Binding of folate (pteroylglutamate) and 5-methyltetrahydrofolate, the major endogenous form of folate, to folate binding protein purified from cows milk was studied at 7°C to avoid degradation of 5-methyltetrahydrofolate. Both folates dissociate rapidly from the protein at pH 3.5, but extremely slowly at pH 7.4, most likely due to drastic changes in protein conformation occurring after folate binding. Dissociation of 5-methyltetrahydrofolate showed no increase at 37°C suggesting that protein-bound-5-methyltetrahydrofolate is protected against degradation. Binding displayed two characteristics, positive cooperativity and a binding affinity that increased with decreasing concentrations of the protein. The α human folate receptor (αhFR), or KB cell folate receptor, gene contains two major promoters that produce transcripts, KB1 and KB4, varying only in the length and sequence of their 5′ untranslated regions (UTRs). Using RNase protection assays specific for each isoform, we show that the level of expression of these two transcripts is tissue-specific, indicating that promoter usage is regulated, not constitutive. RNA stabilities and translational efficiencies of the KB1 and KB4 transcripts were compared to determine the functional significance of the different 5′ UTRs. Analyses of RNA turnover in vivo with actinomycin D to block new transcription and in vitro with a cytoplasmic extract indicate no discernible differences in the stabilities of the two transcripts. Previous experiments indicated that secreted (s) and membrane (m) forms of folate binding protein (FBP) are present in the intrauterine environment of the pig. Our previous studies indicated that the two forms were produced sequentially; the secreted form was present in the intrauterine glands until Day 20 of gestation, whereas binding analysis indicated that folate binding increased dramatically in placental membranes until Day 50 of gestation. However, the cell types expressing mFBP have not been investigated. In this experiment, uterine wall sections from Day 20, 35, 50, 70, 90, and 105 of gestation were collected at slaughter and fixed, and subjected to in situ hybridization analysis for mFBP expression. Folate-protein conjugates have been shown to bind to and enter HeLa and KB cells by receptor-mediated endocytosis [Leamon and Low (1991) Proc. Natl. Acad. Sci. U.S.A. 88, 5572-5576]. Although these cells contain a membrane folate-binding protein (FBP) involved in the uptake of free folate, no studies have been conducted to evaluate whether the folate-protein conjugates enter cells via the same protein. To address this issue, HeLa cell monolayers were treated with folate-labelled 125I-RNAase under various conditions characteristic of FBP-mediated folate uptake. Activated macrophages have been functionally grouped according to their response to pro-Th1/proinflammatory stimuli [lipopolysaccharide, IFNγ, granulocyte macrophage colony-stimulating factor (GM-CSF); M1] or pro-Th2/anti-inflammatory stimuli [interleukin (IL)-4, IL-10, M-CSF; M2]. We report that folate receptor β (FRβ), encoded by the FOLR2 gene, is a marker for macrophages generated in the presence of M-CSF (M2), but not GM-CSF (M1), and whose expression correlates with increased folate uptake ability. 4215 Pemetrexed (PMX, Alimta ®) is a new generation antifolate with activity in a variety of solid tumors. It is an excellent substrate for the reduced folate carrier (RFC) and folate receptor-α (FR-α). The role of FR-α in PMX pharmacological activity is uncertain. While high-level expression may enhance the activity of this agent, it is not clear as to what role constitutive levels of this transporter contribute to PMX activity. In this study, constitutive expression of FR-α was completely abolished by siRNA- induced silencing in HeLa cells and in RFC-null HeLa R5 cells as confirmed by Northern blotting, immuno-histochemistry, and cell surface specific [3H]folic acid binding. PMX growth inhibition over six days was unchanged in HeLa and R5 cells in the absence of FR-α expression. The vitamin folic acid has served as a promising targeting ligand allowing selective delivery of attached radionuclides suitable for imaging of the diseased sites and for therapeutic application. A large number of folate radioconjugates with variable chemical structures have been developed over the last 25 years. Accumulation of radioactivity in healthy organs and tissues was always seen in the kidneys due to the expression of the FR in the proximal tubule cells. In some cases unspecific uptake of radiofolates was also seen in the liver and the intestinal tract. In this case-control and case-only analysis of epithelial ovarian cancer, multivitamin intake was associated with a lower risk of ovarian cancer regardless of FRα expression level. Among women who developed ovarian cancer, however, multivitamin intake was associated with a 3-fold increased risk of developing a strong-expressing FRα tumor. This is further complicated by the notion of polarized macrophages as well as a number of different macrophage phenotypes (which can be changed by signaling from one the same macrophage) that are considered proinflammatory and anti inflammatory. I have questioned experts in the field since I am working on a folate receptor conjugate that can be used to externally image macrophages in atheroma , potentially stratifying fibrous from vulnerable plaques.
