Hemozoin is generally considered a waste deposit that is formed for the sole purpose of detoxification of free heme that results from the digestion of hemoglobin by Plasmodium parasites. However, several observations of parasite multiplication, both in vertebrate and invertebrate hosts are suggestive of a wider, but overlooked, metabolic role for this product. The presence of clinical peripheral blood samples of P. falciparum with high parasitemia containing only hemozoin-deficient (non-pigmented) asexual forms has been repeatedly confirmed. Such samples stand in contrast with other samples that contain mostly pigmented circulating trophozoites and gametocytes, indicating that pigment accumulation is a prominent feature of gametocytogenesis. One of the most studied phosphoinositides is phosphatidylinositol 4,5-bisphosphate (PIP2), which localizes to the plasma membrane, nuclear speckles, small foci in the nucleoplasm, and to the nucleolus in mammalian cells. Here, we show that PIP2 also localizes to the nucleus in prophase I, during the gametogenesis of C. elegans hermaphrodite. The depletion of PIP2 by type I PIP kinase (PPK-1) kinase RNA interference results in an altered chromosome structure and leads to various defects during meiotic progression. We observed a decreased brood size and aneuploidy in progeny, defects in synapsis, and crossover formation. Abstract: Megagametogenesis has been identified in the seagrass Halophila johnsonii, a species with no known seeds, using DAPI staining and manual sectioning methods. Developmental stages were documented and compared with megagametogenesis stages in Halophila decipiens, a related species widely reported to produce viable seeds. Present observations suggest that meiosis occurs in the megasporocyte and, therefore, sexual reproduction should be possible in H. johnsonii. Results of this study, which was part of a multi-level threatened-species recovery plan sponsored by NOAA, are encouraging and suggest that two additional management-related inquiries, 1) the search for male reproductive structures and 2) hybridization with related species are worthwhile. The Retinoblastoma (Rb) protein is a conserved repressor of cell proliferation. In animals and plants, deregulation of Rb protein causes hyperproliferation and perturbs cell differentiation to various degrees. However, the primary developmental impact of the loss of Rb protein has remained unclear. In this study we investigated the direct consequences of Rb protein knockout in the Arabidopsis male germline using cytological and molecular markers. The Arabidopsis germ line derives from the unequal division of the microspore, producing a small germ cell and a large terminally differentiated vegetative cell. A single division of the germ cell produces the 2 sperm cells. We observed that the loss of Rb protein does not have a major impact on microspore division but causes limited hyperproliferation of the vegetative cell and, to a lesser degree, of the sperm cells. Environmental conditions influence the costs and benefits of various life history strategies, so different strategies may be favored across geographic gradients. We describe latitudinal patterns in population density, body size, and gamete production in the clonal sea anemone Diadumene lineata. Fission rate in this species is highly correlated with temperature, so the rate of asexual reproduction declines with latitude. We measured the relationships among body size, gamete production, and environmental variables using individuals from 20 sites in intertidal estuaries and harbors from Florida to Maine across the US Atlantic and Gulf coasts. The history section was well formatted, however more information would have been appreciated as there didnt seem to be much of the actual history, more of a timeline of the frog. Malaria pathogenesis relies on sexual gametocyte forms of the malaria parasite to be transmitted between the infected human and the mosquito host but the molecular mechanisms controlling gametocytogenesis remains poorly understood. Here we provide a high-resolution transcriptome of Plasmodium falciparum as it commits to and develops through gametocytogenesis. The gametocyte-associated transcriptome is significantly different from that of the asexual parasites, with dynamic gene expression shifts characterizing early, intermediate and late-stage gametocyte development and results in differential timing for sex-specific transcripts. The transcriptional dynamics suggest strict transcriptional control during gametocytogenesis in P. falciparum, which we propose is mediated by putative regulators including epigenetic mechanisms (driving active repression of proliferation-associated processes) and a cascade-like expression of ApiAP2 transcription factors.
