Focal dermal hypoplasia is a genetic disorder that primarily affects the skin, skeleton, eyes, and face. Most individuals with this condition are female. Males usually have milder signs and symptoms than females. Although intelligence is typically unaffected, some individuals have intellectual disability. This condition is caused by mutations in the PORCN gene and is inherited in an X-linked dominant manner. Most cases of focal dermal hypoplasia in females result from new mutations in the PORCN gene and occur in people with no history of the disorder in their family. When focal dermal hypoplasia occurs in males, it always results from a new mutation in this gene that is not inherited ...
Focal dermal hypoplasia (FDH) is an uncommon genetic disorder characterized by distinctive skin abnormalities and a wide variety of defects that affect the eyes; teeth; and skeletal, urinary, gastrointestinal, cardiovascular, and central nervous systems. It is usually, but not always, X-linked dominant (lethal in males).
Aicardi syndrome is a sporadic X-linked dominant, presumably male-lethal, neurodevelopmental disorder. It was initially characterized by agenesis of the corpus callosum, neuronal migration defects, eye abnormalities (chorioretinal lacunae, colobomas of the optic nerve and microphthalmia) and severe early-onset seizures and neurodevelopmental delay. It is now well recognized that other brain abnormalities, such as polymicrogyria, agyria, cysts and heterotopias are common features of Aicardi syndrome. We previously hypothesized that the gene causing Aicardi syndrome and possibly additional phenotypically similar disorders with X-linked inheritance, such as Goltz syndrome or Focal Dermal Hypoplasia, are in or near the region on chromosome Xp22 that is deleted in another condition named microphthalmia with linear skin defects syndrome (MLS), because all three have some clinical similarities. However, interim studies have shown that this is likely not the case because no mutations were found in ...

Enhanced sensitivity to Wnts is an emerging hallmark of a subset of cancers, defined in part by mutations regulating the abundance of their receptors. Whether these mutations identify a clinical opportunity is an important question. Inhibition of Wnt secretion by blocking an essential post-translational modification, palmitoleation, provides a useful therapeutic intervention. We developed a novel potent, orally available PORCN inhibitor, ETC-1922159 (henceforth called ETC-159) that blocks the secretion and activity of all Wnts. ETC-159 is remarkably effective in treating RSPO-translocation bearing colorectal cancer (CRC) patient-derived xenografts. This is the first example of effective targeted therapy for this subset of CRC. Consistent with a central role of Wnt signaling in regulation of gene expression, inhibition of PORCN in RSPO3-translocated cancers causes a marked remodeling of the transcriptome, with loss of cell cycle, stem cell and proliferation genes, and an increase in ...

Assembling the photographs of this trip has been something of a challenge because of some problems I had with my camera. After years of banging around boats, my old NIKON SLR developed a light leak into the film compartment, with the unfortunate result that many frames of the film were spoiled completely or contaminated with streaks. To salvage some shots, I have resorted to some unusually tight cropping or, in some cases where there was no alternative, to including some slightly spoiled images in the article. Even if all my rolls had turned out perfectly, I am more fortunate to have the photographs taken by my cruising mates, Larry Goltz and Jim Gibson. They gave me many fine photos which I have used to great advantage to illustrate the events of our trip. In particular, their excellent work with a camera has provided me with many views of my own boat on the journey which otherwise would have been impossible for me to have taken.. As a result of having three photographers, three cameras, three ...

ISSN 2173-9161. http://dx.doi.org/10.1016/j.maxilo.2012.09.008.. Introduction/objective: Gorlin Goltz syndrome (GGS) is an autosomal dominant inherited disorder that mainly predisposes to the proliferation of tumors, such as basal cell carcinomas and jaw keratocysts. GGS is caused by the Patched gene mutation on chromosome 9. Basal cell carcinomas in patients with GGS usually present as multiple tumors, with polymorphic clinical features, a non-gender predilection, sometimes occurring in the early stages of life, and even affecting areas not exposed to sunlight. The clinical behavior may vary, and sometimes can be very aggressive, especially in the face. In order to study the behavior of basal cell carcinomas in GGS patients, a study was performed on the patients who met criteria for the disease and were treated in our hospital ...

