Geneticists have argued that the linear decay in within-population genetic diversity with increasing geographic distance from East Africa is best explained by a phylogenetic process of repeated founder effects, growth, and isolation. However, this serial founder effect (SFE) process has not yet been adequately vetted against other evolutionary processes that may also affect geospatial patterns of diversity. Additionally, studies of the SFE process have been largely based on a limited 52-population sample. In this modestly updated article, originally published in Human Biology in 2016 (vol. 88, no. 3, pp. 219-231), we assess the effects of founder effect, admixture, and localized gene flow processes on patterns of global and regional diversity using a published data set of 645 autosomal microsatellite genotypes from 5,415 individuals in 248 widespread populations. We used a formal tree-fitting approach to explore the role of founder effects. The approach involved fitting global and regional population
Consider the view of Y Combinator, one of the leading incubators of early-stage technology start-ups. Their strategy is based on the belief that successful start-ups rarely, if ever, capitalize on their initial idea. Ideas transform. So its not ideas that matter the most. Far more important than the quality of an idea is the quality of the team. A good team can make a start-up successful because it can discover a good idea by learning how a market works and then do the work to implement the idea. A good team will divide and distribute the labor in a way that takes advantage of individual skills. Y Combinator avoids investing in start-ups that have a single founder not only because a single founder means theres no team to divide up the labor. They avoid single founders for a reason that isnt obvious and yet is fundamental to teamwork: Single founders lack the esprit de corps that prevents individuals from letting their friends down. Teams work harder when things arent going well because ...

There are potential benefits to testing individuals at high risk for carrying a BRCA1 or BRCA2 mutation. However, there are also substantial costs associated with this testing, exceeding $2000 for the combination of genetic counseling and testing. The major expense for this combination is the genetic test itself, for which we used the estimated cost of full gene sequencing. At present, there are ,400 known or suspected deleterious mutations identified for the BRCA1 and BRCA2 genes.5 Many families harbor private mutations that have never been reported but are known to be deleterious. Full gene sequencing is considered to be the most sensitive method of detecting these mutations (34) . However, in certain populations, a few founder mutations appear to account for the majority of detectable alterations in BRCA1 and/or BRCA2. For example, common founder mutations have been reported in individuals of Ashkenazi Jewish (8) , Icelandic (35) , or French Canadian (36) descent. Less expensive tests to ...
Delvecchio M, Ludovico O, Bellacchio E, Stallone R, Palladino T, Mastroianno S, Zelante L, Sacco M, Trischitta V, Carella M. MODY type 2 P59S GCK mutant: founder effect in South of Italy.. Mutations in the glucokinase (GCK) gene are the most frequent cause of maturity onset diabetes of the young (MODY) in Italy. We evaluated GCK mutations in 32 unrelated patients younger than 18 years who had been diagnosed with MODY. Eleven different GCK heterozygous mutations were identified in 22 (68.7%) of the 32 probands. Nine mutations were missense and two were nonsense. Three of these mutations (E17X, P59S and E372X) have not been described previously and were shown to be associated with hyperglycaemia. Several prediction methods suggested that the E17X and E372X mutations result in a premature truncated protein and that the P59S mutation is pathogenic. This idea was further supported by evidence suggesting that Proline 59 is a highly conserved amino acid residue and that the P59S mutation does not ...
The current challenge in biomedical research is to detect genetic risk factors involved in common complex diseases. The power to detect their role is generally poor in populations that have been large for a long time. It has been suggested that the power may be increased by taking advantage of the specificity of founder populations: linkage disequilibrium spanning larger regions and kinship coefficients being stronger than in large populations. A new method is proposed here, the Maximum Identity Length Contrast (MILC) which, in contrast with other existing methods, does not make the assumption of unique ancestry for the genetic risk factors. It is thus appropriate for a search for common genetic risk factors for complex diseases. Statistical properties of the method are discussed in realistic contexts.

After the same splice site mutation in the MLH1 gene (c.589-2A,G) had been observed in four ostensibly unrelated American families with typical LS cancers, its occurrence in comprehensive series of LS cases (Mayo Clinic, Germany and Italy) was determined. It occurred in 10 out of 995 LS mutation carriers (1.0%) diagnosed in the Mayo Clinic diagnostic laboratory. It did not occur among 1,803 cases tested for MLH1 mutations by the German ...

Quite frankly, if I were on the Founders College team, Id read Noumenalselfs criticism and take it to heart. Noumenalself pointed out several problems in their business plan (or at bare minimum, the communication of their business plan to people who should be natural supporters), and he did it for free. Founders has yet to publicly release a concrete selling position that defines their educational mission and how they plan to achieve it. Perhaps the Founders idea is still too early in the planning stages, but at minimum, its principals should be offering statements defining their grand vision and why they are the people to realize it. It seems almost elementary that they would do so, so why the absence ...

The initial limitation of the technique described here is that it does not take into account other factors that might play a critical role in the spread of different viral variants globally. In particular, the authors claim that increasing frequency of the G614 variant is due to higher transmissibility. While this is supported by potentially higher viral loads in patients infected with the G614 variant, there are other equally plausible explanations for the observed patterns. One of these is a pervasive founder effect, wherein the G614 variant simply increased in frequency due to random chance after introduction into Europe from China, and was then reinforced by repeated introductions from Europe to other countries around the world. Without controlling for these other neutral factors through more complex modeling, many of the claims made in the paper on the emergence of dominant strains are unfounded. Although structural modeling is an interesting tool added to this analysis pipeline, it ...

Most startup founders have a tough road to their first round of funding, but the founders of Digital Brain had it a bit tougher than most. The two young founders survived by entering and winning hackathons to pay their rent and put food on the table. One of the ideas they came up with at those hackathons was DigitalBrain, a layer that sits on top of customer service software like Zendesk to streamline tasks and ease the job of customer service agents.
