, i.e., peptic ulcer disease, malabsorption or diarrhea, or with elevated fasting serum gastrin levels. However, symptoms can be masked by the use of proton pump inhibitors and fasting serum gastrin values are not always conclusive. Therefore, the secretin stimulation test is advocated as the principal diagnostic tool to identify the Zollinger-Ellison syndrome ...

Gastric cancer, a disease with a reduced frequency for decades, now appears to be on the rise again in young Americans. The epidemiology of gastric cancer differs between tumors in the cardia and those of the more distal parts of the stomach. The tumors are divided into the intestinal type showing glandular growth pattern and the diffuse type with a different pattern. The latter often expresses neuroendocrine and more specifically ECL-cell markers suggesting that they originate from the ECL cell, the target cell for the antral hormone, gastrin. Helicobacter pylori gastritis is accepted as the major cause of gastric cancer, but only after having induced oxyntic atrophy which reduces gastric acid secretion and thus induces hypoacidity leading to hypergastrinemia. Long-term hypergastrinemia is known to induce malignant neoplasia in the stomach of animals as well as man. Recently treatment with proton pump inhibitor after Helicobacter pylori eradication in patients with gastroesophageal reflux ...

The mechanisms regulating the proliferation and differentiation processes that give rise to and maintain the gastric epithelium have not yet been completely elucidated.. In the present studies, in vitro models were established and the influence of growth factors and extracellular matrix proteins on these processes were investigated. Pentagastrin and hydrocortisone were found to accelerate the development of H,KTPase-positive parietal cells and other epithelial cells from undifferentiated gastric epithelial cells of foetal rats. These undifferentiated cells and also presumably immature epithelial cells in the progenitor zone of adult gastric glands were shown to express cholecystokinin-2 (CCK2) receptors and are therefore targets for the trophic action of gastrin.. H,K-ATPase-positive parietal cells in the progenitor zone of adult glands were also found to express CCK2 receptors, indicating that gastrin may stimulate maturation of the parietal cell lineage even during adult life. The G-protein coupled receptor 56 (GPR56) is known to regulate cell adhesion, migration, proliferation and stem cell biology, but its expression in the gut has not been studied. We hypothesized that the promotion of colorectal cancer by progastrin may be mediated in part through GPR56. Here, we found that GPR56 expresses in rare colonic crypt cells that lineage trace colonic glands consistent with GPR56 marking long-lived colonic stem-progenitor cells. GPR56 was upregulated in transgenic mice overexpressing human ...
The gastrin releasing peptide receptor (GRP-R) is over-expressed on several tumors and cancer cell lines including pancreas, prostate, breasts, gastrointestinal and small cell lung cancer (SCLC). targeted cells is usually a prerequisite to triggering effective internalization. For restorative reasons, efficient internalization from the radioconjugate is apparently important in providing the best radioactivity dose towards the targeted cells. Structural adjustments to these radiopharmaceuticals possess included variance in the peptide series, the nature from the chelate and radiometal, and the space from the spacer attaching both components (Physique 1). For instance, an agonist BBN AUY922 analogue, Perform3A-CH2CO-G-4-aminobenzoyl-Q-W-A-V-G-H-L-M-NH2 (AMBA) was synthesized and tagged with 177Lu for prostate malignancy focusing on (22). 177Lu-AMBA displays a higher affinity for the GRP-R (IC50 = 2.50 0.50 nM) and 76.8% internalization. Bombesin is a 14 amino acid peptide originally isolated from the skin of a frog. It has two known homologues in mammals called neuromedin B and gastrin releasing peptide. It works on the gastrointestinal tract neuroendocrine hormone and it stimulates gastrin release from G cells. It activates three different G-protein coupled receptors known as BBR1 2 & 3. It also activates these receptors in the brain. Together with cholecystokinin it is the second major source of negative feedback signals that stop eating behaviour. Bombesin is also a tumor marker for gastric cancer and neuroblastoma. ...
