Human beta defensins (hBDs) are small cationic peptides, expressed in mucosal epithelia and important agents of innate immunity, act as antimicrobial and chemotactic agents at mucosal barriers. In this perspective, we present evidence supporting a novel strategy by which the oral bacterium Fusobacterium nucleatum induces hBDs and other antimicrobial peptides (AMPs) in normal human oral epithelial cells and thereby protects them from other microbial pathogens. The findings stress (1) the physiological importance of hBDs, (2) that this strategy may be a mechanism that contributes to homeostasis and health in body sites constantly challenged with bacteria and (3) that novel properties identified in commensal bacteria could, one day, be harnessed as new probiotic strategies to combat colonization of opportunistic pathogens. We studied the microbiology of pus from 10 infected sinuses and their corresponding intracranial abscess (IA) (22).BA and SE was present in 5 patients, and both BA and SE were present in one. Four of the patients were children and all had SE. Five had Polymicrobial flora was found in 9 sinuses and 8 IA. A total of 26 isolates (2.6 isolates per specimen, 19 anaerobic, 7 aerobic or facultative and 1 microaerophilic) were recovered from the sinuses, and 17 isolates (1.7 isolates per site, 13 anaerobic, 2 aerobic or facultative and 2 microaerophilic) were found in the IA. PURPOSE: To report the case of a previously healthy young female who developed orbital cellulitis caused by Fusobacterium necrophorum. We are unaware of previous reports of this condition caused by the same anaerobic, gram-negative, nonsporeforming bacterium and could find no reference to it in a computer search using MEDLINE. METHODS: Case report. RESULTS: In the case of orbital cellulitis presented here, the patient required 3 sinus debridement operations and 30 days of intravenous antibiotics. A new genus of cellulolytic, gram-negative, nonsporeforming, anaerobic bacteria is described. The colonies produced by these bacteria on cellulose agar were round, clear, translucent, and cream-colored and had an undulate margin. Single cells of the bacterium were straight to slightly curved rods 0.5 to 0.8 μm wide by 4 to 10 μm long and were motile by means of a single flagellum located one-third of the distance from the end of the cell. Among the various substrates tested, only cellulose, cellobiose, and salicin were able to support growth. The major fermentation products of cellobiose or cellulose degradation were acetic acid, hydrogen, and carbon dioxide. The deoxyribonucleic acid base composition of the type species was 38 mol% guanine plus cytosine. The name Acetivibrio is proposed for this new genus, which is placed in the family Bacteroidaceae. Bacteroides coprosuis Whitehead et al. 2005 belongs to the genus Bacteroides, which is a member of the family Bacteroidaceae. Members of the genus Bacteroides in general are known as beneficial protectors of animal guts against pathogenic microorganisms, and as contributors to the degradation of complex molecules such as polysaccharides. B. coprosuis itself was isolated from a manure storage pit of a swine facility, but has not yet been found in an animal host. The species is of interest solely because of its isolated phylogenetic location. The genome of B. coprosuis is already the 5th sequenced type strain genome from the genus Bacteroides. Summary An agglutination assay for detecting intermicrobial adherence between the cells of Candida albicans and various oral bacteria is described. Strains of Streptococcus sanguis, S. salivarius, S. mutans, S. mitis, Fusobacterium nucleatum and Actinomyces viscosus all coagglutinated with C. albicans. No interaction could be demonstrated between the cells of Bacteroides melaninogenicus and those of C. albicans. Preliminary investigations of these interactions suggest that binding of F. nucleatum and A. viscosus to C. albicans is mediated by bacterial proteins, possibly lectins. Other mechanisms must account for the binding of oral streptococci to C. albicans. The possible implications of these findings in relation to oral mucosal colonisation and oral candidal clearance are discussed.
Streptococcus gordonii is one of several species that can initiate the formation of oral biofilms that develop into the complex multispecies microbial communities referred to as dental plaque. It is in the context of dental plaque that periodontal pathogens such as Porphyromonas gingivalis cause disease. We have previously reported a whole cell quantitative proteomics investigation of P. gingivalis in a model dental plaque community of S. gordonii, P. gingivalis, and Fusobacterium nucleatum. Here we report the adaptation of S. gordonii to the same model. 1122 S. gordonii proteins were detected in S. gordonii control samples, 915 in communities with F. nucleatum, 849 with P. gingivalis, and 649 with all three organisms. Quantitative comparisons showed extensive proteome changes in association with F. nucleatum or P. gingivalis individually or both P. gingivalis and F. nucleatum together.