The PORCN gene provides instructions for making a protein that belongs to a group of related proteins called the porcupine (Porc) family. Although the precise function of the PORCN protein is unknown, proteins in the Porc family are involved in the process of transferring a molecule called palmitoleic acid to Wnt proteins. Wnt proteins participate in chemical signaling pathways in the body and play critical roles in development before birth. Members of the Porc family are located in the endoplasmic reticulum, which is a structure inside the cell that is involved in protein processing and transport. The transfer of palmitoleic acid to Wnt proteins facilitates the release of these proteins from the cell so they can regulate development of the skin, bones, and other structures. Researchers are working to determine the specific role of the PORCN protein within human cells. ...
With Winnipeg quarterback Buck Pierce injured yet again and backup Joey Elliot out for the year with a torn ACL making a tackle after an interception*, the Bombers are somewhat pooched in the quarterback department. This article goes over the problem and mentions the next three quarterbacks on the depth chart, Alex Brink, Justin Goltz and Brandon Summers. The Bombers picking up former Ticat third stringer Adam Trafalis wouldnt be a bad idea. Trafalis at least has been around the CFL for a while now. Sure the Ticats picked Jason Boltus as their third string quarterback over Trafalis this year, but with an excellent defence, you might be able to win some games with someone whos been in the league longer than training camp, with the added bonus of making Matt Dunnigan happy. With the way Bomber quarterbacks are dropping, they could soon be playing raw rookie fourth and fifth stringers. Hopefully against the Ticats ...

The cape porcupine (Hystrix africaeaustralis) is sometimes called the South African porcupine, and can be found in much of sub-Saharan Africa, however, it is not found in the southwestern desert regions. It is an Old World porcupine. The cape porcupine prefers to make its home in hills or rocky outcrops in dens or caves that it burrows.. The female members of this species are the largest rodents in southern Africa, weighing in at an average between forty to sixty-six pounds and they can grow to be two feet in length. The quills of this porcupine can be as long as its body and the hollow spines on the tail are used as a defensive mechanism where the porcupine will shake them to make a loud noise. When defending itself, it will either retreat into its burrow, extending its quills to make it difficult to extract, or it may become aggressive and charge the attacker to stab it with its spines.. Cape porcupines, unlike other rodents, can live to be between fifteen to twenty years and they mature very ...
ANSWER: To get rid of a porcupine, start with general exclusion tactics. Fencing around your trees and shrubs will prevent this rodent from reaching them. Gardens should also be fenced. Porcupines have been known to actively seek cat and dog food.
Natural predators of this species include fishers (a cat-sized mustelid)[20], wolverines[21], coyotes, wolves[22], American black bears[23], and cougars[24], as well as humans. The only known avian predators of this species are golden eagles and great horned owls.[25][26][27] In many cases, injury or even death may occur in the predator from embedded porcupine quills even if they are successful in dispatching the porcupine.[28][29] The North American porcupine is most at risk from the fisher (Martes pennanti). Fishers are related to the weasel, with males sometimes exceeding a mass of 5.5 kg (12 lb). Fishers have two advantages that make them capable hunters of the porcupine. First, they are agile tree climbers. If a fisher locates a porcupine, it cannot hide by fleeing into a tree because the fisher can pursue it and force it down to the ground. A porcupine can sometimes defend itself by facing the trunk on a branch and presenting its tail to the fisher. If the fisher manages to force a ...
Porcupines stick close to the trees. Beyond forests, you may find them alongside river undergrowth and maybe in the trees by a rocky ledge. They live in dens found in rock piles, caves, fallen logs and trees. Generally, they stay close to home leaving their dens for food - porcupines eat a variety of shrubs, bark, water plants and they love anything salty.. A peaceful animal, the porcupine will try to run away if it feels theres danger. Theyll make loud chattering noises as a warning for predators to leave. If they cant get away, their muscles tighten forcing their quills to come out. It will tuck its head in, lean forward and thump its back feet while swinging its tail as a warning. Sometimes loose quills fly out of the tail or if a predator tries to get too close, the quills will stick them. The quills are an amazing defense mechanism - when they get lodged in the skin, body heat makes the barbs swell, making it even harder and more painful to pull them out.. Animals like bears, bobcats, ...

This gene encodes a guanosine triphosphate (GTP)-metabolizing protein that contains a phorbol-ester/diacylglycerol (DAG)-type zinc finger, a Rho-GAP domain, and an SH2 domain. The encoded protein translocates from the cytosol to the Golgi apparatus membrane upon binding by diacylglycerol (DAG). Activity of this protein is important in cell proliferation and migration, and expression changes in this gene have been detected in cancers. A mutation in this gene has also been associated with schizophrenia in men. Alternative transcript splicing and the use of alternative promoters results in multiple transcript variants. Figuring out the sex of a porcupine can be a difficult task. They arent always easy to handle, and when they are young they show no sexual dimorphism. But veterinarians, geneticists, and keepers at the Smithsonians National Zoo have figured out a novel and easy way to determine the sex of prehensile-tailed porcupines, and all it takes is a single quill.
