Functional neuroimaging data embodies a massive multiple testing problem, where 100,000 correlated test statistics must be assessed. The familywise error rate, the chance of any false positives is the standard measure of Type I errors in multiple testing. In this paper we review and evaluate three approaches to thresholding images of test statistics: Bonferroni, random field and the permutation test. Owing to recent developments, improved Bonferroni procedures, such as Hochbergs methods, are now applicable to dependent data. Continuous random field methods use the smoothness of the image to adapt to the severity of the multiple testing problem. Also, increased computing power has made both permutation and bootstrap methods applicable to functional neuroimaging. We evaluate these approaches on t images using simulations and a collection of real datasets. Little is known about whether residual cognitive function occurs in the earliest stages of brain injury. The overarching goal of the work presented in this dissertation was to elucidate the role of functional neuroimaging in assessing brain activity in critically ill patients. The overall objective was addressed in the following four empirical chapters: In Chapter 2, three versions of a hierarchically-designed auditory task were developed and their ability to detect various levels of auditory language processing was assessed in individual healthy participants. The same procedure was then applied in two acutely comatose patients. In Chapter 3, a hierarchical auditory task was employed in a heterogeneous cohort of acutely comatose patients. Math anxiety is a relatively frequent phenomenon and often related to dyscalculia. Thus, a standardised instrument to detect math anxiety as early as possible would be useful. The FRA is the first German math anxiety questionnaire for primary school children (original version: Thomas & Dowker, 2000). Objectives: Is the FRA a reliable and valid instrument? Are there any gender differences concerning math anxiety? Are there any developmental changes in this regard in the course of the early grades? Methods: Together with the dyscalculia test TEDI-MATH (Kaufmann et al., in preparation), the FRA was presented to a total of 450 children from the first to the third grade of primary school (at least 40 girls and 40 boys per semester). Results: The total scale has an internal consistency (Cronbachs alpha) between 0.83 and 0.91. Correlations between arithmetic skills and the FRA scales were mostly significant. The human pulvinar is the largest thalamic area in terms of size and cortical connectivity. Although much is known about regional pulvinar structural anatomy, relatively little is known about pulvinar functional anatomy in humans. Cooccurrence of experimentally induced brain activity is a traditional metric used to establish interregional brain connectivity and forms the foundation of functional neuroimaging connectivity analyses. Because functional neuroimaging studies report task-related coactivations within a standardized space, meta-analysis of many whole-brain studies can define the brains interregional coactivation across many tasks. Such an analysis can also detect and define variations in functional coactivations within a particular region. Functional neuroimaging studies have provided strong support for a critical role of the amygdala in emotional processing. However, several controversies remain in terms of whether different factors-such as sex, valence and stimulus type-have an effect on the magnitude and lateralization of amygdala responses. To address these issues, we conducted a meta-analysis of functional neuroimaging studies of visual emotional perception that reported amygdala activation. Critically, unlike previous neuroimaging meta-analyses, we took into account the magnitude (effect size) and reliability (variance) associated with each of the activations. Our results confirm that the amygdala responds to both positive and negative stimuli, with a preference for faces depicting emotional expressions. We did not find evidence for amygdala lateralization as a function of sex or valence. Instead, our findings provide strong support for a functional dissociation between left and right amygdala in terms of temporal dynamics. ...

Degree, which is the number of connections incident upon a node, measures the relative importance of the node within a network. By computing degree metrics in voxel-wise functional brain networks, many studies performed high-resolution mapping of brain network hubs using resting-state functional magnetic resonance imaging. Despite its extensive applications, defining nodes as voxels without considering the different sizes of brain regions may result in a network where the degree cannot accurately represent the importance of nodes. In this study, we designed a data-driven method to reduce this impact of the region size in degree metrics by (1) disregarding all self-connections among voxels within the same region and (2) regulating connections from voxels of other regions by the sizes of those regions.
